Changes in the haemocyte population of the mosquito, Culex quinquefasciatus, following infection with the filarial parasite, Wuchereria bancrofti.

The mosquito Culex quinquefasciatus Say (Diptera: Culicidae) is the vector of the filarial parasite Wuchereria bancrofti (Cobbold) (Spirurida: Onchocercidae), which causes human bancroftian filariasis. Information on the mosquito humoral response against the filarial parasite during the process of its infection and development is important, as it decides the vector competence of the mosquito. Visible changes in the haemocyte population of mosquito, if any, will be an indicator of the possible humoral factors. The present study was aimed at investigating changes in the populations of various ty

Pharmacological validation of an inward-rectifier potassium (Kir) channel as an insecticide target in the yellow fever mosquito Aedes aegypti.

Mosquitoes are important disease vectors that transmit a wide variety of pathogens to humans, including those that cause malaria and dengue fever. Insecticides have traditionally been deployed to control populations of disease-causing mosquitoes, but the emergence of insecticide resistance has severely limited the number of active compounds that are used against mosquitoes. Thus, to improve the control of resistant mosquitoes there is a need to identify new insecticide targets and active compounds for insecticide development. Recently we demonstrated that inward rectifier potassium (Kir) chann

Nationwide larval mosquito sampling in Cambodian households: Vector species in anthropogenic breeding sites.

Vector control remains a key strategy in reducing mosquito-borne disease transmission. Understanding mosquito species distribution, diversity, and breeding habitat ecology is crucial for effective surveillance and to define targeted vector control interventions. We conducted a study to understand the diversity and habitat preferences of mosquito larvae across Cambodia during the rainy season from July to September 2024. Mosquito larvae were collected from a variety of breeding habitats located around households across all 25 provinces. The national sampling was conducted once during the rainy

Design, synthesis, characterization and insecticidal screening of novel anthranilic diamides comprising acyl thiourea substructure.

BACKGROUND: Mosquito-borne pathogens constitute a major health problem worldwide. The extermination of the mosquito remains a significant issue in public health. Chemical insecticides have been used to control mosquitoes for decades. However, resistance has become a limiting factor for their control. The anthranilic diamide insecticides possess excellent insecticidal activities against Lepidoptera and its resistant strains by draining internal calcium stores on activating insect ryanodine receptors. However, the reports on the effect on mosquitoes are scarce and hence a series of novel anthran

Contrasting patterns of tolerance between chemical and biological insecticides in mosquitoes exposed to UV-A.

Mosquitoes are vectors of major human diseases, such as malaria, dengue or yellow fever. Because no efficient treatments or vaccines are available for most of these diseases, control measures rely mainly on reducing mosquito populations by the use of insecticides. Numerous biotic and abiotic factors are known to modulate the efficacy of insecticides used in mosquito control. Mosquito breeding sites vary from opened to high vegetation covered areas leading to a large ultraviolet gradient exposure. This ecological feature may affect the general physiology of the insect, including the resistance …

Mosquito-Borne Viral Pathogens Detected in Zambia: A Systematic Review.

Emerging and re-emerging mosquito-borne viral diseases are a threat to global health. This systematic review aimed to investigate the available evidence of mosquito-borne viral pathogens reported in Zambia. A search of literature was conducted in PubMed and Google Scholar for articles published from 1 January 1930 to 30 June 2020 using a combination of keywords. Eight mosquito-borne viruses belonging to three families, Togaviridae, Flaviviridae and Phenuiviridae were reported. Three viruses (Chikungunya virus, Mayaro virus, Mwinilunga virus) were reported among the togaviruses whilst four (den

Visualization and live imaging analysis of a mosquito saliva protein in host animal skin using a transgenic mosquito with a secreted luciferase reporter system.

Mosquitoes inject saliva into a vertebrate host during blood feeding. The analysis of mosquito saliva in host skin is important for the elucidation of the inflammatory responses to mosquito bites, the development of antithrombotic drugs, and the transmission-blocking of vector-borne diseases. We produced transgenic Anopheles stephensi mosquitoes expressing the secretory luciferase protein (MetLuc) fused to a saliva protein (AAPP) in the salivary glands. The transgene product (AAPP-MetLuc) of transgenic mosquitoes exhibited both luciferase activity as a MetLuc and binding activity to collagen a

A peridomestic Aedes malayensis population in Singapore can transmit yellow fever virus.

The case-fatality rate of yellow fever virus (YFV) is one of the highest among arthropod-borne viruses (arboviruses). Although historically, the Asia-Pacific region has remained free of YFV, the risk of introduction has never been higher due to the increasing influx of people from endemic regions and the recent outbreaks in Africa and South America. Singapore is a global hub for trade and tourism and therefore at high risk for YFV introduction. Effective control of the main domestic mosquito vector Aedes aegypti in Singapore has failed to prevent re-emergence of dengue, chikungunya and Zika vi

In silico DNA barcoding surpasses whole genome sequencing for species identification from vector surveillance pools.

Mosquito-borne diseases are responsible for over 600,000 deaths annually, mainly in sub-Saharan countries. Mosquito surveillance is a crucial element of vector control programmes to assure they remain effective. This study focused on optimizing the use of the MinION sequencer for interrogating mosquito pools from surveillance programmes, with simplified bioinformatic workflows for use in African laboratories nearer to the field. Mosquito pools were created using different human disease vector species, mimicking possible field trap contents. Some pools were spiked with DNA from the malaria para

Sterilising effects of pyriproxyfen on Anopheles arabiensis and its potential use in malaria control.

BACKGROUND: Insecticide resistance poses a major threat to current vector control campaigns. Insecticides with novel modes of action are therefore in high demand. Pyriproxyfen (PPF), a conventional mosquito pupacide, has a unique mode of action that also sterilises adult mosquitoes (unable to produce viable offspring) upon direct contact. However, the timing of PPF exposure in relation to when mosquitoes take a blood meal has an important impact on that sterilisation. This study investigated the relationship between different blood feeding and PPF exposure timings to determine the potential of

Do vegetated rooftops attract more mosquitoes? Monitoring disease vector abundance on urban green roofs.

Green roof, an increasingly common constituent of urban green infrastructure, can provide multiple ecosystem services and mitigate climate-change and urban-heat-island challenges. Its adoption has been beset by a longstanding preconception of attracting urban pests like mosquitoes. As more cities may become vulnerable to emerging and re-emerging mosquito-borne infectious diseases, the knowledge gap needs to be filled. This study gauges the habitat preference of vector mosquitoes for extensive green roofs vis-à-vis positive and negative control sites in an urban setting. Seven sites in a univer

Effect of controlled burning on survival of floodwater Aedes eggs in Kenya.

The effect of controlled burning on the survival of Aedes mosquito eggs was evaluated in 2 distinct dambo habitats. In a dambo dominated by grasses, egg survival was 3.3% after burning compared with 43.8% in a similar dambo that was not burned. In a dambo dominated by sedges, egg survival was 0.7% after burning compared with 28.5% in a similar dambo that was not burned. Mortality of mosquito eggs appeared to be caused by high temperatures associated with the fire and not elapsed time since egg survival did not decrease with time after burning. The potential for burning to control the mosquito

Mosquito Magnet® traps as a potential means of monitoring blackflies of medical and veterinary importance.

Mosquito Magnet® traps, deployed in widespread parts of England as part of nationwide mosquito surveillance projects, also caught blackflies. As many as 1242 blackflies were caught in a trapping session lasting 4 days. Principal among the species caught were Simulium equinum, Simulium lineatum and Simulium ornatum s.l. As S. ornatum s.l. is a vector that transmits Onchocerca linealis to cattle and S. equinum is responsible for dermatitis ('sweet itch') in cattle and horses, it is suggested that Mosquito Magnet® traps could be used to monitor and partially control these pests, as well as nuisan

Carlos Finlay and yellow fever: triumph over adversity.

The eradication of yellow fever in Havana, Cuba, was achieved by a fruitful collaboration between American and Cuban physicians. Carlos Finlay, a Cuban physician who proposed the mosquito-vector theory in 1881, shared his ideas, his publications, and a sample of mosquito eggs with the U.S. Army Yellow Fever Commission. The commission, headed by MAJ Walter Reed, used human volunteers to confirm Finlay's theory. MAJ William Gorgas adopted mosquito-control measures in his sanitation program and, within 6 months, yellow fever was controlled in Havana for the first time. Finlay held fast to his ide

Heritable strategies for controlling insect vectors of disease.

Mosquito-borne diseases are causing a substantial burden of mortality, morbidity and economic loss in many parts of the world, despite current control efforts, and new complementary approaches to controlling these diseases are needed. One promising class of new interventions under development involves the heritable modification of the mosquito by insertion of novel genes into the nucleus or of Wolbachia endosymbionts into the cytoplasm. Once released into a target population, these modifications can act to reduce one or more components of the mosquito population's vectorial capacity (e.g. the
