Assessment of human cancer risk from animal carcinogen studies is severely limited by inadequate experimental data at environmentally relevant exposures and by procedures requiring modeled extrapolations many orders of magnitude below observable data. We used rainbow trout, an animal model well-suited to ultralow-dose carcinogenesis research, to explore dose-response down to a targeted 10 excess liver tumors per 10000 animals (ED(001)). Peliosis hepatis, characterized by the presence of blood-filled spaces within hepatic parenchyma, developed in C57Bl mice implanted subcutaneously with melanoma cells 23 days previously. Pharmacokinetic studies with dichloroacetate (DCA) provide insights into the likelihood that trichloroethylene-induced liver cancers arise from formation of DCA as a metabolite and the mode of action by which DCA induces liver cancer. The present study reports the effect of dietary selenium deficiency and supplementation on the growth of the transplantable MT-W9B mammary tumor in female Wistar-Furth rats. Supplementation of the diet with 2 ppm of selenium inhibited tumor growth and reduced the final tumor weight by approximately 50% compared to the control rats receiving 0.1 ppm of selenium. The inhibitory response was selective, without inducing any weight loss in the animals. On the other hand, selenium deficiency (less than 0.02 ppm) had no influence on the growth of this tumor.
