The effects of two diazepines (diazepam and Ro 11-7800) and 3 barbiturates (thiamylal, pentobarbitol and phenobarbital) on GABA-mediated recurrent inhibition were assessed on single hippocampal pyramidal cells and on population spikes using extracellular recording techniques. Recurrent inhibition was evoked in spontaneously active CA1 pyramidal cells by stimulation of the fimbria or the alveus with single shocks. Microiontophoretic application of Ro 11-7800 or systemic application of diazepines or barbiturates resulted in an increase of the duration of the inhibition and in a concomitant depre

The inhibitory response to nerve stimulation was studied in three isolated preparations of the rat stomach. Bethanidine and guanethidine in doses sufficient to block adrenergic neurotransmission reduced but never abolished the inhibitory response to field stimulation of the rat stomach strip. In similar concentration of these drugs, the response to vagus nerve stimulation was diminished; higher doses of blocking agent abolished this response. Relaxation to both field and vagus nerve stimulation could be evoked in stoamchs removed from rats which had received sufficient reserpine to deplte tiss

1. We have investigated the effects of stimulating carotid sinus baroreceptors upon the activity of single cortical pyramidal tract cells (PT-cells) in anaesthetized cats.2. Extracellular potentials were recorded from PT-cells, which were driven orthodromically (1/sec) by stimulating thalamic nuclei (N. ventralis lateralis, N. ventralis posterolateralis) or afferent nerves in the contra-lateral forepaw. Baroreceptors were stimulated by inflating small balloons placed in the bifurcations of one or both common carotid arteries.3. Distension of the carotid sinus caused a prolonged depression of t

In local application of substances with a mediator action (adrenaline, acetylcholine, histamine) and blockers of specific receptors (dihydroergotoxin, inderal, atropine and of subcutaneous dimedrol) together with dicaine it was possible to reveal a distinct presence in the cornea of the eye of the mediator receptors capable of influencing one another. Pharmacological analysis permitted a supposition to be made that there were two receptor systems in the sensory nerve endings of the cornea; these systems were in reciprocal inhibitory relations; due to this their regular influence on the process

A complete system for neural regulation of insulin and glucagon secretion from the ventral hypothalamus to the autonomic cholinergic and adrenergic nerves of the endocrine pancreas is described. Both physiologic and pathophysiologic states of altered metabolism can be partly explained by activation of this neural system. Direct hypothalamic humoral control of the endocrine pancreas has been evaluated by studies of the hypothalamic hormone somatostatin, or somatotrophin release inhibiting factor (SRIF). Somatostatin has been shown to inhibit pancreatic glucagon and insulin secretion when infuse

Seesaw nystagmus is characterized by the rhythmic combination of vertical and torsional dysconjugate oscillations where one eye moves up and inward while the other moves down and outward. Common association of seesaw nystagmus with accessory optic track lesions lead to traditional hypothesis that it is due to the mismatch in the vision and vestibular systems. Here we propose a novel mechanism for seesaw nystagmus. We hypothesize that reverberations due to abnormal increases in the excitability of the reciprocally innervating circuit of excitatory burst neuron in the midbrain interstitial nucle

The cortico-striatal-thalamo-cortical (CSTC) pathway is a brain circuit that controls movement execution, habit formation and reward. Hyperactivity in the CSTC pathway is involved in obsessive compulsive disorder (OCD), a neuropsychiatric disorder characterized by the execution of repetitive involuntary movements. The striatum shapes the activity of the CSTC pathway through the coordinated activation of two classes of medium spiny neurons (MSNs) expressing D1 or D2 dopamine receptors. The exact mechanisms by which balanced excitation/inhibition (E/I) of these cells controls the network dynamic

The rostral ventrolateral medulla (RVLM) contains neurons critical for cardiovascular, respiratory, metabolic, and motor control. The activity of these neurons is controlled by inputs from multiple identified brain regions; however, the neurochemistry of these inputs is largely unknown. Gamma-aminobutyric acid (GABA) and enkephalin tonically inhibit neurons within the RVLM. The aim of this study was to identify all brain regions that provide GABAergic or enkephalinergic input to the rat RVLM. Neurons immunoreactive for cholera toxin B (CTB-ir), retrogradely transported from the RVLM, were asse

Repetitive training of isolated movements induces reorganization of motor cortical representations. To elucidate the mechanisms of practice-dependent cortical plasticity within the lesioned central motor system at the time of the application of a therapeutic intervention, we examined the effect of repetitive movements on intracortical facilitation (ICF) and inhibition of agonist (extensor carpi radialis [ECR]) and antagonist (flexor carpi ulnaris) muscles of the hand shortly after the movements, by the paired-pulse technique in patients with cortical (n = 9) and subcortical strokes (n = 11). Prepulse inhibition (PPI) of the acoustic startle response and open-field locomotor activity were measured after bilateral infusion of N-methyl-D-aspartate into the ventral (0.10, 0.25, 0.50 microg/side) and dorsal (0.10, 0.25, 0.50, 0.70 microg/side) hippocampus of Wistar rats. Dose-dependent hyperactivity and disruption of PPI--behavioral effects related to psychotic symptoms--were observed after ventral infusions but were virtually absent after dorsal infusions. This functional dorsal-ventral difference might be related to the different connections of the dorsal and ventral hippocampus with

Descending inhibition plays an important role in modulating spinal nociceptive neurotransmission. Barbiturates have been suggested to be poor analgesics or anti-analgesic because they block descending inhibition from supraspinal centers to the spinal cord. Opiate analgesics, on the other hand, are postulated to increase descending inhibition. We tested this hypothesis in an isolated brain stem-spinal cord preparation from neonatal rats, using as the test response a nociceptive-related slow ventral root potential (sVRP) recorded in the lumbar region. Brain stem and spinal cord were separately p

Carr and Coons (1982a, 1982b) found that lateral hypothalamic (LH) stimulation ameliorates the aversiveness of stimulation of pain-implicated nucleus gigantocellularis (NGC), but this finding disagrees with other findings. To resolve this disagreement, we tested whether amelioration is differentially associated with the ability of LH stimulation to support self-stimulation (SS), to support responding to escape LH stimulation (LH escape), or to elicit stimulation-bound feeding (SBF). LH stimulation not yielding SBF always increased responding to escape from NGC stimulation (NGC escape) and was

Rostral septal lesions accelerate somatic growth in adult hamsters. This study tested the hypothesis that this effect results from damage to fibers of passage by observing the effects of transections of septohippocampal and septohypothalamic connections on growth. We attempted to identify these fibers further by (a) measuring spectrofluorometrically changes in the monoamine concentrations in hippocampus, cerebral cortex, corpus striatum, and diencephalon, (b) staining the degenerating axons after septal lesions and the two cuts, and (c) examining the correspondence between such damage and the

Cortical circuits exhibit variable yet bounded activity patterns, suggesting operation near-but not within-fully chaotic regimes. Here we develop a minimal three-variable rate model for primary visual cortex (V1) that reveals how biologically motivated feedback mechanisms can function as intrinsic chaos controllers. We adopt the simplest known chaotic Lotka-Volterra system as a phenomenological scaffold and introduce three biologically motivated modifications: excitatory-to-inhibitory (E→I) feedback coupling, homeostatic regulation of modulatory drive, and orientation-tuned sensory input. Thes

In amyotrophic lateral sclerosis (ALS) motor cortex, hyperexcitability is detected in both familial and sporadic cases, suggesting its centrality in the ALS phenotype; the underlying mechanisms, however, remain largely obscure. Here we utilize male and female UCHL1-eGFP (UeGFP) mice, in which the corticospinal neurons of the motor cortex are labeled with green fluorescent protein, to investigate the intrinsic excitability and synaptic inhibitory inputs on distinct neuron populations in WT-UeGFP and presymptomatic AlsinKO-UeGFP mice, which lack Alsin function and are a well-characterized mouse

Neuronal hyperexcitability is a common pathophysiological feature of many neurological diseases. Neuron-glia interactions underlie this process but the detailed mechanisms remain unclear. Here, we reveal a critical role of microglia-mediated selective elimination of inhibitory synapses in driving neuronal hyperexcitability. In epileptic mice of both sexes, hyperactive inhibitory neurons directly activate surveilling microglia via GABAergic signaling. In response, these activated microglia preferentially phagocytose inhibitory synapses, disrupting the balance between excitatory and inhibitory s

INTRODUCTION: Cerebellar brain inhibition (CBI) is a way to quantify the cerebellar influence on the motor cortex in humans. Studies suggest that the orientation of the transcranial magnetic stimulation (TMS) coil influences which motor networks are activated. This study investigated the influence of cerebellar transcranial direct current stimulation (ctDCS) on CBI as a function of coil orientation (anterior-posterior [AP] vs. posterior-anterior [PA]). An interstimulus interval (ISI) of 7 ms (CBI-AP-7) was used for AP orientation and 5 ms (CBI-PA-5) for PA orientation.
METHODS: Young and heal
