Targeting fusion proteins in solid tumors: from oncogenic mechanisms to clinical interventions.

Fusion proteins are implicated in a wide range of solid tumors, including lung cancer, breast cancer, and prostate cancer, and play a critical role in tumor initiation and progression through diverse molecular mechanisms. In this review, we provide an overview of the mechanisms underlying the generation of fusion proteins in solid tumors and their oncogenic roles, classifying these proteins based on different carcinogenic mechanisms. We then highlight the current research progress on targeted intervention strategies for fusion proteins in solid tumors, including small molecule inhibitors, anti

Proteomic profiling identifies an oncogene ITGA2 and its downstream targets in gastric cancer.

PURPOSE: Gastric cancer is a leading cause of cancer-related mortality worldwide, with metastasis being the primary cause of death. Exosomes secreted by tumor cells are key mediators of intercellular communication and can prepare distant sites for metastasis by altering the local microenvironment. We hypothesized that exosomes released by gastric cancer cells deliver cargo that regulates specific proteins in recipient gastric cancer cells, thereby enhancing tumor progression and metastatic potential. METHODS: Human gastric cancer cell lines were treated with exosomes isolated from the conditi

Oncogenic and tumor-suppressive forces converge on a progenitor niche at the benign-to-malignant transition.

The benign-to-malignant transition is a defining step in cancer progression. To investigate when and how malignancy initiation occurs and tissue reorganization proceeds, we combine single-cell and spatial transcriptomic profiling in mouse models of pancreatic ductal adenocarcinoma (PDAC) that capture spontaneous p53 loss. Among Kras-mutant cells, we find that oncogenic and tumor-suppressive programs, including those controlled by p53, CDKN2A, and SMAD4, are co-activated in a discrete progenitor-like population, engaging senescence-like responses. Using a framework we developed for spatial anal

FGFR2 is an oncogenic driver critical for successful human gammaherpesvirus latent infection and oncogenesis.

Epstein-Barr virus (EBV) and Kaposi's sarcoma-associated herpesvirus (KSHV) drive multiple aggressive lymphomas, yet effective targeted therapies for these virus-associated malignancies remain limited. Using an unbiased kinome-wide screen combined with analysis of virus-positive patient tumors, we identified fibroblast growth factor receptor 2 (FGFR2) as a selectively activated host kinase in EBV- and KSHV-associated lymphomas. Importantly, FGFR2 is required for efficient establishment of EBV latent infection, and its knockdown markedly impairs the formation of viral latency programs. Viral la

Host membrane cholesterol constrains constitutive signaling of the oncogenic KSHV GPCR ORF74.

UNLABELLED: Cholesterol is a major structural component of the plasma membrane and a key allosteric regulator of G protein-coupled receptors … ANCE: Constitutively active viral G protein-coupled receptors (GPCRs) promote viral persistence and oncogenesis, yet how excessive signaling is restrained has remained unknown. We identify a cholesterol-binding allosteric pocket that stabilizes the inactive conformation of the Kaposi's sarcoma-associated herpesvirus GPCR ORF74, thereby suppressing constitut …

POU2F2 as a dual oncogenic and immunomodulatory driver in kidney renal clear cell carcinoma.

Kidney renal clear cell carcinoma (KIRC) features an immunosuppressive tumor microenvironment (TME). We investigated the role of transcription factor POU2F2 in KIRC oncogenesis and macrophage polarization. Single-cell and spatial transcriptomics showed that POU2F2 is upregulated in KIRC and associated with metastasis and poor prognosis. POU2F2 knockdown inhibited KIRC cell proliferation, migration, and invasion in vitro. POU2F2 expression correlated with macrophage infiltration and JAK-STAT signaling. Co-culture assays showed that POU2F2 silencing reduced macrophage recruitment and shifted M2 …

Immune Dysregulation and Viral Persistence in HPV-HIV Coinfection: Oncogenic and Clinical Implications.

INTRODUCTION: Human papillomavirus (HPV) associated cancers constitute a major global health concern, particularly among people living with human immunodeficiency virus (HIV). Accumulating evidence suggests that HPV-HIV co-infection substantially increases the risk of genital, cervical, and oral cancers; however, the molecular and immunological mechanisms underlying this association remain incompletely elucidated. OBJECTIVE: To critically review the current evidence regarding HPV-HIV interactions, focusing on immunological synthesis, translation, clinical implications, and development of a co

The commensal microbiome in respiratory tract tumors: From oncogenic mechanisms to clinical translation.

BACKGROUND: The human commensal microbiome, commonly recognized as a "second genome", exerts a crucial regulatory role in host metabolism, immune homeostasis maintenance, and the progression of various diseases. Respiratory tract tumors remain a leading cause of global cancer-related deaths due to their high invasiveness and late-stage diagnosis. With the rapid development of high-throughput sequencing technology, the complex associations between microbial communities and respiratory tract tumors have been gradually uncovered. In lung cancer, distinct microbiota-related signatures, including t

Influence of oncogenic drivers and signaling pathways on immune checkpoint inhibitor efficacy in NSCLC.

Non-small cell lung cancer (NSCLC) is characterized by intricate genetic modifications and mechanisms that allow it to evade immune detection, which complicates the pursuit of lasting therapeutic responses. The effectiveness of treatments is greatly affected by the interaction between tumor genetic profiles and the surrounding immune microenvironment, highlighting the need for carefully designed therapeutic combinations. This review provides a comprehensive analysis of oncogenic drivers and signaling pathways (such as the KRAS, BRAF, and CD73/adenosine pathway) that interact with immune checkp

Spatially resolved transcriptomics identifies epithelial-specific oncogenic signaling networks in ameloblastoma.

BACKGROUND/PURPOSE: Ameloblastoma is a benign odontogenic tumor characterized by locally aggressive growth and a high recurrence rate. Although recurrent mutations such as BRAF V600E have been identified, the epithelial-specific molecular mechanisms underlying tumor initiation and invasion remain incompletely defined. Previous transcriptomic studies have been limited by stromal contamination and lack of spatial resolution. MATERIALS AND METHODS: An integrative transcriptomic strategy was applied, combining bulk RNA sequencing of fresh ameloblastoma and dental follicular tissues, RNA sequencin

LIMCH1 in Cancer: A Context-Dependent Modulator Beyond the Tumor Suppressor-Oncogene Dichotomy.

LIM and calponin homology domain-containing protein 1 (LIMCH1) has recently gained attention as a functionally perplexing factor in human cancers, with studies attributing both tumor-suppressive and tumor-promoting properties that defy simple categorization. Originally described as an actin-associated cytoskeletal protein that promotes non-muscle myosin-IIA (NM-IIA) activity and thereby curtails cell migration, LIMCH1 has more recently been linked to diverse oncogenic pathways across a range of tumor types. In lung adenocarcinoma and clear-cell renal cell carcinoma, diminished LIMCH1 expressio

The oncogenic role of hypomethylated ZNF793 in gastric carcinoma: a focus on cell survival and stemness.

BACKGROUND: Although genome-wide promoter CpG island hypermethylation of Epstein-Barr virus-associated gastric carcinoma (EBV GC) is well known, ZNF793 is rarely methylated in EBV GC but is frequently methylated in other molecular subtypes of GC, including microsatellite instability-high GC. Based on the hypothesis that ZNF793 may be important for cell survival and stemness in EBV GC, the oncogenic role of ZNF793 was investigated. METHODS: ZNF793 expression was knocked out and then restored in EBV and non-EBV GC cell lines, and its effects on cell proliferation, cell migration, cell invasion,

Proteomic analyses identify targets, pathways, and cellular consequences of oncogenic KRAS signaling.

Mutations that activate the small GTPase KRAS are a frequent genetic alteration in cancer, and drug discovery efforts have led to inhibitors that block KRAS activity. We sought to better understand oncogenic KRAS signaling and the cytostatic effects of drugs that target this system. We performed proteomic analyses to investigate changes in protein abundance and posttranslational modifications in inhibitor-treated human KRAS-mutant pancreatic (KRAS G12C and G12D) and lung cancer (KRAS G12C) cells. The inhibitors used target these mutant forms of KRAS, the downstream effectors MEK and ERK, and t

A timeless chronicle: Effects of cigarette smoke on GPR15 receptor and its oncogenic potential.

G-protein-coupled receptors (GPCRs) represent the largest family of membrane receptors encoded in the human genome, with diverse physiological and pathological functions. GPR15, a recently characterized member of this family, has emerged as a receptor of interest due to its significant upregulation and hypomethylation in response to cigarette smoking, a modifiable environmental risk factor implicated in numerous diseases. Initially, it was identified as a coreceptor for viruses, human immunodeficiency virus (HIV) and simian immunodeficiency virus (SIV). It was later deorphanized with the disco

The nuclear receptor NR6A1 plays an oncogenic role through reprogramming glycolysis in tumors.

NR6A1 is a member of the nuclear receptor superfamily of ligand-activated transcription factors and can bind to conserved DNA sequences, acting as a transcriptional repressor. NR6A1 has been reported to promote prostate cancer, gastric cancer, and testicular germ cell tumor progression. In the present study, we confirmed the oncogenic role of NR6A1 in several types of cancer cells, including HeLa, TFK1, and A549 cells, and revealed that NR6A1 knockdown led to a decrease in lung adenocarcinoma cell proliferation, a reduction in glucose consumption, a reduction in lactic acid production, and dec
