Bone remodeling: a central mechanism in prostate cancer bone metastasis.

Prostate cancer (PCa) commonly metastasizes to bone, leading predominantly to osteoblastic lesions driven by intricate cellular interactions … h factor 23 (FGF-23), and platelet-derived growth factor (PDGF) further enhance tumor colonization. Osteocytes (OCYs), the most abundant and long-lived bone cells, directly interact with PCa cells and, in response to altered mechanotransduction, release pro-metastatic mediators including CCL5 and matrix metalloproteinases (MMPs). Moreover, PCa cells actively …

Hydrogen activates ACOD1-itaconate pathway to ameliorate steroid-associated osteonecrosis.

Steroid-associated osteonecrosis (SAON) remains a challenging clinical condition as there are few effective preventive measures. This study investigates the effects of hydrogen (H2) administrated via saturated hydrogen-rich water (HRW) in mice received high dose of glucocorticoids (for inducing SAON model). Here we find that HRW treatment significantly reduces osteocyte apoptosis, improves deteriorated trabecular architecture, increases osteoblast numbers and the bone formation, while decreases osteoclast numbers and the bone resorption. Additionally, HRW-treated mice exhibit improved serum li

There is a relationship between obesity and … singly, the relationship between bone and obesity is not unidirectional; there is now evidence that osteocytes are able to regulate body weight by acting as weighing machines.

Expression regulation and functional analysis of RGS2 and RGS4 in adipogenic and osteogenic differentiation of human mesenchymal stem cells.

BACKGROUND: Understanding the molecular basis underlying the formation of bone-forming osteocytes and lipid-storing adipocytes will help provide insights into the cause of disorders originating in stem/progenitor cells and develop therapeutic treatments for bone- or adipose-related diseases. In this study, the role of RGS2 and RGS4, two members of the regulators of G protein signaling (RGS) family, was investigated during adipogenenic and osteogenenic differentiation of human mesenchymal stem cells (hMSCs). RESULTS: Expression of RGS2 and RGS4 were found to be inversely regulated during adipo…

Influence of magnesium depletion on matrix-induced endochondral bone formation.

The effect of magnesium deficiency on bone cell differentiation and bone formation was investigated using in vivo matrix-induced endochondral ossification as a test system. Demineralized bone matrix was implanted subcutaneously in young (35-day-old) male Long-Evans rats that had been fed a semisynthetic Mg-deficient diet (50 ppm Mg) for 7 days. Plasma Mg levels were reduced to 25-30% of control values at that time. Control rats were paired the same diet, supplemented to contain 1000 ppm Mg. The implants were harvested 7, 9, 11, 15, and 20 days after implantation and analyzed for Mg and Ca cont

[Effect of space flight on rat skeletal bones (an optical light and electron microscopic study)].

By light and electron microscopy long tubular bones of white Wistar rats were examined after a 22-day space flight and ground-based synhcroneous experiment. Approximately half of the flight rats showed loosened spongiose of metaphyses which was normally combined with a reduced area of the primary spongiose near the cartilaginous growth plate. This was suggestive of inhibited growth of bones in-flight. The light and electron microscopic examination of bones of flight rats revealed wide osteocyte cavities (lacunae) that could be associated with perilacunar osteolysis. The study of synchroneous a

Distribution of Immunomodulation, Protection and Regeneration Factors in Cleft-Affected Bone and Cartilage.

BACKGROUND: Craniofacial clefts can form a significant defect within bone and cartilage, which can negatively affect tissue homeostasis and … e reacting to stressors caused by the local tissue defect. A significant increase in Col-I-positive osteocytes in cleft patients might indicate increased bone remodeling and osteocyte activity due to the presence of a cleft. Correlations between factors indicate notable differences in molecular interactions within each group.

Overexpression of Fam20C in osteoblast in vivo leads to increased cortical bone formation and osteoclastic bone resorption.

Fam20C, which phosphorylates many secretory proteins with S-x-E/pS motifs, is highly expressed in bone and tooth tissues, implying that Fam20C-mediated phosphorylation is critical for regulation of these mineralized tissues. Previous studies of Fam20C-deficient mice revealed that Fam20C plays important roles in bone formation and mineralization. However, Fam20C-deficient mice develop hypophosphatemia, a systemic factor that masks the local effect of Fam20C in the bone tissue; consequently, the local role of Fam20C remains unknown. To elucidate the local function of Fam20C in bone tissue, we st

RISK-FACTORS, PATHOGENESIS, AND PHARMACEUTICAL APPROACHES FOR TREATMENT OF STEROID-INDUCED BONE INFARCTION OF FEMORAL HEAD.

During first year of steroid usage, osteocyte necrosis and blood vessel blockage may occur, which subsequently may produce steroid-induced bone infarction (SIBI) resulting in painful movement of patient. For treatment of SIBI, pharmaceutical strategy is the basic approach. It involves the use of various pharmacologically active compounds including bisphosphonates, hyperbaric oxygen (HBO), coenzyme Q10, erythropoietin, antihyperlipidemics, anticoagulants, antioxidants, and tissue repair protein. Out of these, there is no pharmaceutical agent that may completely treat this disease because many f

Lipogranuloma with osseous metaplasia in the breast that developed after "Bu-Hwang" oriental medicine treatment.

A lipogranuloma is an inflammatory reactive process associated with exogenous or endogenous lipids, and it's occurrence in the breast has rarely been reported. Osseous metaplasia, which is used to describe bone formation in abnormal locations, can develop from several conditions such as trauma or a tumor. However, few studies have reported benign breast lesions that have been seen as osseous metaplasia. We present a case of a benign calcified breast lesion that developed after a traumatic treatment process called "Bu-Hwang", and it was confirmed as a lipogranuloma with osseous metaplasia. To t

[Glucocorticoid and bone metabolism and disease].

Glucocorticoid (GC) is widely used to treat a variety of inflammatory and allergic diseases and about one million people in Japan are known to take oral GC. GC causes a number of significant side effects and among them osteoporosis and degenerative bone fracture are major complications of GC therapy. GC induces apoptosis of osteoblasts and osteocytes and prolongs lifespan of osteoclast, resulting in severe osteoporosis. However, few physicians are aware of the increased risk of fracture caused by GC. Recent multiple evidence strongly indicates that bisphosphonate is effective for treatment and…

Histologic evaluation of a threaded hydroxyapatite-coated root-form implant located at a dehisced maxillary site and retrieved from a human subject: a clinical report.

This clinical report describes the microscopic analysis of a threaded hydroxyapatite-coated root-form implant retrieved from an 89-year-old subject after 10 months of service. The implant was never loaded and was removed because it was nonrestorable. Clinically, the buccal area of the implant was covered by soft tissue, whereas the palatal side was covered by bone. Light microscopic evaluation revealed tight contact between hydroxyapatite and bone with no sign of coating dissolution. Osteocytes were present, and Haversian canals were in close proximity to the implant surface. The buccal side o…
