Oxidative damage in brains of mice treated with apomorphine and its oxidized derivative.

Increasing evidence suggests that some of the neurobiological and neurotoxic actions of apomorphine and other dopamine receptor agonists might be mediated by their oxidation derivatives. The aim of the present study was to evaluate the effects of apomorphine and its oxidation derivative, 8-oxo-apomorphine-semiquinone (8-OASQ), on oxidative stress parameters and antioxidant enzyme activity. Adult male CF-1 mice were treated with a systemic injection of apomorphine (0.4, 4.0 or 40.0 mg/kg) or 8-OASQ (0.4, 4.0 or 40.0 mg/kg). Animals were sacrificed by decapitation 24 h after treatment, and the f

Unique oxidative mechanisms for the reactive nitrogen oxide species, nitroxyl anion.

The nitroxyl anion (NO-) is a highly reactive molecule that may be involved in pathophysiological actions associated with increased formation of reactive nitrogen oxide species. Angeli's salt (Na2N2O3; AS) is a NO- donor that has been shown to exert marked cytotoxicity. However, its decomposition intermediates have not been well characterized. In this study, the chemical reactivity of AS was examined and compared with that of peroxynitrite (ONOO-) and NO/N2O3. Under aerobic conditions, AS and ONOO- exhibited similar and considerably higher affinities for dihydrorhodamine (DHR) than NO/N2O3. Qu

Nitric oxide scavengers as a therapeutic approach to nitric oxide mediated disease.

The essential role of nitric oxide (NO) in normal physiology and its involvement in the pathophysiology of a variety of diseases render the compound an attractive therapeutic target. NO donor drugs are used in the treatment of hypotension and angina where abnormalities in the L-arginine-nitric oxide pathway have been implicated. Overproduction of NO has been associated with a number of disease states including septic shock, inflammatory diseases, diabetes, ischaemia-reperfusion injury, adult respiratory distress syndrome, neurodegenerative diseases and allograft rejection. NO is produced by a

Oxidative chemistry of nitric oxide: the roles of superoxide, peroxynitrite, and carbon dioxide.

The roles of superoxide (O2.-), peroxynitrite, and carbon dioxide in the oxidative chemistry of nitric oxide (.NO) are reviewed. The formation of peroxynitrite from .NO and O2.- is controlled by superoxide dismutase (SOD), which can lower the concentration of superoxide ions. The concentration of CO2 in vivo is high (ca. 1 mM), and the rate constant for reaction of CO2 with -OONO is large (pH-independent k = 5.8 x 10(4) M(-l)s(-1)). Consequently, the rate of reaction of peroxynitrite with CO2 is so fast that most commonly used scavengers would need to be present at very high, near toxic levels

Oxidant and anti-oxidant effects on arachidonate metabolism by rabbit ocular tissues.

Ocular tissues have the capacity to metabolize arachidonate to prostaglandins and related materials, such as hydroxy arachidonate derivatives (HETEs) which are potent mediators of the inflammatory response. Reactive oxygen species are also present during the inflammatory response, resulting in an altered oxidative environment within the eye. This study was designed to evaluate the possible impact of the oxidant hydrogen peroxide and anti-oxidants, ascorbic acid and glutathione, upon arachidonate metabolism. It was found that hydrogen peroxide was observed to potently inhibit arachidonate metab

Bioinspired metal oxide particles as efficient wet air oxidation and photocatalytic oxidation catalysts for the degradation of acetaminophen in aqueous phase.

The catalytic performances of the biomimetic metal oxides were tested in photo Fenton-like oxidation and catalytic wet air oxidation processes. Biomimetic copper oxide, iron oxide, and cobalt oxide catalysts were prepared by using pollen grains as biotemplate. The surface characteristics of the biomimetic metal oxides were characterized. SEM micrographs of the biomimetic catalysts demonstrated that pollen grains were successfully mimicked by metal oxide structures. The influences of UV light intensity, catalyst loading, and the initial hydrogen peroxide concentration on acetaminophen degradati…

Beyond arsenite: Arsenite-oxidizing prokaryotes drive sulfur compound oxidation.

Arsenite-oxidizing prokaryotes (AOPs) contribute significantly to the biogeochemical processes governing arsenic cycling. Since AOPs can oxidize As(III) to As(V), markedly reducing arsenic toxicity and decreasing its migration potential, they have been widely utilized in the restoration of As(III)-contaminated environment and in the design of bioreactors for treating As(III)-contaminated groundwater. However, it is still unclear whether AOPs possess additional catalytic activities that may interfere with their bioremediation capacity, highlighting a critical knowledge gap that warrants further

Oxygen production via nitric oxide dismutation in diverse ammonia oxidizers.

Ammonia-oxidizing archaea (AOA) are widespread and highly abundant in nature. Despite their typical aerobic metabolism, they thrive in ecosystems where oxygen is scarce. Recently, the AOA Nitrosopumilus maritimus SCM1 was shown to produce oxygen and N2O from nitrite upon oxygen depletion, with N2O subsequently converted to dinitrogen (N2) supporting nitric oxide (NO) dismutation as the proposed metabolic pathway. Here, we explore the ability of other ammonia oxidizers, with diverse phylogenetic affiliations and from different environmental settings to produce oxygen via NO-dismutation. We stud

Reduction by Oxidation: Selective Hydrodehalogenation of Aryl Halides by Mediated Oxalate Oxidation.

Electro-organic reduction reactions are canonically carried out at a cathode at which a significant negative potential is applied. Specifically, at carbon electrodes, aryl bromides and chlorides undergo heterogeneous reduction in organic solvents at potentials more negative than -2 V vs E0' for the Fc/Fc+ couple (Fc = ferrocene). To decrease the overpotential for reduction reactions, homogeneous or heterogeneous electrocatalysis strategies are often employed. Here, we present an electrochemical method to reduce aryl bromides and chlorides that is initiated by an oxidation reaction at very mild

Low-Temperature Oxidation of Methane to Methanol on a Zeolitic Octahedral Metal Oxide.

Direct oxidation of methane to methanol utilizing molecular oxygen under mild conditions is an important yet challenging process due to the difficulty in activating of methane under such conditions. In this research, we report on zeolitic octahedral metal oxides based on cobalt vanadotungstates, which act as the catalysts for the oxidation of methane using molecular oxygen as the oxidant without co-reductants at a low temperature of 90 °C even as low as 60 °C. This catalytic process results in the high-yield production of methanol as the primary product. The micropores within the catalyst are …

Dysfunction of Microcirculation in Atherosclerosis: Implications of Nitric Oxide, Oxidative Stress, and Inflammation.

Cardiovascular diseases (CVDs) are the leading causes of death worldwide, and most of them are connected with atherosclerosis (AS). Hypertension (HT), hyperlipidemia (HPL), and hyperglycaemia (HG) are the main risk factors responsible for CVD and have become a significant public health issue. AS might be a prime causative factor in CVD, and it originates from endothelial cell dysfunction. On the other hand, the factors mentioned above might cause endothelial cell damage as a consequence of endothelial dysfunction (ED) or might be regarded as a consequence of ED. Thus, endothelial cells are cri

Investigation of oxidative-stress-induced axon beading using photo-oxidation.

Oxidative stress caused by excessive reactive oxygen species adversely affects cellular function. Here, we investigate axonal beading induced by the photo-oxidation of a membrane-based fluorescent dye. Our experiments show that, apart from using oxygen scavengers, chelation of free calcium, or stabilization of microtubules or actin filaments by pharmacological agents can mitigate oxidative-stress-induced beading. Furthermore, we take advantage of this light-induced oxidative stress to explore axonal responses to spatially confined perturbations. We demonstrate that low excitation causes locali

Dissolved organic carbon diminishes manganese oxide-driven oxidation of chromium.

Oxidation of chromium (Cr)-bearing minerals by manganese (Mn) oxides is viewed as the dominant mechanism controlling geogenic production of Cr(VI) and its contamination of groundwater. This process may be modulated by other chemical constituents found in the natural environment, but such confounding factors have not been quantified. Here, we evaluated the mechanism of Cr(III) oxidation by mixed-valence Mn oxide in the presence of citric and gallic acids, two natural organic matter (NOM) constituents commonly found in the soil environment. Incubation experiments showed that each organic acid en…

Methanol-Promoted Oxidation of Nitrogen Oxide (NOx) by Encapsulated Ionic Liquids.

The removal of nitrogen oxides (NOx) has been extensively studied due to their harmful effects to health and environment. In this work, encapsulated ionic liquids (ENILs) are used as catalysts for the NO oxidation at humid conditions and low temperatures. Hollow carbon capsules (CCap) were first synthesized to contain different amounts of 1-butyl-3-methylimidazolium nitrate IL ([bmim][NO3]), responsible for the catalytic oxidation. Then, the materials were characterized using different techniques, by analyzing microstructure, porosity, elemental composition, and thermal stability. The catalyti…

Comparison of Nitrogen Oxide Metabolism among Diverse Ammonia-Oxidizing Bacteria.

Ammonia-oxidizing bacteria (AOB) have well characterized genes that encode and express nitrite reductases (NIR) and nitric oxide reductases (NOR). However, the connection between presence or absence of these and other genes for nitrogen transformations with the physiological production of nitric oxide (NO) and nitrous oxide (N2O) has not been tested across AOB isolated from various trophic states, with diverse phylogeny, and with closed genomes. It is therefore unclear if genomic content for nitrogen oxide metabolism is predictive of net N2O production. Instantaneous microrespirometry experime

Photochemical Oxidative Growth of Iridium Oxide Nanoparticles on CdSe@CdS Nanorods.

We demonstrate a procedure for the photochemical oxidative growth of iridium oxide catalysts on the surface of seeded cadmium selenide-cadmium sulfide (CdSe@CdS) nanorod photocatalysts. Seeded rods are grown using a colloidal hot-injection method and then moved to an aqueous medium by ligand exchange. CdSe@CdS nanorods, an iridium precursor and other salts are mixed and illuminated. The deposition process is initiated by absorption of photons by the semiconductor particle, which results with formation of charge carriers that are used to promote redox reactions.
