MYH11 upregulation attenuates autophagy dysfunction through modulation of ATG4A in metabolic dysfunction-associated steatotic liver disease.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a globally prevalent chronic liver disorder for which effective therapeutic options are scarce. Autophagy is pivotal in sustaining hepatic lipid homeostasis, and cytoskeletal proteins modulate autophagic flux. However, the role of myosin heavy chain 11 (MYH11) in the pathogenesis of MASLD remains elusive. Bioinformatic mining of Gene Expression Omnibus (GEO) datasets identified MYH11 as a key autophagy-associated differentially expressed gene that exhibited robust diagnostic performance in MASLD. MYH11 expression was markedly

Nonalcoholic Steatohepatitis Exacerbates Inflammatory Bone Destruction and Macrophage NLR Family Pyrin Domain-Containing 3 Inflammasome Activation in Ligature-Induced Periodontitis.

Metabolic disorders and accompanying complications pose considerable risks to systemic organ health. Clinical studies have revealed a significant correlation between nonalcoholic steatohepatitis (NASH) and increased severity of periodontitis. However, directed mechanistic insights remain limited. This study aimed to elucidate the direct impact of NASH on periodontitis progression and to investigate the underlying mechanisms using both in vivo and in vitro models. In mice, a NASH diet induced severe liver damage and steatosis compared with chow-fed controls. Furthermore, NASH significantly exac

CD36 inhibition corrects lipid-FetuinA mediated insulin secretory defects by preventing intracellular lipid accumulation and inflammation in the pancreatic beta cells.

CD36 is a multifunctional protein involved in long chain fatty acid uptake and immune modulation in different cells. Recently it was reported that increased expression of CD36 is evident in the islets of diabetic obese individuals. In this present study we investigated the role of CD36 in regulating intracellular lipid accumulation and inflammation in beta cells and its implication on secretory dysfunction. Additionally, we have elucidated the potential role of fetuinA, a circulatory glycoprotein and an endogenous ligand of TLR4, for aggravating lipid accumulation and insulin secretory defects

Changes of flavin-containing monooxygenases and trimethylamine-N-oxide may be involved in the promotion of non-alcoholic fatty liver disease by intestinal microbiota metabolite trimethylamine.

Evidence shows that trimethylamine (TMA)/trimethylamine-N-oxide (TMAO) is closely related to non-alcoholic fatty liver disease (NAFLD). The conversion of TMA to TMAO is mainly catalyzed by flavin-containing monooxygenases 3 (FMO3) and FMO1. In this study, we explored the role of TMA in the process of NAFLD. The human NAFLD liver puncture data set GSE89632 and rat TMAO gene chip GSE135856 was downloaded for gene differential expression analysis. Besides, oleic acid (OA) combined with palmitate were used to establish high-fat cell model. TMA, TMAO and FMO1-siRNA were used to stimulate L02 cells.

Effects of feeding on plasma concentrations of vitamin A in captive African penguins (Spheniscus demersus).

Vitamin A comprises vitamin A1 and vitamin A2; vitamin A1 is retinol and its fatty-acid esters and vitamin A2 is 3,4-didehydroretinol and its fatty-acid esters. Although vitamin A1 is generally recognized as the major vitamin A, vitamin A2 is found in some birds and mammals that eat fish containing vitamin A2. Plasma concentration of retinyl esters, but not retinol, is known to increase postprandially in humans. The objectives of this study were to confirm the presence of vitamin A2 in fish fed to penguins, and in penguin plasma, and the postprandial changes in vitamin A concentration in pengu

Polydatin attenuates diet-induced nonalcoholic steatohepatitis and fibrosis in mice.

Scope: Non-alcoholic steatohepatitis (NASH) is characterized by lipid accumulation in hepatocytes and inflammatory cell infiltration. In view of the anti-oxidative and anti-inflammatory effects of polydatin, the current study aimed to investigate the pharmacological effects of polydatin on NASH and its related fibrosis. Methods: C57BL/6 mice were fed with methionine-choline deficient (MCD) diet to induce NASH and liver fibrosis, and treated with or without polydatin (5 mg/kg, every other day, i.p) for 4 weeks. HepG2 cells induced by palmitic acid (PA) were treated with polydatin. Results: The

Hydrogen-rich water protects against liver injury in nonalcoholic steatohepatitis through HO-1 enhancement via IL-10 and Sirt 1 signaling.

Nonalcoholic steatohepatitis (NASH) could progress to hepatic fibrosis in the absence of effective control. The purpose of our experiment was to investigate the protective effect of drinking water with a high concentration of hydrogen, namely, hydrogen-rich water (HRW), on mice with nonalcoholic fatty liver disease to elucidate the mechanism underlying the therapeutic action of molecular hydrogen. The choline-supplemented, l-amino acid-defined (CSAA) or the choline-deficient, l-amino acid-defined (CDAA) diet for 20 wk was used to induce NASH and fibrosis in the mice model and simultaneously tr

Instant controlled pressure drop processing modulates phenolic composition and fatty acid profile in Amaranthus hypochondriacus grain.

Instant Controlled Pressure Drop (DIC) is a high-pressure/short-time thermo-mechanical pretreatment that can expand grain structure and improve phytochemical accessibility. This study assessed how DIC conditions (0.10-0.40 MPa/10-90 s) modulate phenolic composition and lipid fingerprints in two Amaranthus hypochondriacus grain varieties ("Criollo" and "Laura") using a 13-run central composite design. Ethanolic extracts were profiled by HPLC, while GC-MS characterized the extracted oils. HPLC identified gallic acid, catechin, p-hydroxybenzoic acid, ellagic acid, trans-4-hydroxycinnamic acid, qu

Protective effects of whey camel milk proteins conjugate with metal-organic frameworks (MOF) as antimicrobial and against cobalt-induced chloragogen cell toxicity in earthworms.

Camel whey protein (CWP) is rich in potent antioxidant and antimicrobial compounds, while the iron-based metal-organic framework (MOF)-235 offers high biocompatibility for drug delivery. This study synthesised and characterised CWP/MOF-235 nano-conjugates to enhance CWP stability, bioavailability, and therapeutic efficacy. GC-MS analysis confirmed the presence of hepatoprotective and antimicrobial fatty acids in CWP, including oleic acid, palmitic acid methyl ester, and cis-vaccenic acid. Antimicrobial assays revealed that CWP/MOF-235 exhibited greater activity against Candida albicans than CW

Integrated transcriptome and metabolome study reveal the therapeutic effects of nicotinamide riboside and nicotinamide mononucleotide on nonalcoholic fatty liver disease.

Nicotinamide mononucleotide (NMN) and nicotinamide riboside (NR) have received considerable attention as anti-aging and anti-metabolic disease nutraceuticals. However, few studies have focused on their role in ameliorating hepatic metabolic disturbances. In the present study, the effects of NMN and NR on the liver of mice with nonalcoholic fatty liver disease (NAFLD) were investigated via transcriptome and metabolome analyses. NMN and NR reduced body weight gain, improved glucose homeostasis, regulated plasma lipid levels, and ameliorated liver injury, oxidative stress, and lipid accumulation

Targeted Mass Spectrometry Reveals Interferon-Dependent Eicosanoid and Fatty Acid Alterations in Chronic Myeloid Leukaemia.

Bioactive lipids are involved in cellular signalling events with links to human disease. Many of these are involved in inflammation under normal and pathological conditions. Despite being attractive molecules from a pharmacological point of view, the detection and quantification of lipids has been a major challenge. Here, we have optimised a liquid chromatography-dynamic multiple reaction monitoring-targeted mass spectrometry (LC-dMRM-MS) approach to profile eicosanoids and fatty acids in biological samples. In particular, by applying this analytic workflow to study a cellular model of chronic

NAFLD causes selective CD4(+) T lymphocyte loss and promotes hepatocarcinogenesis.

Hepatocellular carcinoma (HCC) is the second most common cause of cancer-related death. Non-alcoholic fatty liver disease (NAFLD) affects a large proportion of the US population and is considered to be a metabolic predisposition to liver cancer. However, the role of adaptive immune responses in NAFLD-promoted HCC is largely unknown. Here we show, in mouse models and human samples, that dysregulation of lipid metabolism in NAFLD causes a selective loss of intrahepatic CD4(+) but not CD8(+) T lymphocytes, leading to accelerated hepatocarcinogenesis. We also demonstrate that CD4(+) T lymphocytes

Palmitoylation of cysteine 69 from the COOH-terminal of band 3 protein in the human erythrocyte membrane. Acylation occurs in the middle of the consensus sequence of F--I-IICLAVL found in band 3 protein and G2 protein of Rift Valley fever virus.

One of the major physiologic functions of erythrocytes is the mediation of chloride-bicarbonate exchange in the transport of carbon dioxide from the tissues to the lungs. The anion exchange is mediated by a typical polytopic transmembrane protein in the cell membrane, designated Band 3. A carboxyl-terminal peptide of Band 3 was affinity-labeled with pyridoxal phosphate, a substrate for the anion transport system, and then sequenced (Kawano, Y., Okubo, K., Tokunaga, F., Miyata, T., Iwanaga, S., and Hamasaki, N. (1988) J. Biol. Chem. 263, 8232-8238). The 10th amino acid residue of the peptide co

Effects of term infant formulas containing high sn-2 palmitate with and without oligofructose on stool composition, stool characteristics, and bifidogenicity.

OBJECTIVES: Levels of stool fatty acid soaps and beneficial bacteria differ between formula-fed and breast-fed infants; addition of specific formula ingredients may reduce these differences. This study evaluated the effects of a term infant formula containing high sn-2 palmitate term infant formula (sn-2) or an identical formula supplemented with oligofructose (OF) at 2 concentrations (sn-2+3 g/L OF, sn-2+5 g/L OF) on stool composition, stool characteristics, and fecal bifidobacteria. METHODS: Healthy, term formula-fed infants 7 to 14 days old (n = 300) were randomized in a double-blind manne

Enhanced fatty acid oxidation in adipocytes and macrophages reduces lipid-induced triglyceride accumulation and inflammation.

Lipid overload in obesity and type 2 diabetes is associated with adipocyte dysfunction, inflammation, macrophage infiltration, and decreased fatty acid oxidation (FAO). Here, we report that the expression of carnitine palmitoyltransferase 1A (CPT1A), the rate-limiting enzyme in mitochondrial FAO, is higher in human adipose tissue macrophages than in adipocytes and that it is differentially expressed in visceral vs. subcutaneous adipose tissue in both an obese and a type 2 diabetes cohort. These observations led us to further investigate the potential role of CPT1A in adipocytes and macrophages

Metabolic remodelling of glucose, fatty acid and redox pathways in the heart of type 2 diabetic mice.

KEY POINTS: Hearts from type 2 diabetic animals display perturbations in excitation-contraction coupling, impairing myocyte contractility and delaying relaxation, along with altered substrate consumption patterns. Under high glucose and β-adrenergic stimulation conditions, palmitate can, at least in part, offset left ventricle (LV) dysfunction in hearts from diabetic mice, improving contractility and relaxation while restoring coronary perfusion pressure. Fluxome calculations of central catabolism in diabetic hearts show that, in the presence of palmitate, there is a metabolic remodelling invo
