The time course of production in the mouse circulation, after different schemes of immunization, of reagin-like and γ1 antibodies, as measured by the ability of the antisera to induce thermolabile long-latence and thermostable short-latence PCA reactions, has been studied. γ1 antibody was obtained in all schemes of immunization and its amount in antiserum increased with time. The reaginic antibody response to alum-precipitated ovalbumin (OA) and the dialyzed water-soluble extracts of ragweed (DWSR) and Alternaria tenuis (DWST) in several strains of rats appeared to be wholly an IgE response. The IgG1 and IgE homocytotropic antibody responses of LAF and C3H mice to timothy pollen antigens are defined. The inbred LOU/Wsl rat strain presents a high incidence of spontaneous ileocoecal immunocytoma which secrete monoclonal immunoglobulins. These tumours are transplantable in histocompatible animals and their secreting properties are maintained over many passages. A screening of monoclonal immunoglobulins secreted by 250 such tumours revealed sixty-three proteins which shared class-specific determinants, physicochemical features and biological properties different from those of rat IgM, IgA, IgG1, IgG2a, IgG2b and IgG2c classes, and characteristic of proteins of the IgE class.

Intracutaneous injection of the low molecular weight fragment (C3a) of the third component of human complement caused pruritus, wheal and axon-reflex erythema in human skin. The response was maximal at 5–10 min after injection and dose-dependent between 15 and 600 ng. On a molar basis, C3a was more effective than histamine. The influence of antigen dosage on reagin formation to haemocyanin in rabbits was studied. Using large doses of antigen, reagin formation was more readily elicited, the induction period was shorter and the persistence of reagins in serum was longer than when low antigen doses were employed. Reagin synthesis was readily induced after intravenous administration of antigen.
