α-Catenin phosphorylation is tension-sensitive and supports epithelial barrier functions via afadin.

Zonula adherens junctions (zAJs) are spatially proximal to tight junctions (TJs), in a superstructure known as the apical junctional complex (AJC). A key component of the AJC is a circumferential ring of F-actin, but how actomyosin contractility drives AJC structure and epithelial barrier function is incompletely understood. Here, we show that a central mechanosensitive component of zAJ, α-catenin (α-cat), undergoes force-dependent phosphorylation in an unstructured linker region. This modification in turn primes the α-cat middle region for effector binding. We credential afadin, a TJ/AJ scaff…

TBK1-mediated phosphorylation of WIPI2 links bacteria recognition to autophagosome formation in xenophagy.

Xenophagy, a selective autophagy pathway, is a critical innate immune defense mechanism that targets pathogens for lysosomal degradation. However, the molecular mechanisms enabling autophagosomes to specifically recognize and engulf bacteria remain incompletely understood. Here, we identify WIPI2, a core component that drives autophagosome biogenesis, as a novel phosphorylation substrate of TBK1 during Salmonella Typhimurium infection. We demonstrate that TBK1 phosphorylates WIPI2 at Ser96, which enhances its interaction with ATG16L1 and its binding to PtdIns3P. Crucially, the recruitment of W…

KCC2 phosphorylation dynamics relate to oligomerization during development and after spinal cord injury.

Effective synaptic inhibition relies on KCC2, a K+-Cl- cotransporter that forms oligomers to extrude chloride. However, how phosphorylation regulates oligomerization remains unclear. To address this, we studied the phosphorylation of KCC2 in the lumbar spinal cord of neonatal rats by subcellular fractionation and phospho-specific western blotting across three paradigms with distinct expression profiles: developmental upregulation, spinal cord injury (SCI)-induced downregulation, and pharmacological rescue with the 5-HT2A/2C agonist DOI. KCC2 moved from cytoplasm to membrane during development,…

Histamine H1 Receptor-Mediated CREB Phosphorylation via Gq Protein Signaling and Arrestin Modulation.

Background/Objectives: Histamine H1 receptors mediate multiple physiological and pathophysiological processes, including inflammation and allergy, by regulating downstream gene expression via transcription factors. cAMP response element-binding protein (CREB) is a major transcription factor whose phosphorylation is regulated by multiple signaling pathways. Although CREB is closely involved in multiple physiological and pathophysiological processes, the detailed intracellular signaling pathway of H1 receptor-mediated CREB phosphorylation remains to be elucidated. We investigated the roles of Gq…

Integrative phosphoproteomic analysis reveals co-regulatory phosphorylation networks of rhotekin in cancer progression.

Rhotekin (RTKN), a Rho GTPase effector, promotes the development of malignancies, including breast, gastric, and colon cancers, by enhancing cell proliferation and migration while inhibiting apoptosis. Despite its oncogenic role, the phosphoregulatory network of RTKN remains largely unexplored, with no experimental evidence on its upstream kinases and functional phosphosites. To characterize RTKN-associated phosphorylation dynamics, PubMed-indexed studies were systematically retrieved using predefined MeSH terms to compile large-scale cellular phosphoproteomics datasets. Analysis of 618 quanti…

Characterization of posttranslational modification in metabolic dysfunction-associated steatohepatitis: acetylation, lactylation, and phosphorylation.

In metabolic dysfunction-associated steatohepatitis (MASH), abnormalities in posttranslational modification (PTM) are both a consequence of the pathological process and a driving force for disease progression. Therefore, this study intends to characterize the PTM features (acetylation, lactylation, and phosphorylation) of MASH, which will provide a crucial theoretical basis for the diagnosis and development of new targets for MASH. In this study, protein/modification profiles of clinical liver tissues from Normal and MASH were analyzed by LC-MS to identify differentially expressed proteins (DE…

ANOS1 Facilitated Tumorigenesis in Gastric Cancer through the Regulation of Oxidative Phosphorylation Pathway.

Gastric cancer (GC) is the fifth leading cause of cancer-related deaths, with poor early detection and prognosis. Biomarkers related to its … oted migration, invasion, and proliferation. Mechanistically, knockdown of ANOS1 promotes oxidative phosphorylation via the receptor tyrosine kinase pathway, suppresses glycolysis, and inhibits the proliferative, migratory, and invasive capacities of tumor cells. The results of immune infiltration analysis revealed that ANOS1 expression was exhibited a positive c …

Procyanidin Capsules Relieve Inflammation in Hypertriglyceridemic Acute Pancreatitis via Regulating Oxidative Phosphorylation Signaling.

Hypertriglyceridemic acute pancreatitis (HAP) is characterized by mitochondrial oxidative phosphorylation (OXPHOS) impairment and excessive reactive oxygen species (ROS), leading to severe inflammation and organ damage. Existing therapies often lack targeted antioxidant efficacy and long-term stability. In this study, we innovatively designed and synthesized procyanidin capsules using a one-step calcium carbonate template method, which enables sustained release of procyanidin for over one month and exhibits exceptional scavenging capacity for both ROS and RNS. The capsules demonstrated high bi…

Targeting NDUFS4 Disrupts Oxidative Phosphorylation and Induces Ferroptosis in Olaparib-Resistant Prostate Cancer.

Resistance to poly(ADP-ribose) polymerase inhibitors (PARPi) remains a major challenge in the treatment of advanced prostate cancer. Although metabolic rewiring has been implicated in this process, the molecular drivers and therapeutic vulnerabilities underlying this adaptation remain poorly defined. We integrated transcriptomic, functional, and clinical analyses to identify mitochondrial regulators of PARPi resistance. RNA sequencing and gene set enrichment analysis revealed robust enrichment of oxidative phosphorylation (OxPhos) pathways in PARPi-resistant prostate cancer cells, with consist…

Olfactory Dysfunction Exacerbates Hippocampal Aβ Accumulation, Tau Phosphorylation and Memory Deficits in Mice.

Olfactory dysfunction is a frequent feature in patients with neurodegenerative disorders such as Alzheimer’s disease (AD). However, whether olfactory impairment is the cause or consequence of AD is unknown. We previously found that olfactory dysfunction impairs learning and memory in mice in multiple experimental paradigms, but whether olfactory dysfunction increases AD-related neuropathological changes such as Aβ deposition and tau protein phosphorylation is not clear. In this study, mice were treated with bilateral intranasal zinc sulfate (ZnSO4) solution infusion, which resulted in olfactor…

Site-specific phosphorylation of Ser352 drives aggregation of Tau R4 under acidosis conditions.

In Alzheimer's disease, neurons undergo acidosis as their pH drops from ∼7.1 to ∼6.5. Here we elucidate the molecular mechanism of specific Tau aggregation only at this lower pH. We show that a specific phosphorylation event in the Tau R4 domain is coupled to this pH drop to drive a defined phase transition from condensates to fibrils. Using a combination of experimental and computational studies, our results demonstrate that site-specific phosphorylation of Ser352 is the molecular switch that induces aggregation of Tau only at the more acidic, disease-related pH. We designed and synthesized a…

Src-mediated PHB2 phosphorylation disrupts mitochondrial cristae through cardiolipin dissociation in hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) displays mitochondrial dysfunction characterized by disrupted redox homeostasis and cristae disorganization, … eakage and oxidative stress. This mitochondrial dysfunction drives a metabolic shift from oxidative phosphorylation toward glycolysis, promoting tumor growth in xenograft models. Phosphomimetic PHB2 mutants (Y34E/Y77E) exacerbate these effects, whereas phosphorylation-resistant mutants (Y34F/Y77F) restore cristae integrity, normalize redox balance, and suppress t …

Copper-Catalyzed Enantioselective C-H Phosphorylation Enabled by a 3,3'-Difluorinated BINOL Ligand.

The asymmetric construction of C-P bonds via transition-metal-catalyzed enantioselective C-H activation remains a significant and unresolved challenge. Herein, we report an unprecedented copper-catalyzed enantioselective C-H phosphorylation through oxidative C-H/P-H coupling. This method employs earth-abundant copper catalyst and readily accessible 3,3'-difluorinated BINOL (1,1'-bi-2-naphthol) ligand, which proved essential for achieving high reactivity and enantioselectivity. A broad range of unmasked H-phosphonates and secondary phosphine oxides (SPOs) serve as practical coupling partners, d…

Structure-guided optimisation of fenofibrate-derived oxidative phosphorylation inhibitors to modify tumour hypoxia.

Solid tumours frequently manifest regions of abnormally low levels of oxygen (hypoxia), which negatively impacts cancer treatment outcomes. This is particularly detrimental to radiotherapy which requires oxygen to exert maximal therapeutic effects. Tumour hypoxia can be abolished by reducing oxygen consumption rates (OCR) through inhibition of oxidative phosphorylation (OXPHOS), though to date no hypoxia modifying OXPHOS inhibitors have successfully translated into routine clinical practise. Here, we demonstrate that the well-tolerated, pro-drug fenofibrate, which has moderate OXPHOS inhibitor…

Myocardial mitochondrial morphology and oxidative phosphorylation capacity in wild-type transthyretin amyloid cardiomyopathy.

BACKGROUND: The effects of amyloid deposition on myocyte mitochondrial morphology and oxidative function in wild-type transthyretin cardiac amyloidosis (ATTRwt) are poorly understood. METHODS: Endomyocardial biopsies of patients with ATTRwt were investigated with electron microscopy to determine mitochondrial structure, numbers, and size. High-resolution respirometry was used to determine mitochondrial oxidative phosphorylation. Amyloid load was assessed using both the stereological counting method, cardiac resonance imaging, and echocardiography. Regression analysis was performed to assess a…

Antiandrogen therapy induces mitochondrial oxidative phosphorylation to promote castration resistance in prostate cancer.

Tumor recurrence and therapy resistance are frequently accompanied by alterations in cellular metabolism. However, how metabolic remodeling occurs and contributes to castration-resistant prostate cancer (CRPC) remains largely elusive. Here, we demonstrate that mitochondrial oxidative phosphorylation (OXPHOS) is critical for development of androgen receptor signaling inhibitors (ARSI) resistance. Our findings indicate that prostate cancer cells exhibit increased mitochondrial OXPHOS following ARSI treatment. Notably, there is no significant change in glycolytic activity. Importantly, this metab…
