Systemic resistance and lipoxygenase-related defence response induced in tomato by Pseudomonas putida strain BTP1.

BACKGROUND: Previous studies showed the ability of Pseudomonas putida strain BTP1 to promote induced systemic resistance (ISR) in different host plants. Since ISR is long-lasting and not conducive for development of resistance of the targeted pathogen, this phenomenon can take part of disease control strategies. However, in spite of the numerous examples of ISR induced by PGPR in plants, only a few biochemical studies have associated the protective effect with specific host metabolic changes. RESULTS: In this study, we showed the protective effect of this bacterium in tomato against Botrytis

Inactivated Cowpea Mosaic Virus in Combination with OX40 Agonist Primes Potent Antitumor Immunity in a Bilateral Melanoma Mouse Model.

Viral immunotherapies are being recognized in cancer treatment, with several currently approved or undergoing clinical testing. While contemporary approaches have focused on oncolytic viral therapies, our efforts center on the development of plant virus-based cancer immunotherapies. In a previous work, we demonstrated the potent efficacy of the cowpea mosaic virus (CPMV), a plant virus that does not replicate in animals, applied as an in situ vaccine. CPMV is an immunostimulatory drug candidate, and intratumoral administration remodels the tumor microenvironment leading to activation of local

Characterization, genetic analysis and gene identification of an EMS-mutagenized lesion-mimic mutant with enhanced powdery mildew resistance in bread wheat (Triticum aestivum L.).

Lesion mimic mutants (LMMs) are ideal for dissecting plant immunity mechanisms. Here, we characterized lm10373, a stable LMM isolated from an EMS-induced library of wheat cultivar AK58. lm10373 developed light-dependent lesion spots on leaves from the late tillering stage, accompanied by programmed cell death (PCD) and reactive oxygen species (ROS) accumulation. Phenotypically, lm10373 showed reduced photosynthetic capacity and yield-related traits, but enhanced powdery mildew resistance at the heading stage. Genetic analysis revealed the lesion trait was controlled by a single semi-dominant n…

Baicalin enhances respiratory mucosal immunity by modulating antiviral protein expression and T-cell homeostasis during H9N2 infection.

INTRODUCTION: The H9N2 avian influenza virus, although not highly pathogenic, still poses ongoing risks to poultry health and food security due to its ability to resist vaccines and its potential to spread to humans. METHODS: This study investigated the effects of baicalin, a flavonoid derived from Scutellaria baicalensis, on respiratory mucosal immunity during H9N2 infection. In vitro experiments were conducted using MLE-12 alveolar epithelial cells, and in vivo evaluations were performed in a mouse model of H9N2 infection. RESULTS: Baicalin treatment enhanced the expression of antiviral pr

Revisiting the Immunometabolic Basis for the Metabolic Syndrome from an Immunonutritional View.

Metabolic syndrome (MetS) implies different conditions where insulin resistance constitutes a major hallmark of the disease. The disease incurs a high risk for the development of cardiovascular complications, and takes its toll in regard to the gut-liver axis (pancreas, primary liver and colorectal)-associated immunity. The modulation of immunometabolic responses by immunonutritional factors (IFs) has emerged as a key determinant of the gut-liver axis' metabolic and immune health. IFs from plant seeds have shown in vitro and pre-clinical effectiveness primarily in dealing with various immunome

Highly immunogenic influenza virus-like particles containing B-cell-activating factor (BAFF) for multi-subtype vaccine development.

Virus-like particle (VLP) technology is an attractive platform for the development of seasonal and pandemic influenza vaccines. Influenza VLPs can be obtained by the overexpression of HA, M1, NA, and/or M2 viral proteins in insect, mammalian, or plant cells. In this study, we reported to obtain highly immunogenic influenza VLPs by molecular incorporation with B-cell-activating factor (BAFF) or proliferation-inducing ligand (APRIL). Since BAFF and APRIL act as homotrimers to interact with their receptors, we engineered the VLPs by direct fusion of BAFF or APRIL to the transmembrane anchored dom

Arbuscular Mycorrhizal Fungi: Boosting Crop Resilience to Environmental Stresses.

Amid escalating challenges from global climate change and increasing environmental degradation, agricultural systems worldwide face a multitude of abiotic stresses, including drought, salinity, elevated temperatures, heavy metal pollution, and flooding. These factors critically impair crop productivity and yield. Simultaneously, biotic pressures such as pathogen invasions intensify the vulnerability of agricultural outputs. At the heart of mitigating these challenges, Arbuscular Mycorrhizal Fungi (AM fungi) form a crucial symbiotic relationship with most terrestrial plants, significantly enhan

Coated Hematite Nanoparticles Alleviate Iron Deficiency in Cucumber in Acidic Nutrient Solution and as Foliar Spray.

Micronutrient iron (Fe) deficiency poses a widespread agricultural challenge with global implications. Fe deficiency affects plant growth and immune function, leading to reduced yields and contributing to the global "hidden hunger." While conventional Fe-based fertilizers are available, their efficacy is limited under certain conditions. Most recently, nanofertilizers have been shown as promising alternatives to conventional fertilizers. In this study, three nanohematite/nanoferrihydrite preparations (NHs) with different coatings were applied through the roots and shoots to Fe-deficient cucumb

Novel perspectives on cysteine-dependent enzymes and processes in photosynthetic organisms.

Present in various subcellular compartments, cysteine is the major source of reduced sulfur and thus represents a key metabolite for various biosynthetic pathways as well as for redox homeostasis as a component of glutathione. As photosynthetic organisms assimilate inorganic sulfate and reduce it into sulfide before its incorporation into cysteine, there are strong relationships between cysteine homeostasis and all pathways involved in its synthesis and utilization. Over the last decade, cysteine degradation leading to hydrogen sulfide release has been linked to different physiological respons

Natural biomolecules and derivatives as anticancer immunomodulatory agents.

Despite advancements in chemotherapy, the issue of resistance and non-responsiveness to many chemotherapeutic drugs that are currently in clinical use still remains. Recently, cancer immunotherapy has gathered attention as a novel treatment against select cancers. Immunomodulation is also emerging as an effective strategy to improve efficacy. Natural phytochemicals, with known anticancer properties, been reported to mediate their effects by modulating both traditional cancer pathways and immunity. The mechanism of phytochemical mediated-immunomodulatory activity may be attributed to the remode

Inducers of plant systemic acquired resistance regulate NPR1 function through redox changes.

NPR1 is an essential regulator of plant systemic acquired resistance (SAR), which confers immunity to a broad-spectrum of pathogens. SAR induction results in accumulation of the signal molecule salicylic acid (SA), which induces defense gene expression via activation of NPR1. We found that in an uninduced state, NPR1 is present as an oligomer formed through intermolecular disulfide bonds. Upon SAR induction, a biphasic change in cellular reduction potential occurs, resulting in reduction of NPR1 to a monomeric form. Monomeric NPR1 accumulates in the nucleus and activates gene expression. Inhib

Expression of a conserved synthetic multiepitopic malaria protein CS712 in duckweed: immune response evaluation after oral delivery in BALB/c mice.

BACKGROUND: Malaria remains a significant global health challenge, with hundreds of millions of cases reported annually and limited access to effective and affordable vaccines. Owing to their scalability, safety, and potential for oral administration, plant-based vaccine platforms have emerged as promising alternatives. In this study, we evaluated the immunogenicity of a conserved, synthetic, and multiepitopic chimeric circumsporozoite protein, CS712, derived from Plasmodium vivax and expressed in transgenic Lemna minor, as a candidate antigen for oral malaria vaccine development. Immune regulation by polysaccharides: implications for skin cancer.

UV radiation causes sunburn, premature aging of the skin and is the major environmental carcinogen for squamous cell and basal cell skin cancer in humans. Besides causing mutations in DNA, UV radiation contributes to carcinogenesis by suppressing immune responses to highly antigenic, newly arising neoplasms. Strategies aimed at preventing UV-induced immune suppression, the mechanism of action of the agents used, and the significance of immune protection for prevention of skin cancer are reviewed. This review focuses on the use of plant polysaccharides to prevent immune damage triggered by UV r

Schistosoma mansoni-infected mice produce antibodies that cross-react with plant, insect, and mammalian glycoproteins and recognize the truncated biantennaryN-glycan Man3GlcNAc2-R.

To reveal the role of cross-reactive carbohydrate determinants in the host immune response in helminth infections and allergenicity, we developed monoclonal antibodies (mAbs) that recognize glycan epitopes present on glycoconjugates from both helminths and plants. An IgM mAb (100-4G11-A) was selected from a panel of anti-glycan mAbs generated from Schistosoma-infected or immunized mice because it recognized both a plant glycoprotein horseradish peroxidase and phospholipase A2 from honeybee venom. On further characterization, it was shown that mAb 100-4G11-A recognizes the truncated biantennary

Mechanistic and Therapeutic Approach of Plants as an Anti-inflammatory Agent: A Review.

Chronic inflammation is a prolonged immune response that leads to the development of numerous diseases, including arthritis, cardiovascular disorders, and metabolic syndromes. Currently, synthetic anti-inflammatory medications are widely used to treat chronic inflammation due to their high efficacy. However, their long-term use is often associated with serious side effects. This led to an urgent need to explore the anti-inflammatory drugs that are more efficient and exhibit few or no adverse effects. Several studies have revealed the anti-inflammatory properties of various plants. However, mos

Overexpression of a small GTP-binding protein Ran1 in Arabidopsis leads to promoted elongation growth and enhanced disease resistance against P. syringae DC3000.

Plants resist infection through an innate immune response, which is usually associated with slowing of growth. The molecular mechanisms underlying the trade-off between plant growth and defense remain unclear. The present study reveals that growth/defense trade-offs mediated by gibberellin (GA) and salicylic acid (SA) signaling pathways are uncoupled during constitutive overexpression of transgenic AtRAN1 and AtRAN1Q72L (active, GTP-locked form) Arabidopsis plants. It is well known that the small GTP-binding protein Ran (a Ras-related nuclear protein) functions in the nucleus-cytoplasmic trans
