Do Melanocytes Have a Role in Controlling Epidermal Bacterial Colonisation and the Skin Microbiome?

In addition to producing melanin to protect epidermal keratinocytes against DNA damage, melanocytes may have important roles in strengthening innate immunity against pathogens. We have developed a functional, pigmented, human full-thickness 3D skin equivalent to determine whether the presence of melanocytes impacts epidermal bacterial growth and regulates the expression of genes involved in the immune response. We introduced primary epidermal melanocytes to construct a 3-cell full-thickness skin equivalent with primary dermal fibroblasts and epidermal keratinocytes. Immunohistochemistry verifi

Activity of cloxacillin against Cutibacterium acnes: MIC evaluation and utility of gradient test.

BACKGROUND: Cutibacterium acnes is a cause of implant-associated infections, often co-occurring with methicillin-susceptible Staphylococcus aureus (MSSA), especially in orthopedic settings. In Scandinavia, cloxacillin may be used as initial empiric intravenous treatment if MSSA implant-associated infection is suspected. Clinical breakpoints for C. acnes susceptibility to cloxacillin or other isoxazolyl-penicillins are lacking to guide therapy if C. acnes is subsequently cultured after empiric cloxacillin therapy has been initiated. OBJECTIVE: We aimed to compare MIC distributions of C.acnes t

Unveiling acne microbiome signatures: the role of the microbiome and the impact of a facewash formula containing plant-derived monoterpenes.

BACKGROUND: Acne vulgaris is a common skin disorder associated with a dysbiotic skin microbiome. While changes in the skin microbiome profile in acne have been reported, its role in acne pathogenesis and resolution, as well as the potential to rebalance the acne microbiome through facewash products, remain to be elucidated. OBJECTIVES: To investigate the microbiome signatures of participants with acne, at baseline and 4 weeks after intervention with a facewash product containing plant-derived monoterpenes, and 1 week after a regression phase with no product application. METHODS: Volunteers w

Novel antimicrobial peptides against Cutibacterium acnes designed by deep learning.

The increasing prevalence of antibiotic resistance in Cutibacterium acnes (C. acnes) requires the search for alternative therapeutic strategies. Antimicrobial peptides (AMPs) offer a promising avenue for the development of new treatments targeting C. acnes. In this study, to design peptides with the specific inhibitory activity against C. acnes, we employed a deep learning pipeline with generators and classifiers, using transfer learning and pretrained protein embeddings, trained on publicly available data. To enhance the training data specific to C. acnes inhibition, we constructed a phylogen

Intradermal injection of Cutibacterium acnes and staphylococcus: A pustular acne-like murine model.

BACKGROUND: Skin 16S microbiome diversity analysis indicates that the Staphylococcus genus, especially Staphylococcus aureus (S. aureus), plays a crucial role in the inflammatory lesions of acne. However, current animal models for acne do not fully replicate human diseases, especially pustular acne, which limits the development of anti-acne medications. AIMS: The aim is to develop a mouse model for acne, establishing an animal model that more closely mimics the clinical presentation of pustular acne. This will provide a new research platform for screening anti-acne drugs and evaluating the ef

Sarcoidosis Associated with Enlarged Mediastinal Lymph Nodes with the Detection of Streptococcus gordonii and Cutibacterium acnes Using a Clone Library Method.

A 77-year-old Japanese woman with mediastinal lymphadenopathy and uveitis was diagnosed with sarcoidosis. The bacterial flora in biopsied samples from mediastinal lymph nodes was analyzed using a clone library method with Sanger sequencing of the 16S rRNA gene, and Streptococcus gordonii (52 of 71 clones) and Cutibacterium acnes (19 of 71 clones) were detected. No previous study has conducted a bacterial floral analysis using the Sanger method for the mediastinal lymph node in sarcoidosis, making this case report the first to document the presence of S. gordonii and C. acnes in the mediastinal

Preventative effects of antioxidants on changes in sebocytes, outer root sheath cells, and Cutibacterium acnes-pretreated mice by particulate matter: No significant difference among antioxidants.

OBJECTIVES: Particulate matter (PM) is an air pollutant that can damage human skin; antioxidants have shown some efficacy in alleviating PM-induced skin inflammation. We investigated the antioxidant effects of punicalagin, epigallocatechin-3-gallate (EGCG), and resveratrol on PM-induced changes in cultured human sebocytes, outer root sheath (ORS) cells, and Cutibacterium acnes-pretreated mice. METHODS: Sebocytes and ORS cells were cultured with 100 μg/mL PM10 and 5 μM punicalagin, 1 μM EGCG, or 1 μM resveratrol for 24 h. In C. acnes-pretreated mice, inflammatory nodules were treated with 100

[Effect of IL-1β and NLRP3 on the inflammatory response of acne vulgaris].

To investigate the pathogenesis of acne vulgaris, and to provide new ideas for non-antibiotic therapy for acne vulgaris.
Methods: Normal human epidermal keratinocyte (NHEK) was exposed to Propionibacterium acnes (P. acnes) [multiplicity of infection (MOI)=10, 20, 30] for 12, 24, or 36 hours. The enzyme-linked immunosorbent assay (ELISA) and real-time PCR were used to detect the protein and mRNA of IL-1β in NHEK. Three groups were set up as follows: A negative control group (no NHEK pretreatment), a positive control group (P. acnes was used to stimulate NHEK), and a siRNA group (pretreated NHE…

Clinical evidence for washing and cleansers in acne vulgaris: a systematic review.

PURPOSE: Washing and over-the-counter cleansers are common interventions in acne vulgaris (AV), but the clinical evidence for their benefit is poorly understood. This systematic review presents clinical studies of washing and cleanser efficacy in acne vulgaris to guide treatment recommendations of dermatologists. MATERIALS AND METHODS: We surveyed English-language articles indexed in MEDLINE (1951-March 2017) and EMBASE (1974-March 2017). Articles were required to be prospective studies of a single over-the-counter cleanser or washing intervention in AV with an objective AV outcome measuremen

Bacteriological resistance in acne: A call to action.

Acne is common in adolescence but is also increasingly seen in adulthood, with about 40% of adults being affected. Topical and systemic oral antibiotics have been used for more than 40 years in the treatment of acne lesions. In the 1970s, evidence of resistance to topical erythromycin and clindamycin was reported and, since then, antibiotic resistance in acne has been increasing worldwide. Antibiotic exposure can be significant in acne treatment because the patient population is large and there is a tendency for prolonged treatment regimens to be prescribed. The overuse of antibiotics is now c

Infections associated with orthopedic implants.

PURPOSE OF REVIEW: We review recent advances in the prevention, diagnosis and treatment of infections associated with joint prostheses and internal fixation devices. RECENT FINDINGS: The perioperative antimicrobial prophylaxis should be administered 60-30 min before incision or before inflation of the tourniquet. New diagnostic approaches include sonication of removed implants to dislodge adherent microorganisms growing in biofilms and the use of molecular techniques to improve diagnostic yield. Treatment of implant-associated infections without removal of the device is an established option

Non-specific resistance to Babesia divergens in the Mongolian gerbil (Meriones unguiculatus).

Inoculation of mature gerbils with BCG gave protection to subsequent infection with B. divergens when inoculated by the intracardiac and intraperitoneal routes, the latter showing a dose dependent relationship. BCG vaccination was most effective in immature gerbils (less than 4 weeks old), which are innately resistant to B. divergens. Vaccination of gerbils with killed Propionesbacterium acne and zymosan A failed to elicit a protective response, which contrasts conspicuously with rodent babesia studies. Incubation of B. divergens-infected gerbil blood with hydrogen peroxide produced parasite i

Effect of betamethasone valerate on the normal human facial skin flora.

Eighteen volunteers were randomly divided into two groups and allocated either an active corticosteroid preparation (Betamethasone valerate) or the basal formulation only (placebo). The cream was applied to the face twice daily for one month. The treated area was sampled by the scrub-wash method immediately before treatment began and after 2 and 4 weeks, and microorganisms were enumerated and identified. Application of either cream produced a very slight increase (less than or equal to 0.5 log cycle) in the skin flora during the first 2 weeks of treatment. There were no significant differences

The prevalence of anaerobic bacteria in suprapubic bladder aspirates obtained from pregnant women.

The incidence of asymptomatic bacteriuria due to aerobic, facultative anaerobic and strictly anaerobic bacteria during pregnancy was determined in 185 patients admitted for complications of pregnancy, using suprapubic bladder aspiration. Anaerobic bacterial counts of 103 per ml urine or higher were detected in 6.4 per cent of the specimens. The incidence of anaerobic bacteriuria seems comparable to that of bacteriuria due to aerobic and facultative anaerobic micro organisms. In our group of patients the incidence of the latter was 5.4%. The presence of anaerobic bacteria was not confined to th
