Systemic effects of orally-administered zinc and tin (IV) metalloporphyrins on heme oxygenase expression in mice.

Some metalloporphyrins (Mps) inhibit heme oxygenase (HO), the rate-limiting enzyme in the production of bilirubin, and are potential compounds for the treatment of neonatal jaundice. We studied the safety and efficacy of Mps following oral administration. Adult HO-1-luc reporter mice were administered 30 micromol/kg body weight of tin mesoporphyrin (SnMP), zinc bis glycol deuteroporphyrin (ZnBG), or zinc protoporphyrin (ZnPP), or vehicle by oral gavage. Bilirubin production was measured as total body carbon monoxide (CO) excretion (VeCO). HO activity was quantitated via CO measurements by gas

Native Proteomics Reveals Protease-Mediated Hemophore Release by Bacterial Pathogens.

Iron is an essential micronutrient for nearly all forms of life, including pathogenic microbes that must acquire it from their host during infection. At the host-pathogen interface, humans restrict microbial access to iron through nutritional immunity, which many pathogens overcome by secreting hemophores that scavenge extracellular heme (iron protoporphyrin IX). However, identifying hemophores and other ligand-binding proteoforms in complex proteomes remains challenging using conventional peptide-based bottom-up mass spectrometry (MS). Here, we introduce ProteoMIX (Proteome Analysis by Mixing

Tumor/Lymph Node Dual-Targeting Ultrasonic Nanoconverter Orchestrates Spatiotemporal ROS Regulation for Dual-Zone Programmed Sono-STING Immunotherapy.

Tumor-draining lymph node (tdLN) metastasis remains a formidable challenge in treating breast cancer. Current anticancer treatments encounter difficulties in delivering therapeutic agents to tumors and tdLNs, impeding effective inhibition of tumor invasion and metastasis. Herein, a dual-targeting ultrasonic nanoconverter (OPD@PSF) is elaborately engineered through in situ polymerization to co-deliver a sonosensitizer protoporphyrin IX (PpIX) and a stimulator of interferon genes (STING) agonist Vadimezan (DMXAA) to achieve dual-zone programmed sono-STING immunotherapy (DPSSI) in tumors and tdLN

Combined autofluorescence and diffuse reflectance spectroscopy for rapid metabolic and vascular characterizations of orthotopic tongue tumors in vivo.

Precise label-free quantification of tissue metabolic and vascular dynamics in vivo represents a critical challenge for cancer therapy prediction and longitudinal treatment assessment. In this study, we demonstrated a portable autofluorescence and diffuse reflectance spectroscopy device along with novel spectroscopic algorithms to quantify tissue vascular and metabolic parameters of orthotopic head and neck cancer models in vivo. Tissue-mimicking phantom studies were used to verify the dual-modal optical spectroscopy and easy-to-use spectroscopic algorithms for rapid and accurate estimation of

Fluorescence Imaging of Cancer.

Optical imaging offers a high potential for noninvasive detection and therapy of cancer in humans. Recent advances in instrumentation for diffuse optical imaging have led to new capabilities for the detection of cancer in highly scattering tissue such as the female breast. In particular, fluorescence imaging is a promising method to image molecular probes in vivo. We review recent developments in the detection of breast cancer and fluorescence-guided surgery of the breast by contrast agents available for application on humans. Detection of cancer has been investigated with the unspecific contr

Post-Synthetic Functionalization of Porous Materials (MOFs and Silica) with Tetrapyrrole Macrocycles as Antiviral Materials against Coronaviruses and Hepatitis C Virus.

Mesoporous silica SBA-15 and two Metal-Organic Frameworks (MOFs), UiO-66(Zr)-NH2 and MIL-125(Ti)-NH2, were functionalized with tetrapyrrole macrocycles (TpM), two chlorin, and two porphyrin molecules with a simple coupling reaction, creating a strong covalent amide bond. Next, their antiviral activity was assessed against HCoV-229E, two highly pathogenic coronaviruses, SARS-CoV-2 and MERS-CoV, and Hepatitis C virus (HCV). Infection tests in vitro took place under dark or visible light exposure for different contact times, and it was found that 15 min of exposure for pheophorbide A (PhA)-functi

Identification of common heme-coordinating polymorphisms in human intracellular and secreted proteomes.

Heme (Iron II-Protoporphyrin IX) is the pigment of life among all organisms and as a prosthetic group in hemoproteins contributes to vital multifunctional processes. In plasma, free heme is scavenged by hemopexin, albumin and several other heme-binding proteins (HeBPs). Based on our earlier identification of human intracellular HeBPs with putative heme-binding motifs (HBMs) that exhibit pathology-associated encoded genetic variations, we expanded our efforts to cover hundreds of additional catalogued intracellular and secreted-extracellular species aiming to identify HeBPs carrying common poly

Combining 5-ALA-PDT with berbamine as an in vitro multimodal therapy approach against bladder cancer cells.

Bladder cancer is a common urogenital malignancy that remains difficult to treat, particularly due to therapeutic resistance, such as resistance to cisplatin, in which cancer stem cells (CSCs) play a central role. This study investigates the combination of 5-aminolevulinic acid-mediated photodynamic therapy (5-ALA-PDT) and berbamine as a potential multimodal treatment strategy using the bladder cancer cell lines RT112 and J82, their cisplatin-resistant variants, and generated CSC-like cells. Berbamine is a natural plant compound and was confirmed in this study to have anticancer properties by

Immunoblot-based activity assay for heme-containing histidine kinases.

Histidine kinases (HKs) are essential bacterial signal transduction proteins and attractive drug targets due to their critical cellular functions. Direct measurement of their autophosphorylation activity is crucial for developing inhibitors and advancing disease treatments. While [γ-32P]-ATP radiolabeling has long been a conventional method for kinase activity measurements, its reliance on 32P introduces inherent limitations. The isotope’s short half-life imposes time-sensitive constraints on experiments, and stringent radiation safety and compliance requirements significantly increase operati

Multi-wavelength photobleaching kinetics of endogenous fluorophores for label-free tissue characterization with potential for tumor margin estimation using machine learning.

Autofluorescence photobleaching encodes valuable information related to tissue morphology, metabolism, and fluorophore microenvironment; however, it remains mostly underexplored as a quantitative imaging biomarker. This study investigates multi-wavelength photobleaching kinetics of endogenous fluorophores, including NADH, FAD, lipofuscin, and protoporphyrin, as label-free contrast biomarkers for breast tissue characterization with potential for tumor margin estimation. Time-lapse fluorescence images are acquired using a spectral imaging system equipped with a liquid crystal tunable filter unde

Effects of Exposure to Ambient Air Pollution on Human Metabolome in Patients with Coronary Artery Disease.

Despite the established associations between air pollution and cardiovascular diseases (CVD), the biological mechanisms underlying its toxic effects remain largely unknown. We investigated whether exposure to both short- and long-term ambient air pollutants is associated with systemic perturbations in the human metabolome. Using a metabolome-wide association (MWAS) study framework with pathway enrichment and chemical annotation, we analyzed plasma samples from 244 patients with coronary artery disease (CAD) in the Emory Cardiovascular Biobank cohort. Individual residential exposures to air pol

Whole exome sequencing identifies rare variants involved in PPIX-metabolizing pathway in anti-TB drug-induced hepatitis.

AIMS: To screen and validate rare variants in the protoporphyrin IX (PPIX)-metabolizing pathway associated with anti-tuberculosis drug-induced hepatitis (AT-DIH). METHODS: A two-stage matched case-control study was conducted. Firstly, a 1:1 matching design (50 cases and 50 controls) was adopted to screen for rare variants using whole exome sequencing (WES). Secondly, a 1:4 matching design (132 cases and 528 controls) was employed to validate these rare variants via TaqMan genotyping. The association between these variants and AT-DIH risk was evaluated using odds ratios (ORs) with 95% confiden

Lipoic Acid Modulates Nrf2/HO-1 Pathway to Suppress Inflammation in Fungal Keratitis Through Host-Directed Cytoprotective Mechanisms.

Lipoic acid (LA), a dithiol antioxidant with an exemplary ocular safety profile, was investigated for its capacity to temper the oxidative-inflammatory milieu that drives fungal keratitis. Human corneal epithelial cells tolerated LA up to 60 µM without viability loss and, upon Aspergillus fumigatus challenge, LA induced a three-fold escalation of nuclear factor erythroid 2-related factor 2 (Nrf2) nuclear translocation and a five-fold surge in its downstream effector heme oxygenase-1 (HO-1). Pharmacological silencing of Nrf2 with ML385 or catalytic blockade of HO-1 with tin protoporphyrin IX ab

Spectrally Resolved and Highly Parallelized Raman Difference Spectroscopy for the Analysis of Drug-Target Interactions between the Antimalarial Drug Chloroquine and Hematin.

Malaria is a severe disease caused by cytozoic parasites of the genus Plasmodium, which infiltrate and infect red blood cells. Several drugs have been developed to combat the devastating effects of malaria. Antimalarials based on quinolines inhibit the crystallization of hematin into hemozoin within the parasite, ultimately leading to its demise. Despite the frequent use of these agents, there are unanswered questions about their mechanisms of action. In the present study, the quinoline chloroquine and its interaction with the target structure hematin was investigated using an advanced, highly

Iron Deficiency and Iron Deficiency Anemia in Women with and without Obesity: NHANES 2001-2006.

Obesity has been linked to numerous health and nutritional problems, including impaired iron metabolism, a common cause of anemia. We aimed to determine the prevalence of anemia, iron deficiency (ID), and iron deficiency anemia (IDA) among women aged 20-49 years based on body mass index (BMI) status. We used measures of iron status and body mass index from the 2001-2006 National Health and Nutrition Examination Survey (NHANES). Mean serum ferritin, erythrocyte protoporphyrin, and soluble transferrin receptor were higher, while those of serum iron, percent transferrin saturation, and mean cell

Consecutive Liver and Bone Marrow Transplantation for Erythropoietic Protoporphyria: Case Report and Literature Review.

BACKGROUND: Erythropoietic protoporphyria (EPP) is a rare inherited disease of heme biosynthesis resulting in the accumulation of protoporphyrin, characterized by liver failure in a minority of cases. Although liver transplant (LT) is the therapeutic strategy for advanced hepatic disease, it does not correct the primary defect, which leads to recurrence in liver graft. Thus, hematopoietic stem cell transplantation (HSCT) is an approach for treating EPP. METHODS: We aim to describe the first sequential LT and HSCT for EPP performed in Latin America, besides reviewing the present-day literature
