Interacting factors that influence long-term storage of live Pasteurella tularensis vaccine and Rift Valley fever virus.

Studies were conducted on the interaction of various parameters which affect the storage stability and growth potential of liquid cultures of Pasteurella tularensis live vaccine strain (LVS) and Rift Valley fever virus Van Wyk strain (RVFV). Storage variables studied with LVS included four storage temperatures (4, -20, -65, -175 C), single and multiple freeze-thaw cycles, two freezing and two thawing rates (slow and fast), various inoculum levels (1, 3, 5, and 10%) for the determination of growth potential, and the retention of immunizing potential (mice and guinea pig) after storage. Neither …

Systematic review and meta-analysis for the occurrence of Crimean-Congo hemorrhagic fever, Tularemia, and Rift Valley fever in pastoralist systems in Africa.

INTRODUCTION: Pastoralism is a livestock production system practiced in areas with harsh environmental conditions and is characterized by low investment in animal health, increasing the risk of animal disease outbreaks. Zoonotic diseases such as Crimean-Congo hemorrhagic fever (CCHF), Rift Valley fever (RVF), and tularemia may spread through livestock product value chains. METHODS: Published studies on the three diseases were systematically searched in PubMed, Web of Science, Scopus, and Google Scholar using predetermined search terms. Eligible studies were screened, and full texts were revie…

The Temperature-Associated Effects of Rift Valley Fever Virus Infections in Mosquitoes and Climate-Driven Epidemics: A Review.

Rift Valley fever virus (RVFV) is a mosquito-borne zoonotic disease within the genus Phlebovirus. Symptoms of the disease in animals range from moderate to severe febrile illness, which significantly impacts the livestock industry and causes severe health complications in humans. Similar to bunyaviruses in the genus Orthobunyavirus transmitted by mosquitoes, RVFV progression is dependent on the susceptibility of the physical, cellular, microbial, and immune response barriers of the vectors. These barriers, shaped by the genetic makeup of the mosquito species and the surrounding environmental t…

Incidence rate of Rift Valley fever exposure in humans and livestock from a longitudinal study in Northern Kenya.

Unravelling the mechanisms of Rift Valley fever virus (RVFV) maintenance in endemic areas during interepidemic periods is critical for enhancing early detection and response. Unfortunately, data on key epidemiological parameters, such as incidence rates, which are crucial for risk assessments and designing targeted interventions, are almost nonexistent. We conducted a longitudinal study of 1,938 pastoral livestock and 814 livestock keepers in an endemic region of northern Kenya from March 2022 to May 2023 to estimate the incidence rate of RVFV exposure and determine risk factors for infection.…

One Health investigation and response to a nationwide outbreak of Rift Valley fever in Rwanda - March to December 2022.

Rift Valley fever (RVF) is an emerging zoonotic mosquito-borne disease caused by Rift Valley Fever virus (RVFV), affecting both humans and animals. It is endemic to Rwanda and Tanzania and Uganda which are adjacent countries, with possible transboundary transmissions. Despite the various outbreak reports in Rwanda since 2012, information on the intensity and spread of these outbreaks and their management is scarce. We describe the 2022 outbreak that happened in Rwanda and provide insights into the One Health response implemented during the outbreak. There were no human cases officially reporte…

Serological Evidence of Antibodies to Rift Valley Fever Virus in Wild and Domestic Animals in Bauchi State, Nigeria.

Rift Valley fever (RVF) is an arthropod-borne zoonotic disease responsible for severe outbreaks in livestock and humans with concomitant economic losses in many countries in sub-Saharan Africa. The study, therefore, investigated the seroprevalence of the Rift Valley fever virus (RVFV) among wild and domestic animals. Blood samples were collected between 2013 and 2015 from 106 wild animals, 300 cattle (Bos indicus), and 200 horses (Equus caballus), respectively, in Yankari Game Reserve (YGR) and Sumu Wildlife Park (SWP) in Bauchi state, Nigeria. Harvested sera from blood were evaluated for the …

Characterization of the Molecular Interactions That Govern the Packaging of Viral RNA Segments into Rift Valley Fever Phlebovirus Particles.

Rift Valley fever phlebovirus (RVFV) has a single-stranded, negative-sense RNA genome, consisting of L, M, and S segments. The virion carries two envelope glycoproteins, Gn and Gc, along with ribonucleoprotein complexes (RNPs), composed of encapsidated genomes carrying N protein and the viral polymerase, L protein. A quantitative analysis of the profile of viral RNA segments packaged into RVFV particles showed that all three genomic RNA segments had similar packaging abilities, whereas among antigenomic RNA segments, the antigenomic S RNA, which serves as the template for the transcription of …

Evaluation of Fluorescence Microsphere Immunoassay for Detection of Antibodies to Rift Valley Fever Virus Nucleocapsid Protein and Glycoproteins.

Rift Valley fever virus (RVFV) is a mosquito-borne, zoonotic virus that infects ruminants, including cattle, sheep, goats, camels, and buffalo. Multiplexing diagnostic assays that can simultaneously detect antibodies against multiple RVFV antigens offer a high-throughput test for disease surveillance and vaccine evaluations. We describe the improvement and evaluation of a previously developed fluorescence microsphere immunoassay (FMIA) for the detection of IgG and IgM antibodies against the RVFV glycoprotein (Gn) and the immunogenic nucleocapsid protein (Np). Well-characterized vaccinated and …

Vascular permeability in the brain is a late pathogenic event during Rift Valley fever virus encephalitis in rats.

Rift Valley fever virus (RVFV) is a zoonotic disease of livestock that causes several clinical outcomes in people including febrile disease, hemorrhagic fever, and/or encephalitis. After aerosol infection with RVFV, Lewis rats develop lethal encephalitic disease, and we use this as a model for studying disease mechanisms of RVFV infection in the brain. Permeability of the brain vasculature in relation to virus invasion and replication is not known. Here, we found that vascular permeability in the brain occurred late in the course of infection and corresponded temporally to expression of matrix…

The first imported case of Rift Valley fever in China reveals a genetic reassortment of different viral lineages.

We report the first imported case of Rift Valley fever (RVF) in China. The patient returned from Angola, a non-epidemic country, with an infection of a new reassortant from different lineages of Rift Valley fever viruses (RVFVs). The patient developed multiorgan dysfunction and gradually recovered with continuous renal replacement therapy and a short regimen of methylprednisolone treatment. The disordered cytokines and chemokines in the plasma of the patient revealed hypercytokinemia, but the levels of protective cytokines were low upon admission and fluctuated as the disease improved. Whole-g…

Biologically Informed Individual-Based Network Model for Rift Valley Fever in the US and Evaluation of Mitigation Strategies.

Rift Valley fever (RVF) is a zoonotic disease endemic in sub-Saharan Africa with periodic outbreaks in human and animal populations. Mosquitoes are the primary disease vectors; however, Rift Valley fever virus (RVFV) can also spread by direct contact with infected tissues. The transmission cycle is complex, involving humans, livestock, and multiple species of mosquitoes. The epidemiology of RVFV in endemic areas is strongly affected by climatic conditions and environmental variables. In this research, we adapt and use a network-based modeling framework to simulate the transmission of RVFV amon…

[Infectious, immunogenic, and protective characteristics of the Rift Valley fever virus depending on the passage level and storage conditions].

The report discusses the research into the impact of some factors, especially the passage in a suspension of continuous cells BHK-21/13 and storage at different temperatures, upon immunobiological characteristics of the Rift Valley fever (RVF) virus strain 1974-VNIIVViM. The limits for the passage levels and optimal storage conditions providing maximal infectious and immunogenic activity, as well as protection of the attenuated RVF strain 1974- VNIIVViM, were determined. It was found that the RVF virus growth in VHK-21123 cell suspension in the course of 20 consecutive passages and storage at …

A model for the coupling of the Greater Bairam and local environmental factors in promoting Rift-Valley Fever epizootics in Egypt.

OBJECTIVES: Rift-Valley Fever (RVF) is a zoonotic mosquito-borne disease in Africa and the Arabian Peninsula. Drivers for this disease vary by region and are not well understood for North African countries such as Egypt. A deeper understanding of RVF risk factors would inform disease management policies. STUDY DESIGN: The present study employs mathematical and computational modeling techniques to ascertain the extent to which the severity of RVF epizootics in Egypt differs depending on the interaction between imported ruminant and environmentally-constrained mosquito populations. METHODS: An

Efficacy of a recombinant Rift Valley fever virus MP-12 with NSm deletion as a vaccine candidate in sheep.

Rift Valley fever virus (RVFV), a mosquito-borne virus in the Bunyaviridae family and Phlebovirus genus, causes RVF, a disease of ruminants and man, endemic in Sub-Saharan African countries. However, outbreaks in Yemen and Saudi Arabia demonstrate the ability for RVFV to spread into virgin territory and thus the need exists to develop safe and efficacious vaccines that can be used outside the endemic zones. Commercial RVFV vaccines are available but have limitations that prevent their use in disease-free countries. Consequently, there are ongoing efforts to develop and/or improve RVFV vaccines…

Simulation modelling of population dynamics of mosquito vectors for rift valley Fever virus in a disease epidemic setting.

BACKGROUND: Rift Valley Fever (RVF) is weather dependent arboviral infection of livestock and humans. Population dynamics of mosquito vectors is associated with disease epidemics. In our study, we use daily temperature and rainfall as model inputs to simulate dynamics of mosquito vectors population in relation to disease epidemics. METHODS/FINDINGS: Time-varying distributed delays (TVDD) and multi-way functional response equations were implemented to simulate mosquito vectors and hosts developmental stages and to establish interactions between stages and phases of mosquito vectors in relation…

Chemotactic and inflammatory responses in the liver and brain are associated with pathogenesis of Rift Valley fever virus infection in the mouse.

Rift Valley fever virus (RVFV) is a major human and animal pathogen associated with severe disease including hemorrhagic fever or encephalitis. RVFV is endemic to parts of Africa and the Arabian Peninsula, but there is significant concern regarding its introduction into non-endemic regions and the potentially devastating effect to livestock populations with concurrent infections of humans. To date, there is little detailed data directly comparing the host response to infection with wild-type or vaccine strains of RVFV and correlation with viral pathogenesis. Here we characterized clinical and …

A temperature-limited assessment of the risk of Rift Valley fever transmission and establishment in the continental United States of America.

The rapid spread of West Nile virus across North America after its introduction in 1999 highlights the potential for foreign arboviruses to become established in the United States of America. Of particular concern is Rift Valley fever virus (RVFV), which has been responsible for multiple African epidemics resulting in death of both humans and livestock, as well as major economic disruption due to livestock loss and trade restrictions. Modern globalization, travel, and commerce allow viruses to easily jump from one continent to another; and it is likely only a matter of time before RVFV reaches…

The hexamer structure of Rift Valley fever virus nucleoprotein suggests a mechanism for its assembly into ribonucleoprotein complexes.

Rift Valley fever virus (RVFV), a Phlebovirus with a genome consisting of three single-stranded RNA segments, is spread by infected mosquitoes and causes large viral outbreaks in Africa. RVFV encodes a nucleoprotein (N) that encapsidates the viral RNA. The N protein is the major component of the ribonucleoprotein complex and is also required for genomic RNA replication and transcription by the viral polymerase. Here we present the 1.6 Å crystal structure of the RVFV N protein in hexameric form. The ring-shaped hexamers form a functional RNA binding site, as assessed by mutagenesis experiments.…
