This study reports the results of the retention and quantitation of barbiturates in fresh tissue, in liver tissue fixed in formalin, and in the formalin solutions in which the same tissues were fixed for six months. BACKGROUND: Both the illicit drug heroin and the prescription drug codeine are metabolized to morphine, which tends to complicate interpretation of opiate-positive samples. We report here the concentrations of morphine and codeine, the morphine/codeine ratios, and 6-acetylmorphine (6-AM) in blood specimens from individuals arrested for driving under the influence of drugs (DUID) in Sweden. We describe a new derivatization method for phenmetrazine using 4-carbethoxyhexafluorobutyryl chloride. The applicability of the immunoassay TRIAGE (E. Merck, Darmstadt, Germany) as a simple and rapid assay for antemortem drug abuse was tested on 100 urine samples from forensic autopsies. Residual samples of blood spots, which are routinely collected on almost all newborns in the United States, can be used to determine seroprevalence information on newborns and maternal exposures to various substances, including drugs of abuse. Automating the initial screening of postmortem blood and other tissues for drugs of abuse requires a pipetting procedure that is compatible with immunoassay screening tests. The deproteinization procedure uses a zinc sulfate-5-sulfosalicylic acid reagent to precipitate blood proteins. This procedure for deproteinization of whole blood and tissue homogenates is compatible with coated tube radioimmunoassays (RIA) and can therefore be used in automating the initial screening step.
