Identification and prediction of Parkinson's disease subtypes and progression using machine learning in two cohorts.

The clinical manifestations of Parkinson's disease (PD) are characterized by heterogeneity in age at onset, disease duration, rate of progression, and the constellation of motor versus non-motor features. There is an unmet need for the characterization of distinct disease subtypes as well as improved, individualized predictions of the disease course. We used unsupervised and supervised machine learning methods on comprehensive, longitudinal clinical data from the Parkinson's Disease Progression Marker Initiative (n = 294 cases) to identify patient subtypes and to predict disease progression. T…

HYPER-Net: Physics-Conditioned Self-Supervised Reconstruction for Fourier Light-Field Microscopy.

The rapid convergence of optical innovation and machine intelligence is reshaping biological imaging by enabling platforms that jointly advance image formation and computational reconstruction for highspeed, high-resolution volumetric microscopy. However, broadly accessible three-dimensional imaging at high spatiotemporal resolution remains limited by the reliance of existing supervised methods on large modality-matched training datasets, the computational burden of conventional iterative reconstruction, and sensitivity to optical mismatch arising from small deviations in the spatial-angular p

Exploring Radiomics Features Based on H&E Images as Potential Biomarkers for Evaluating Muscle Atrophy: A Preliminary Study.

Radiomics features have been widely used as novel biomarkers in the diagnosis of various diseases, but whether radiomics features derived from hematoxylin and eosin (H&E) images can evaluate muscle atrophy has not been studied. Therefore, this study aims to establish a new biomarker based on H&E images using radiomics methods to quantitatively analyze H&E images, which is crucial for improving the accuracy of muscle atrophy assessment. Firstly, a weightless muscle atrophy model was established by laying macaques in bed, and H&E images of the shank muscle fibers of the control and bed rest (BR)

Optimization in Sparse 2D to Dense 3D Weakly Supervised Learning: Application to Multi-Label Segmentation of Large ex vivo MRI Data.

INTRODUCTION —: The fully supervised segmentation of 3D medical images featuring large amounts of slices (1,000+), such as serial block-face electron microscopy, or high-resolution ex vivo MRI, imposes prohibitive cost of volumetric manual annotation. Oftentimes, researchers resort to limited ground truth annotations consisting of sparse 2D slices, forcing a reliance on models that lack volumetric context. Incorporating across-slice information in the model can be done using a weakly supervised Sparse-to-Dense framework. However, guidelines for training such models remain ambiguous, specifical

Computational Mass Spectrometry Imaging in the Era of AI.

Mass spectrometry imaging (MSI) is a tool-of-choice for mapping and understanding the spatial organization of biomolecules, including small metabolites, lipids, peptides, and many others. As the MSI instrument and spatial biology inquiries evolve, researchers and practitioners are constrained by the inherent trade-offs in spatial resolution, chemical detail, and acquisition time. Here, we review how the rapidly growing interplay between MSI and machine learning/artificial intelligence (ML/AI)-powered computational approaches is addressing these issues. We begin by highlighting key steps in MSI

Pain Recognition With Electrocardiographic Features in Postoperative Patients: Method Validation Study.

BACKGROUND: There is a strong demand for an accurate and objective means of assessing acute pain among hospitalized patients to help clinicians provide pain medications at a proper dosage and in a timely manner. Heart rate variability (HRV) comprises changes in the time intervals between consecutive heartbeats, which can be measured through acquisition and interpretation of electrocardiography (ECG) captured from bedside monitors or wearable devices. As increased sympathetic activity affects the HRV, an index of autonomic regulation of heart rate, ultra-short-term HRV analysis can provide a re

Harnessing vaginal inflammation and microbiome: a machine learning model for predicting IVF success.

Humans are the only species with a commensal Lactobacillus-dominant vaginal microbiota. Reproductive tract microbes have been linked to fertility outcomes, as has intrauterine inflammation, suggesting immune response may mediate adverse outcomes. In this pilot study, we compared vaginal microbiota composition and immune marker concentrations between patients with unexplained or male factor infertility (MFI), as a control. We applied a supervised machine learning algorithm that integrated microbiome and inflammation data to predict pregnancy outcomes.Twenty-eight participants provided vaginal s…

Federated CT foundation models for multi-center detection of lymph node metastasis in pancreatic cancer.

Pancreatic ductal adenocarcinoma (PDAC) remains one of the most lethal malignancies, with prognosis strongly influenced by the presence of lymph node metastasis (LNM). However, preoperative LNM assessment from computed tomography (CT) is limited by low sensitivity, high inter-observer variability, and substantial heterogeneity across imaging protocols. This retrospective multi-center study (546 patients from three institutions) introduces a privacy-preserving deep learning framework that integrates large-scale CT foundation model pre-training with heterogeneity-aware federated optimization to

Optimizing preoperative planning for total hip arthroplasty using random forest models to predict stem size and compatibility.

BACKGROUND: Preoperative planning is essential for optimal outcomes post-total hip arthroplasty (THA). Appropriate stem size and compatibility with femoral dimensions are essential in THA to prevent distal fixation. Previous studies have proposed deep learning-based models with image recognition for preoperative planning to determine the best-fit stem; however, whether size and compatibility are adequately optimized remains unclear. This study developed methods that may assist surgeons in preoperative planning to estimate the stem size and compatibility using supervised machine learning models…
