Dermatitis Herpetiformis (DH) is an immunobullous skin disease with an associated gluten-sensitive enteropathy. Withdrawal of gluten from the diet leads to the resolution of both the skin lesions and the enteropathy. We have previously reported that Lyt2+ cytotoxic T lymphocytes (CTL) can be raised against Japanese encephalitis virus (JEV) in BALB/c mice. We used a rat model of orthotopic penetrating keratoplasty to study splenic cell cytotoxicity in the host. DA (RT1avl) rats received grafts of Fischer (F344, RT1lv1) rat corneas. Cytotoxic T lymphocyte (CTL) activity was measured by the cell-mediated lymphocytotoxicity assay using 4/4R.M.-4 cells as target cells. CTL activity became detectable in vitro 2 weeks after grafting and peaked at 3 weeks. This simple method of quantifying CTL activity in the orthotopic penetrating keratoplasty model should provide a reliable tool for studying the CTL response in corneal transplantation.

The importance of cytosolic free calcium level ([Ca2+]i) in lymphocyte activation prompted us to investigate changes in [Ca2+]i in T cells caused by mercury compounds, which have been shown to have immunomodulatory and immunotoxic properties. Using fura-2 as fluorescent Ca2+ indicator, we found that both methyl-mercury (MeHg; 0.02-2 microM) and inorganic mercury (HgCl2; 0.01-1 microM) increased [Ca2+]i in lymphocytes from rat spleen in a concentration-dependent manner. Polyribosilribitolphosphate (PRP), the capsular polysaccharide from Haemophilus influenzae type b, is a T-cell-independent type 2 antigen.
