Innovations in preclinical biology: ex vivo engineering of a human kidney tissue microperfusion system.

Kidney disease is a public health problem that affects more than 20 million people in the US adult population, yet little is understood about the impact of kidney disease on drug disposition. Consequently there is a critical need to be able to model the human kidney and other organ systems, to improve our understanding of drug efficacy, safety, and toxicity, especially during drug development. The kidneys in general, and the proximal tubule specifically, play a central role in the elimination of xenobiotics. With recent advances in molecular investigation, considerable information has been gat

Polyamine delivery as a tool to modulate stem cell differentiation in skeletal tissue engineering.

The first step in skeleton development is the condensation of mesenchymal precursors followed by any of two different types of ossification, depending on the type of bone segment: in intramembranous ossification, the bone is deposed directly in the mesenchymal anlagen, whereas in endochondral ossification, the bone is deposed onto a template of cartilage that is subsequently substituted by bone. Polyamines and polyamine-related enzymes have been implicated in bone development as global regulators of the transcriptional and translational activity of stem cells and pivotal transcription factors.

Tissue engineering with recombinant human platelet-derived growth factor BB for implant site development.

Currently, PDGF-BB is FDA-approved for periodontal regeneration as part of a dental bone-filling device only. Although this device uses beta-TCP as the scaffold carrier, there has been considerable clinical interest in combining this growth factor with other bone replacement grafts, particularly bone allografts. This article reports on clinical experiences using rhPDGF-BB with bone allografts for implant site development. After careful evaluation of clinical parameters and consideration of current and emerging evidence, the off-label use of rhPDGF-BB was determined in the following case report

Engineering bioartificial tracheal tissue using hybrid fibroblast-mesenchymal stem cell cultures in collagen hydrogels.

We aimed at providing the first in vitro and in vivo proof-of-concept for a novel tracheal tissue engineering technology. We hypothesized that bioartificial trachea (BT) could be generated from fibroblast and collagen hydrogels, mechanically supported by osteogenically-induced mesenchymal stem cells (MSC) in ring-shaped 3D-hydrogel cultures, and applied in an experimental model of rat trachea injury. Tube-shaped tissue was constructed from mixtures of rat fibroblasts and collagen in custom-made casting molds. The tissue was characterized histologically and mechanically. Ring-shaped tissue was …

Fabrication of cultured oral gingiva by tissue engineering techniques without materials of animal origin.

BACKGROUND: Cultured gingival substitute has been found to be a useful graft material for treatment of gingival recession. However, such substitutes include xenograft derivative materials that involve concomitant risk of viral contamination. To eliminate this risk, we designed new gingival substitutes made of recombinant human collagen types I and III sponges and cultured these substitutes in animal-free media (HFDM-1). METHODS: Gingival fibroblasts were seeded onto sponges of type I or III recombinant collagen. These sponges were cultured in Dulbecco's modified Eagle's medium (DMEM) with 10%

Auricular cartilage tissue engineering: from making cartilage to regenerating a shape-stable elastic organ.

Auricular cartilage tissue engineering has long been grouped under the broader category of craniofacial cartilage reconstruction, but this classification is becoming insufficient. Unlike articular cartilage repair, which mainly aims to restore load-bearing function and relieve pain, auricular reconstruction must reproduce a complex three-dimensional structure, preserve elastic recoil, resist long-term contraction, tolerate soft-tissue coverage, and remain surgically and aesthetically acceptable over time. These demands make auricular cartilage engineering fundamentally distinct from generic ca…

Graphene-Based Nanomaterials as Bioactive Agents and Drug Delivery Systems for Bone Tissue Engineering.

Graphene-based nanomaterials (GBNs), such as graphene oxide and reduced graphene oxide, have been considered as promising functional materials for bone tissue engineering due largely to their intrinsic biological functions and great drug delivery capacity. As bioactive materials, GBNs possess antibacterial activity, photothermal effect, and the ability to regulate immune microenvironment and osteogenesis-related signaling pathways such as Wnt/β-catenin, MAPK, and BMP/Smad. As drug delivery systems, the large surface area and multiple functionalization sites allow GBNs to have great potential i…

Bone Tissue Engineering Strategies To Treat Critically Sized Defects in Compromised Wound Healing Environments.

Critically sized bone defects are difficult to treat, necessitating tissue engineering strategies to restore form and function. However, translation of these approaches is often constrained by preclinical models that fail to replicate systemic comorbidities commonly seen in clinical practice, such as diabetes, prior irradiation, osteonecrosis, and osteoporosis, and instead favor healthy wound environments that may overestimate efficacy. This comprehensive review aimed to provide a detailed overview of in vivo bone regeneration strategies for critically sized defects specifically within comprom…

Photoactivatable CRISPR/Cas13d via upconversion nanoparticles for deep tissue RNA engineering and orthopedic therapy.

Spatiotemporal control of RNA therapeutics remains a fundamental challenge limiting clinical translation. Here, we develop a photoactivatable CRISPR/Cas13d (paCas13d) system that enables non-invasive, light-controlled RNA manipulation in deep tissues. Through structure-guided engineering, we identify optimal split sites within RfxCas13d and create light-switchable fragments using CRY2PHR/CIBN optogenetic dimerization. To overcome the limited tissue penetration of blue light, we engineer polyethylenimine-functionalized upconversion nanoparticles (UCNPs-PEI) that serve dual roles as gene carrier

Engineering epicardium-integrated human iPSC-derived heart tissue for modelling Hutchinson-Gilford progeria syndrome.

Engineered heart tissues (EHTs) have been used in cardiovascular research for nearly three decades, addressing limitations of animal models and oversimplified two-dimensional single-cell systems in recapitulating cardiac (patho) physiology. Recently, epicardium-integrated EHTs have gained attention as therapeutic tools for remuscularizing the failing heart. The epicardium, the outermost mesothelial layer of the heart, plays a crucial role in cardiac development and disease by serving as a source of progenitor cells and by providing essential paracrine signalling. Hutchinson-Gilford progeria sy

Collagen-based nanomaterials for tissue engineering: A review from nanocollagen to collagen-based nanocomposites.

Collagen is widely used as a scaffold material and drug delivery vehicle in tissue engineering due to its excellent biocompatibility and low immunogenicity. However, conventional collagen-based biomaterials suffer from inadequate mechanical strength, poor thermal stability, and excessively rapid degradation. Nanotechnology provides effective strategies to overcome these limitations. Collagen-based nanomaterials generally exhibit significantly enhanced mechanical properties and thermal stability, prolonged drug half-life, and optimized drug release behavior, showing great promise for repairing …

Refractive Index Engineering for Human Corneal Tissue by Femtosecond Laser Two-Photon Cross-Linking.

Clinical refractive surgeries for myopia correction generally entail stromal tissue removal with risks of postoperative complications. NIR femtosecond laser-induced corneal two-photon collagen cross-linking (two-photon CXL) through photochemical effects can lead to localized refractive index (RI) changes in the cornea, promising for non-ablative and non-incisional visual correction. However, previous studies mainly focused on hydrogels and animal corneal tissues, adopting a diffraction grating method that is difficult for biological tissue. Here we systematically investigate femtosecond laser-
