The syntheses of trans-3-(4-oxo-4H-1-benzypyran-3)acrylic acid and a number of analogs shown to be highly active in antiallergic bioassays are described. These compounds are of possible value in the treatment of asthma. The structural requirements for biological activity are discussed with reference to the type of the substituents on the chromone ring or positions of linkage of the acrylic acid on the pyrone ring.

BACKGROUND: Transcription cofactors related to Drosophila Schnurri facilitate the transcriptional programs regulated by BMP signaling in C. elegans, Drosophila, Xenopus, and mouse. In different systems, Schnurri homologs have been shown to act as either agonists or antagonists of Smad function, and as either positive or negative regulators of transcription. How Schnurri proteins achieve this diversity of activities is not clear. The C. elegans sma-9/schnurri locus undergoes alternative splicing, including an unusual trans-splicing event that could generate two non-overlapping shorter transcrip

Aromatic amine antioxidants (AAs), as rubber additives, and their ozone photochemical oxidation products of p-phenylenediamine quinone (PPD-Qs) have attracted great attention recently due to their wide environmental occurrences and toxicity. However, there is currently no research on the exposure risks during pregnancy and their trans-placental transfer mechanisms. Herein, 20 AAs and six PPD-Qs were analyzed in 60 maternal urine and fifty-six amniotic fluid samples (n = 53 pairs). ΣAAs (median: 8.57 and 15.4 ng/mL) and ΣPPD-Qs (0.236 and 2.29 ng/mL) were both observed, where the median concent

The catalytic dechlorination of chlorinated ethylenes by 5,10,15,20-tetrakis(4-carboxyphenyl)porphyrin cobalt ((TCPP)Co), a cobalt complex structurally similar to vitamin B12, was studied. It was found to have superior aqueous-phase dechlorination activity on chlorinated ethylenes (CEs) relative to vitamin B12. Bimolecular rate constants for the degradation of CEs by (TCPP)Co of 250, 24, 0.24, and 1.5 M(-1) s(-1) were found for perchloroethylene (PCE), trichloroethylene (TCE), cis-dichloroethylene (cDCE), and trans-dichloroethylene (tDCE), respectively. Through kinetic analysis, the rate laws

BACKGROUND: Anterior cruciate ligament (ACL) tears in skeletally immature patients are increasingly common. Evidence comparing the outcomes of all-epiphyseal versus trans-epiphyseal ACL reconstruction in skeletally immature patients is limited, and the current literature could benefit from a comprehensive systematic review. The present study compared all-epiphyseal versus trans-epiphyseal ACL reconstruction in skeletally immature patients. The outcomes of interest were to compare joint laxity, patient-reported outcome measures (PROMs), return to sport, and complications.
METHODS: This study w

Cytochrome P450 1A1 (CYP1A1) activity is associated with increased susceptibility to lung cancer induced by polycyclic aromatic hydrocarbons such as benzo[a]pyrene (BP). In non-hepatic human tissues, CYP1A1 is the principal enzyme responsible for the metabolic activation of the proximate BP mutagenic metabolite, (-)-benzo[a]pyrene-trans-7,8-dihydrodiol, to (+)-anti-benzo[a]pyrene-trans-7,8-dihydrodiol-9,10-epoxide, the ultimate BP mutagen. We have genetically engineered both DNA repair-deficient (xeroderma pigmentosum group A) and DNA repair-proficient human skin fibroblasts to express human C

Production of granulocyte-macrophage colony-stimulating factor (GM-CSF) by normal T lymphocytes requires activation by antigen, mitogen or lectin, whereas T-cell lines transformed by human T-cell leukemia virus type I (HTLV-I) or type II (HTLV-II) constitutively produce high levels of GM-CSF. Using transient cotransfection assays, we demonstrate that introduction of the tax gene of either HTLV-I or HTLV-II is sufficient to activate GM-CSF promoter constructs in an unstimulated T-cell line. The GM-CSF 5' flanking sequences previously shown to be sufficient for GM-CSF induction following T-cell

Directing groups play an important role in controlling the selectivity of C-H bond activation. Here we demonstrate that for iron, the nature of the directing group (e.g., ketone, ester, or amide) influences the C-H activation process. In this study the C-H bond activation step either occurs with or without the assistance of the directing group resulting in distinct cis- and trans-isomers of the corresponding iron hydride.

Plant organ size depends on the coordination of cell proliferation and expansion, a process regulated by numerous factors, including transcription factors and small RNAs. However, how different small-RNA pathways are integrated with growth-regulating transcriptional networks remains only partly understood. Here, we studied how the Arabidopsis TRANS-ACTING siRNA3A (TAS3A)-derived trans-acting small RNA pathway controls organ size in Arabidopsis thaliana. We found that TAS3A overexpression increased leaf and floral organ size, whereas tas3a mutants showed reduced organ growth. We show that TAS3A

Routing chemically distinct small-molecule aptamers into a common nuclease trans-cleavage pathway is constrained by the target-specific output handles of conventional structure-switching aptamers (SSAs). Shared-output aptamer-gated DNA nanodevices are designed so that recognition cores control release of the same short trigger DNA (tDNA). The released tDNA hybridizes with a common guide RNA and activates NbaSPARDA, a short prokaryotic Argonaute-associated nuclease complex, for reporter trans-cleavage. Analyses across two sequence scaffolds reveal sequence-dependent activation and identify a pr

Celiac disease (CeD) is a complex T cell-mediated enteropathy induced by gluten. Although genome-wide association studies have identified numerous genomic regions associated with CeD, it is difficult to accurately pinpoint which genes in these loci are most likely to cause CeD. We used four different in silico approaches-Mendelian randomization inverse variance weighting, COLOC, LD overlap, and DEPICT-to integrate information gathered from a large transcriptomics dataset. This identified 118 prioritized genes across 50 CeD-associated regions. Co-expression and pathway analysis of these genes i

Transcription of the Bacillus anthracis structural genes for the anthrax toxin proteins and biosynthetic operon for capsule is positively regulated by AtxA, a transcription regulator with unique properties. Consistent with the role of atxA in virulence factor expression, a B. anthracis atxA-null mutant is avirulent in a murine model for anthrax. In culture, multiple signals impact atxA transcript levels, and the timing and steady-state level of atxA expression are critical for optimal toxin and capsule synthesis. Despite the apparent complex control of atxA transcription, only one trans-acting

Pulmonary infections are characterized by neutrophil recruitment into the lung driven by chemokine ligands of CXCR2, which is expressed on neutrophils, but also present in non-hematopoietic lung cells, in which its role remains unclear. We hypothesize that CXCR2 in epithelial and endothelial cells contributes to neutrophil recruitment into the lung by modifying the availability of its cognate chemokines in lung alveoli. Using conditional endothelial and epithelial CXCR2 knockout mice, we demonstrate that selective CXCR2 deletion in either compartment impairs neutrophil recruitment into the lun

Peptidyl-prolyl cis/trans isomerases (PPIases) are present in a wide variety of microorganisms, including protozoan parasites such as Trypanosoma cruzi, Trypanosoma brucei, Trichomonas vaginalis, Leishmania major, Leishmania donovani, Plasmodium falciparum, Plasmodium vivax, Entamoeba histolytica, Giardia intestinalis, Cryptosporidium parvum, and Cryptosporidium hominis, all of which cause important neglected diseases. PPIases are classified as cyclophilins, FKBPs, or parvulins and play crucial roles in catalyzing the cis-trans isomerization of the peptide bond preceding a proline residue. Thi

Diphtheria toxin kills human cells because it delivers its enzyme domain DTA into their cytosol where it inhibits protein synthesis. After receptor-mediated uptake of the toxin, DTA translocates from acidic endosomes into the cytosol, which might be assisted by host cell factors. Here we investigated the role of Hsp90 and its co-chaperones during the uptake of native diphtheria toxin into human cells and identified the components of the Hsp90 machinery including Hsp90, Hsp70, Cyp40 and the FK506 binding proteins FKBP51 and FKBP52 as DTA binding partners. Moreover, pharmacological inhibition of
