Two Aquaporins Mitigate Growth-Defence Trade-Offs by Facilitating CO2 and H2O2 Transport in Wheat.

The growth-defence trade-offs pose a major challenge to breeding high-yield and disease-resistant crops. Aquaporins are membrane channels that facilitate the transport of water and other small compounds, therefore regulating the growth-defence trade-offs. However, the molecular mechanism that governs the function of aquaporins in the trade-offs remains unclear. Here, we report that Triticum aestivum TaPIP1;6 and TaPIP2;10, aquaporins of the plasma membrane intrinsic protein (PIP) family, function as dual substrate channels that concurrently enhance plant growth and resistance to powdery mildew…

Biological control of Botrytis cinerea in tomato and Fusarium graminearum in wheat using the novel species Burkholderia mycopellens and Burkholderia crassaminum.

The use of beneficial microbes to reduce plant stress upon fungal pathogens is a promising strategy for sustainable crop protection. In this study, we evaluated five novel Burkholderia strains for their capacity to mitigate Botrytis cinerea infection in tomato (Solanum lycopersicum L.) and Fusarium graminearum infection in wheat (Triticum aestivum L.). All strains significantly suppressed B. cinerea infection symptoms in tomato plants, hallmarked by a smaller decrease in photosynthetic activity and chlorophyll content compared to infected control. Using a GFP-tagged F. graminearum strain, we s…

Biocontrol of Fusarium culmorum and enhanced wheat seedling growth by Streptomyces sp. D15 from Algerian steppe soil: performance and genome analysis.

Wheat (Triticum aestivum L.), a staple crop in Algeria and other semi-arid regions, faces significant challenges from fungal pathogens like Fusarium culmorum, which cause substantial yield losses and threaten food security. This study investigates the biocontrol and plant growth-promoting potential of Streptomyces sp. D15, an actinobacterial strain isolated from steppe soil in Algeria. Phylogenomic analysis confirmed the classification of strain D15 as Streptomyces lavendofoliae. During in planta experiments on wheat seedlings, strain D15 improved shoot length with increases of 67% relative to…

Comprehensive Analysis of IDD Transcription Factors and Their Expression Profiling Under Pathogen Stress in Wheat.

INDETERMINATE DOMAIN (IDD) transcription factors are plant-specific regulators essential for plant development and stress adaptation. As a globally important staple crop, common wheat (Triticum aestivum L.) is frequently threatened by fungal diseases such as powdery mildew and stripe rust. To date, however, the IDD gene family in wheat has not been systematically characterized, and its roles in biotic stress responses remain unclear. In this study, we performed genome-wide identification and a comprehensive analysis of the TaIDD gene family. A total of 41 TaIDD genes were identified, which wer…

A slow-then-rapid spike development trajectory enables wheat to escape late-spring cold spells via transcriptional and epigenetic reprogramming.

Late-spring cold spells (LSCs) threaten bread wheat (Triticum aestivum L.) when cold-sensitive spike development coincides with ephemeral spring chilling. The leading Chinese cultivar Xinong 511 (XN511) exhibits superior LSC tolerance, yet the underlying developmental and molecular basis remains unclear. Phenotypic monitoring across eight sequential sampling points, designated T1-T8, revealed that XN511 displays a distinctive "slow-then-rapid" developmental trajectory relative to its parents, Xinong 2000 (XN2000) and Shaanmai 159 (SM159): an extended early vegetative phase followed by accelera…

Functional Characterization of Wheat Seed Endophytic Bacteria Reveals Plant Growth-Promoting Traits and Potential Biocontrol Activity Against Fusarium Pathogens.

Seed-associated microbiota are emerging as key determinants of early plant establishment and resilience, yet their functional potential in wheat remains underexplored. Here, we isolated and functionally characterized culturable bacterial endophytes from seeds of Triticum aestivum cvs. Saekeumkang, Baeggang, and Ariheuk to assess plant growth promotion and pathogen suppression. Sixteen bacterial strains, belonging to Pseudomonadota, Actinomycetota, and Bacillota, were identified using 16S rRNA gene sequencing. Functional assays revealed the high prevalence of plant growth-promoting traits, with…

Development and validation of KASP markers for the greenbug resistance gene Gb7 and the Hessian fly resistance gene H32 in wheat.

Greenbug and Hessian fly are important pests that decrease wheat production worldwide. We developed and validated breeder-friendly KASP markers for marker-assisted breeding to increase selection efficiency. Greenbug (Schizaphis graminum Rondani) and Hessian fly [Mayetiola destructor (Say)] are two major destructive insect pests of wheat (Triticum aestivum L.) throughout wheat production regions in the USA and worldwide. Greenbug and Hessian fly infestation can significantly reduce grain yield and quality. Breeding for resistance to these two pests using marker-assisted selection (MAS) is the m…

Biosolids affect soil barium in a dryland wheat agroecosystem.

In December 2003, the USEPA released an amended list of 15 "candidate pollutants for exposure and hazard screening" with regard to biosolids land application, including Ba. Therefore, we decided to monitor soil Ba concentrations from a dryland wheat (Triticum aestivum L.)-fallow agroecosystem experiment. This experiment received 10 biennial biosolids applications (1982-2003) at rates from 0 to 26.8 dry Mg ha(-1) per application year. The study was conducted on a Platner loam (Aridic Paleustoll), approximately 30 km east of Brighton, CO. Total soil Ba, as measured by 4 M HNO(3), increased with …

Characterization of the binding of the TC-83 strain of Venezuelan Equine encephalomyelitis virus to BW-J-M, a mouse macrophage-like cell line.

The first step in virus replication is attachment of the virus to the host cell surface. To investigate this process, the binding of TC-83 ( … inhibit TC-83 binding to BW-J-M cells. Canavalia ensiformis agglutinin, Glycine max agglutinin and Triticum vulgaris agglutinin were potent inhibitors of virus binding. This evidence suggests that TC-83 binds to a specific receptor on the BW-J-M cell and that the receptor may be glycoprotein.

Genome-wide identification of wheat USP gene family and functional dissection of TaUSP85 involved in heat tolerance.

A collection of conserved proteins known as universal stress protein (USP) is present in a wide range of species, including plants, fungi, bacteria, and animals. USPs are named for their ability to respond to a variety of stress conditions, such as heat stroke, osmotic stress, nutrient limitation, and exposure to toxins or antibiotics. While the USP response to different stress conditions in plants has been reported, little is known about the USP family in wheat (Triticum aestivum L.). In wheat, we identified 88 USP genes distributed across 21 chromosomes classified into four subfamilies. Phyl…

Mapping of a novel major effect Hessian fly field partial-resistance locus in southern soft red winter wheat line LA03136E71.

Hessian fly resistance has centralized around resistance loci that are biotype specific. We show that field resistance is evident and controlled by a single locus on chromosome 7D. Hessian flies (Mayetiola destructor Say) infest and feed upon wheat (Triticum aestivum L) resulting in significant yield loss. Genetically resistant cultivars are the most effective method of Hessian fly management. Wheat breeders in the southern USA have observed cultivars exhibiting a "field resistance" to Hessian fly that is not detectable by greenhouse assay. The resistant breeding line "LA03136E71" and suscepti…

Virus-induced gene silencing suggests (1,3;1,4)-β-glucanase is a susceptibility factor in the compatible russian wheat aphid-wheat interaction.

The Russian wheat aphid (RWA), Diuraphis noxia (Kurdjumov), is a significant insect pest of wheat (Triticum aestivum L.) and has a major economic impact worldwide, especially on winter wheat in the western United States. The continuing emergence of new RWA biotypes virulent to existing resistance genes reinforces the need for more durable resistance. Studies have indicated that resistance in previously susceptible plants can be produced by knock-down of susceptibility genes or other genes involved in host plant susceptibility. Therefore, investigation into genes involved in compatible RWA-whea…

Transcriptional-metabolic networks in beta-carotene-enriched potato tubers: the long and winding road to the Golden phenotype.

Vitamin A deficiency is a public health problem in a large number of countries. Biofortification of major staple crops (wheat [Triticum aestivum], rice [Oryza sativa], maize [Zea mays], and potato [Solanum tuberosum]) with β-carotene has the potential to alleviate this nutritional problem. Previously, we engineered transgenic "Golden" potato tubers overexpressing three bacterial genes for β-carotene synthesis (CrtB, CrtI, and CrtY, encoding phytoene synthase, phytoene desaturase, and lycopene β-cyclase, respectively) and accumulating the highest amount of β-carotene in the four aforementioned …

[Cloning and transcriptional profiling of TaZIP7 gene in wheat with different zinc concentrations].

TaZIP7 belongs to a family of zinc finger proteins in wheat. Given that the TaZIP7 gene is highly associated with zinc content in wheat grains, this study selected nine spring wheat cultivars with significantly divergent grain zinc contents to clone and analyze the full-length sequences (including the promoter region and 3' untranslated region) and coding sequences (CDS) of the TaZIP7 gene, and further investigated the transcript expression characteristics of this gene in mature wheat grains, thus providing valuable genetic resources and theoretical support for the genetic improvement of zinc n

The hijacking of a wheat Exo70-like protein by a fungal pathogen triggers disease.

Tan spot is an economically important fungal disease of wheat that occurs globally. In this necrotrophic pathosystem, pathogen effectors are recognized by wheat genes in an inverse gene-for-gene manner, leading to the hijacking of host defense mechanisms and ultimately disease susceptibility. Here, we identified the tan spot susceptibility gene Tsc2 through positional cloning, mutagenesis, and transgenic complementation. Tsc2 encodes a diverged form of the exocyst subunit Exo70FX15 that localizes to the nucleus and cytoplasm and does not interact directly with the fungal effector. A 21‑nucleot
