Astacin metalloproteases in human-parasitic nematodes.

Parasitic nematodes infect over 2 billion individuals worldwide, primarily in low-resource areas, and are responsible for several chronic an … the whipworm Trichuris trichiura - as well as the human-infective, vector-borne filarial nematodes Wuchereria bancrofti, Onchocerca volvulus, and Brugia malayi. We also review astacin function in parasitic nematodes that infect other mammalian hosts and discuss the potential of astacins as anthelmintic drug targets. Finally, we highlight the molecular and g …

Lymphedema resulting from filariasis successfully treated by surgery.

Filariasis is an infectious disease caused by a filarial worm infection transmitted by mosquito bites. The disease can result in reduced wor … ues were present on the right lower extremity. The blood microfilariae examination was positive for Wuchereria bancrofti. The lymphedema did not resolve despite of antifilarial treatment and surgery was eventually performed to resect the fibrous tissue and subcutaneous edema. The patient responded well to the treatment with a significant reduction in the ede …

Is there a risk of filarial infection during long-term missions in Haiti?

BACKGROUND: Haiti has the highest prevalence of lymphatic filariasis (Wuchereria bancrofti) in the Western Hemisphere. Still, the risk of filarial infection for long-term visitors such as humanitarian aid workers or military personnel is uncertain. The presented study analyzed the exposure to W. bancrofti in Chilean participants of the UN Stabilization Mission in Haiti (MINUSTAH) in 2011. METHODS: Blood samples collected from 531 participants were screened for antifilarial antibodies by IgG ELISA, and, if positive, analyzed by immunofluorescence assay (IFA), IgG4 ELISA, Real-Time PCR, and cir…

Role of vector control in the global program to eliminate lymphatic filariasis.

Lymphatic filariasis (LF) is a major cause of acute and chronic morbidity in the tropical and subtropical parts of the world. The availability of safe, single-dose, drug treatment regimens capable of suppressing microfilaremia to very low levels, along with improvements in techniques for diagnosing infection, has resulted in the targeting of this major mosquito-borne disease for global elimination. The Global Program to Eliminate Lymphatic Filariasis (GPELF) was launched in 2000 with the principal objective of breaking the cycles of transmission of Wuchereria bancrofti and Brugia spp. through …

Production, purification and diagnostic application of filarial recombinant protein WbSXP-1 expressed in salt inducible Escherichia coli.

Wuchereria bancrofti protein WbSXP-1 was identified and established as a potential candidate for the diagnosis of lymphatic filariasis. For the economic production of rWbSXP-1, osmotically (salt) inducible Escherichia coli GJ1158 was preferred. Cultivation and expression was optimized in 3 L airlift bioreactor (ALB) and was successfully extended to 30 L ALB. Purification of rWbSXP-1 his-tag protein was optimized in technical scale using FPLC and the maximal recovery of rWbSXP-1 with significant level of purity was achieved using the combination of IMAC and gel filtration. Quality criteria for …

Biochemical composition and metabolic pathways of filarial worms Setaria cervi: search for new antifilarial agents.

The main problem regarding the chemotherapy of filariasis is that no safe and effective drug is available yet to combat the adult human filarial worms. Setaria cervi, the causal organism of setariasis and lumbar paralysis in cattle, is routinely employed as a model organism for conducting biochemical and enzymatic studies on filarial parasites. In view of the practical difficulties in procuring human strains of Wuchereria bancrofti and Brugia malayi for drug screening, the bovine filarial parasite S. cervi, resembling the human species in having microfilarial periodicity and chemotherapeutic r…

Control of lymphatic filariasis in a hunter-gatherer group in Madang Province.

Diethylcarbamazine (DEC) has been successfully administered to millions of people in established villages and towns, but little or no information exists on the use of this drug to control lymphatic filariasis in isolated seminomadic groups. We have studied the impact of biannual single-dose mass treatment to control filariasis in the Hagahai, an isolated hunter-gatherer, shifting horticulturist group in the fringe highlands of Papua New Guinea. Despite low treatment coverage, 6 mass treatment rounds significantly reduced the overall prevalence of infection with Wuchereria bancrofti, by antigen…

[Combinations of ivermectin and diethylcarbamazine for improved control of lymphatic filariasis].

Since 1993, a three arm double-blind controlled trial was implemented in French Polynesia to compare the efficacy of single doses of diethylcarbamazine (DEC) 6 mg/kg vs. ivermectin (IVR) 400 micrograms/kg vs. the combination IVR 400 + DEC 6 for treatment of Wuchereria bancrofti mf carriers. Of the 57 treated male patients, three groups of 19 were randomly selected and allocated to one of the three treatments. Twelve months after the first then the second treatment (M12 then M24) respectively, 3 (16%) then 10 (53%), 3 (16%) then 5 (26%), 7 (27%) then 10 (53%) of patients were mf negative in gro…

Identification and characterisation of a novel repetitive antigen from Onchocerca spp.

A novel repetitive antigen from the cattle parasite Onchocerca gibsoni was shown to be recognised by sera from humans infected with Onchocerca volvulus, Wuchereria bancroftii or Brugia malayi. The O. gibsoni protein was produced in a recombinant form, and antibodies raised to this protein used to screen cDNA libraries for O. volvulus. A series of clones were isolated which encoded repetitive regions very similar to those in O. gibsoni, but interspersed between these were longer repeating units which we have not so far found in O. gibsoni. The repetitive antigen was shown to be of high molecula…

Diethylcarbamazine in the control of splenomegaly associated with Bancroftian filariasis in the Ok Tedi area of Papua New Guinea.

Bancroftian filariasis is highly endemic in the Ok Tedi region of Papua New Guinea, with a reported mean rate of 39% before the implementati … ated counterparts. Significant differences were reported in the rate of detectable microfilariae of Wuchereria bancrofti, but not of malaria, between the two groups. The number of DEC administrations and the period of time since the first treatment played a significant role immunologically. Significant differences were observed in immunoglobulin (Ig) M and I …

Tropical pulmonary eosinophilia.

Tropical pulmonary eosinophilia is one of the many PIE syndromes [pulmonary infiltrates with eosinophilia (of the peripheral blood)]. It is caused by immunologic hyperresponsiveness to the filarial parasites Wuchereria bancrofti or Brugia malayi. Its clinical presentation includes nocturnal cough, dyspnea, wheezing, fever, weight loss, fatigue, interstitial mottling on chest radiograph, predominantly restrictive but also obstructive lung function abnormalities, and peripheral blood eosinophilia of more than 3000 per microliter. It can be distinguished from other PIE syndromes by the patient's …

Clinical and laboratory aspects of filariasis.

Human filarial infections afflict over 150 million persons worldwide and are major causes of morbidity in many developing countries. Onchocerca volvulus infection is a leading preventable cause of blindness, while bancroftian and brugian filariasis may produce lymphatic obstruction of the genitalia and extremities (elephantiasis). Definitive diagnosis of these helminthic infections currently depends on demonstration of microfilariae in host tissues, i.e., the skin in the case of O. volvulus and the bloodstream in the cases of Wuchereria bancrofti and Brugia malayi. Many investigations are now …

[Lymphatic filariasis in Haiti: historical sequel or future public health problem in this geographic region?].

Wuchereria bancrofti persists in Haiti in localized foci, discontinuously distributed. The parasite, transmitted by Culex quinquefasciatus, develops in mesoendemic or hyperendemic fashion in urban and suburban areas, on the coastal plains sheltered from the prevailing winds, predominantly in the Nord department and around the gulf of La Gonave. Lymphatic filariasis is a major public health problem. Its impact is more evident in males on account of the genital repercussion of the disease. The acute lesions appear from early infancy and are related to microfilarial density. The chronic lesions b…

Observations on the habits and control of Culex pipiens fatigans in Guyana.

Investigations in Guyana in 1961-63 showed that Wuchereria bancrofti was the only filarial parasite causing elephantiasis and that Culex pipiens fatigans was the primary vector. Mansonia titillans and Anopheles aquasalis were found to be secondary vectors.C. p. fatigans bred primarily in pit latrines and secondarily in the clean, confined water of drums and barrels near latrines. No breeding occurred in the relatively clean open water of trenches, ditches and drains.Of 21016 mosquitos collected from houses, 39.9% were found resting on walls, 34.9% on clothing and other hanging objects, and 23.…

The psychosocial and emotional burden of lymphatic filariasis: A systematic review.

BACKGROUND: Lymphatic filariasis (LF) is a neglected tropical disease affecting an estimated 882.5 million people at risk of infection. It is caused by the filarial nematodes Wuchereria bancrofti, Brugia malayi, and Brugia timori, leading to lymphedema, and severe deformation of extremities and resulting in both physical and mental health consequences. Affected individuals often suffer from depression, anxiety, and anger, exacerbated by disability, marginalization, and societal rejection due to their physical disability. METHODS: A systematic review of the literature was conducted following t…

Meta-analysis of the prevalence of lymphatic filariasis infection in mosquito vectors.

BACKGROUND: Lymphatic filariasis, caused by Wuchereria bancrofti, Brugia malayi, and Brugia timori, is transmitted by mosquitoes and persists as a major neglected tropical disease. Despite extensive elimination campaigns, information on vector infection prevalence remains fragmented, hindering evidence-based vector control strategies under the World Health Organization's Global Programme to Eliminate lymphatic filariasis. METHODS: A systematic review and meta-analysis was performed in accordance with PRISMA guidelines. Six databases (PubMed, Web of Science, ScienceDirect, CNKI, VIP, Wanfang) …

Operational research to inform post-validation surveillance of lymphatic filariasis in Tonga study protocol: History of lymphatic filariasis elimination, rational, objectives, and design.

BACKGROUND: Lymphatic filariasis (LF), a mosquito-borne helminth infection, is an important cause of chronic disability globally. The World Health Organization has validated eight Pacific Island countries as having eliminated lymphatic filariasis (LF) as a public health problem, but there are limited data to support an evidence-based approach to post-validation surveillance (PVS). Tonga was validated as having eliminated LF in 2017 but no surveillance has been conducted since 2015. This paper describes a protocol for an operational research project investigating different PVS methods in Tonga
