Acetylcholine-induced current fluctuations and fast excitatory post-synaptic currents in rabbit sympathetic neurones.

Post-synaptic currents and responses to ionophoretically applied acetylcholine (ACh) were recorded at 34-37 degrees C from rabbit superior cervical ganglion neurones clamped at -80 mV membrane potential. Atropine (1 microM) was used to block muscarinic receptors. The fast excitatory post-synaptic current (e.p.s.c.) reversed at -9.6 +/- 1.7 mV and decayed with a single exponential time course. The e.p.s.c. decay time constant, tau d, was 4.5 +/- 0.3 msec and increased as the membrane was hyperpolarized (e-fold increase in tau d corresponded to 140 mV hyperpolarization). Miniature e.p.s.c.s. (m.…

Interaction of cholinergic ligands with the purified acetylcholine receptor protein. I. Equilibrium binding studies.

We have studied the binding equilibria of two fluorescent ligands and several nonfluorescent cholinergic ligands with the purified acetylcholine receptor from Electrophorus electricus. The assay was based on the specifically cholinergic and reversible quenching of fluorescence observed upon complex formation between the receptor protein and N-7-(4-nitrobenzo-2-oxa-1,3-diazole)-5-aminopentanoic acid beta-(N-trimethylammonium) ethyl ester. This way, a large body of accurate, true equilibrium data was obtained. We find 1) all ligands studied compete for the same number of binding sites at the rec…

Notexin preferentially inhibits the release of newly synthesized acetylcholine from rat brain synaptosomal fractions.

An investigation was made of the effects of the snake venom neurotoxin, notexin, on acetylcholine turnover in rat brain P2 fractions using a gas chromatographic mass spectrometric assay for acetylcholine and choline. In contrast to earlier reports, we found a stimulation of the uptake and acetylation of labeled choline by toxin-treated P2 fractions. More significantly, notexin inhibited the release of this newly synthesized transmitter. These effects were found to be dependent on the dose of the toxin and the time of exposure of the P2 fraction to notexin. Longer exposure to notexin or experim…

A Single-Vesicle Electroanalytical Approach Uncovers Deficits of Vesicular Acetylcholine Content in Alzheimer's Disease.

The cholinergic system is essential for cognitive functions including learning, memory, and attention, with its dysfunction serving as a hallmark of neurodegenerative diseases including Alzheimer's disease (AD). However, how acetylcholine (ACh) is stored within individual synaptic vesicles, an essential compartment of cholinergic transmission, remains poorly understood, limiting insight into the nanoscale mechanisms underlying cognitive decline. Here, we present a high-throughput analytical strategy, molecular-empowered vesicle cytometry (MEVC), which employs calix[4]arene-facilitated ion tran…

Spontaneous and stimulus-driven arousal produce distinct acetylcholine dynamics across sensory and prefrontal cortex.

Acetylcholine (ACh) release from the basal forebrain has traditionally been viewed as a slow, spatially diffuse signal regulating cortical arousal across sleep and wakefulness1-3. Recent characterizations with higher resolution optical sensors have revealed rapid, local cholinergic modulation supporting dynamic changes in sensory processing, associative learning, and behavioral state4-7. However, sensory events that recruit cortical ACh often also change arousal and evoke movements, making it difficult to determine whether ACh transients reflect sensory features of environmental stimuli or the…

Small Interfering RNA Approaches for Targeting Nicotinic Acetylcholine Receptors: Current Evidence and Therapeutic Perspectives.

Nicotinic acetylcholine receptors (nAChRs) are pentameric ligand-gated ion channels that regulate diverse physiological processes in neuronal and non-neuronal tissues. In recent years, increasing evidence has shown that nAChRs are also expressed in many cancer cell types, where they contribute to tumor growth, survival, angiogenesis, migration, invasion, and resistance to therapy. The involvement of distinct receptor subunits in these processes has generated interest in nAChRs as potential therapeutic targets in oncology. However, conventional pharmacological agents often show limited subtype …

Insecticidal effects of oxazosulfyl result from synaptic inhibition by targeting the vesicular acetylcholine transporter.

Oxazosulfyl is the first insecticide in the sulfyl (alkylsulfone) class developed for rice protection. While the vesicular acetylcholine transporter (VAChT) has been proposed as its primary molecular target, its mode of action has not yet been extensively examined in major agricultural pests. In this study, we investigated the binding affinity of oxazosulfyl to VAChT derived from the brown planthopper (BPH), a significant rice pest, and evaluated the relationship between its insecticidal activity and binding affinity. Oxazosulfyl bound to VAChT, heterologously expressed in cultured cells, with…

Muscarinic acetylcholine receptor 3 localized to primary endothelial cilia regulates blood pressure and cognition.

We previously demonstrated that the inability of primary endothelial cilia to sense fluid shear stress can lead to nitric oxide (NO) deficiency and cause hypertension (HTN). Decreased biosynthesis of NO contributes to cerebral amyloid angiopathy in Alzheimer's disease (AD) patients through increased deposition of amyloid beta (Aβ). However, the molecular mechanisms underlying the pathogenesis of HTN and AD are incompletely understood. The objective of this study was to examine the pathophysiological roles of vascular primary cilia and muscarinic acetylcholine receptor 3 (CHRM3) in HTN and AD. …

Targeting α7 nicotinic acetylcholine receptors and their protein interactions in Alzheimer's disease drug development.

The decades-old cholinergic hypothesis of Alzheimer's disease (AD) led to clinical testing and FDA approval of acetylcholinesterase inhibitor drugs. Subsequently, the α7 nicotinic acetylcholine receptor (α7nAChR) was proposed as a new drug target for enhancing cholinergic neurotransmission. Nearly simultaneously, soluble amyloid β1-42 (Aβ42 ) was shown to bind α7nAChR with picomolar affinity to activate kinases that hyperphosphorylate tau, the precursor to tau-containing tangles. Multiple biopharmaceutical companies explored α7nAChR as a drug target for AD, mostly to enhance neurotransmission.…

Synaptotagmin-7 facilitates acetylcholine release in splanchnic nerve-chromaffin cell synapses during nerve activity.

Disturbances that threaten homeostasis elicit activation of the sympathetic nervous system (SNS) and the adrenal medulla. The effectors discharge as a unit to drive global and immediate changes in whole-body physiology. Descending sympathetic information is conveyed to the adrenal medulla via preganglionic splanchnic fibers. These fibers pass into the gland and synapse onto chromaffin cells, which synthesize, store, and secrete catecholamines and vasoactive peptides. While the importance of the sympatho-adrenal branch of the autonomic nervous system has been appreciated for many decades, the m

Maternal immunoglobulin treatment can reduce severity of fetal acetylcholine receptor antibody-associated disorders (FARAD).

BACKGROUND: Fetal acetylcholine receptor antibody-associated disorders (FARAD), caused by in utero exposure to maternal antibodies directed against the fetal acetylcholine receptor (AChR), is a rare condition occurring in newborns of myasthenic mothers. Only two cases of FARAD children born to asymptomatic mothers are published. CASE: We report a completely asymptomatic mother of two FARAD children presenting exclusively with positive AChR antibodies. After birth, the first child needed intensive care therapy due to generalized hypotonia, respiratory problems, dysphagia, necessitating tube fe…

Acetylcholine-Binding Protein Affinity Profiling of Neurotoxins in Snake Venoms with Parallel Toxin Identification.

Snakebite is considered a concerning issue and a neglected tropical disease. Three-finger toxins (3FTxs) in snake venoms primarily cause neurotoxic effects since they have high affinity for nicotinic acetylcholine receptors (nAChRs). Their small molecular size makes 3FTxs weakly immunogenic and therefore not appropriately targeted by current antivenoms. This study aims at presenting and applying an analytical method for investigating the therapeutic potential of the acetylcholine-binding protein (AChBP), an efficient nAChR mimic that can capture 3FTxs, for alternative treatment of elapid snake…

A bioassay for neuromuscular junction-restricted complement activation by myasthenia gravis acetylcholine receptor antibodies.

BACKGROUND: Myasthenia gravis (MG) is an autoimmune neuromuscular disorder hallmarked by fluctuating fatigable muscle weakness. Most patients have autoantibodies against acetylcholine receptors (AChRs) at the neuromuscular junction (NMJ). These are thought to have three possible pathogenic mode-of-actions: 1) cross-linking and endocytosis of AChRs, 2) direct block of AChRs and 3) complement activation. The relative contributions of these mechanisms to synaptic block and muscle weakness of individual patients cannot be determined. It likely varies between patients and perhaps also with disease …

Muscarinic Acetylcholine M2 Receptors Regulate Lateral Habenula Neuron Activity and Control Cocaine Seeking Behavior.

The lateral habenula (LHb) balances reward and aversion by opposing activation of brain reward nuclei and is involved in the inhibition of responding for cocaine in a model of impulsive behavior. Previously, we reported that the suppression of cocaine seeking was prevented by LHb inactivation or nonselective antagonism of LHb mAChRs. Here, we investigate mAChR subtypes mediating the effects of endogenous acetylcholine in this model of impulsive drug seeking and define cellular mechanisms in which mAChRs alter LHb neuron activity. Using in vitro electrophysiology, we find that LHb neurons are d…
