A pilot in vivo study: potential ovarian cancer therapeutic by placental extracellular vesicles.

The biological links between cancer and pregnancy are of interest due to parallel proliferative, immunosuppressive, and invasive mechanisms between tumour and placental cells. However, the proliferation and invasion of placental cells are strictly regulated. The understanding of this regulation is largely unknown. Placental extracellular vesicles (EVs) may play an important role in this regulation, as placental EVs are known to contribute to maternal adaptation, including adaptation of the vascular and immune systems. We have previously reported that placental EVs significantly inhibited ovari

Differential expression of skeletal muscle mitochondrial proteins in yak, dzo, and cattle: a proteomics-based study.

Changes in yak mitochondria by natural selection in a hypoxic environment could be utilized to understand adaptation to low-oxygen conditions. Therefore, the differences in proteome profile of skeletal muscle mitochondria from yak, dzo, and cattle were analyzed by mass spectrometry, which were then classified into 3 groups, comparing between yak and dzo, yak and cattle, and dzo and cattle. 376 unique mitochondrial proteins were identified, including 192, 191, and 281 proteins in the yak-dzo, yak-cattle, and dzo-cattle groups, respectively. NRDP1 and COQ8A were expressed at higher levels in yak

Characterization of a heme-protein responsive to hypoxia in Paracoccidioides brasiliensis.

Oxygen is fundamental to the life of aerobic organisms and is not always available to Paracoccidioides cells. During the life cycle stages, reduced oxygen levels directly affect general metabolic processes and oxygen adaptation mechanisms may play a fundamental role on fungal ability to survive under such condition. Heme proteins can bind to oxygen and participate in important biological processes. Several fungi, including Paracoccidioides, express a heme-binding globin (fungoglobin - FglA) presumable to regulate fungal adaptation to hypoxia. However, the characterization of fungoglobin in Par

Sex-specific functional adaptation of the femoral diaphysis to body composition.

OBJECTIVES: The human femoral diaphysis is often used to reconstruct loading histories (mobility, activity, body mass). The proximal femur is known to be differentially affected by changes in total fat-mass (FM), fat-free mass (FFM), and body fat percentage (BF%), but the adaptation of the entire diaphysis to body composition has not been thoroughly characterized to date. Understanding how the femoral diaphysis adapts to body components would benefit biomechanical interpretations of the femoral variation and nutrition-related studies. METHODS: Combining various methods from clinical nutrition

Marine invasions enter the genomic era: three lessons from the past, and the way forward.

The expanding scale and increasing rate of marine biological invasions have been documented since the early 20th century. Besides their global ecological and economic impacts, non-indigenous species (NIS) also have attracted much attention as opportunities to explore important eco-evolutionary processes such as rapid adaptation, long-distance dispersal and range expansion, and secondary contacts between divergent evolutionary lineages. In this context, genetic tools have been extensively used in the past 20 years. Three important issues appear to have emerged from such studies. First, the stud

Unique evolutionary trajectories in repeated adaptation to hydrogen sulphide-toxic habitats of a neotropical fish (Poecilia mexicana).

Replicated ecological gradients are prime systems to study processes of molecular evolution underlying ecological divergence. Here, we investigated the repeated adaptation of the neotropical fish Poecilia mexicana to habitats containing toxic hydrogen sulphide (H2 S) and compared two population pairs of sulphide-adapted and ancestral fish by sequencing population pools of >200 individuals (Pool-Seq). We inferred the evolutionary processes shaping divergence and tested the hypothesis of increase of parallelism from SNPs to molecular pathways. Coalescence analyses showed that the divergence occu

Biological substrates of emotional reactivity and regulation in adolescence during an emotional go-nogo task.

BACKGROUND: Adolescence is a transition period from childhood to adulthood that is often characterized by emotional instability. This period is also a time of increased incidence of anxiety and depression, underscoring the importance of understanding biological substrates of behavioral and emotion regulation during adolescence. Developmental changes in the brain in concert with individual predispositions for anxiety might underlie the increased risk for poor outcomes reported during adolescence. We tested the hypothesis that difficulties in regulating behavior in emotional contexts in adolesce

Product-Triggered Adaptive Evolution From a Pd4L2 to a Pd6L3 Conjoined Cage via Catalytic Dehydration of p‑Quinone Methide Precursors.

Supramolecular chemistry aims to emulate the dynamic structural adaptations and induced-fit conformational changes of enzymes that are fundamental to their biological function. In this work, we synthesized an adaptive Pd4L2 coordination cage using the newly developed pyridinium-based ligand L with cis-capped Pd (II) salt. Upon guest encapsulation, conformational adjustment provides direct structural evidence for the adaptive nature of cage 1 by X-ray crystallographic study. Within the hydrophobic pocket of the capsule, 4-(1-hydroxy-1-phenylethyl) phenol (S1), a known para-quinone methide (p-QM

Psoriasis: a mixed autoimmune and autoinflammatory disease.

In recent years marked progress has been made in our understanding of the critical biologic and immunologic pathways involved in psoriasis. Genetic studies have demonstrated that susceptibility to psoriasis involves components of both the adaptive and innate immune system and not surprisingly activation of both of these arms of the immune system is found in psoriatic skin. While adaptive immune responses predominate in chronic plaque psoriasis, innate and autoinflammatory responses dominate in pustular forms of psoriasis, with other clinical subtypes extending on a spectrum between plaque and

Host- and strain-specific regulation of influenza virus polymerase activity by interacting cellular proteins.

UNLABELLED: Highly pathogenic avian influenza A (HPAI) viruses of the H5N1 subtype have recently emerged from avian zoonotic reservoirs to cause fatal human disease. Adaptation of HPAI virus RNA-dependent RNA polymerase (PB1, PB2, and PA proteins) and nucleoprotein (NP) to interactions with mammalian host proteins is thought to contribute to the efficiency of viral RNA synthesis and to disease severity. While proteomics experiments have identified a number of human proteins that associate with H1N1 polymerases and/or viral ribonucleoprotein (vRNP), how these host interactions might regulate in

Critical Motor Involvement in Prediction of Human and Non-biological Motion Trajectories.

OBJECTIVES: Adaptive interaction with the environment requires the ability to predict both human and non-biological motion trajectories. Prior accounts of the neurocognitive basis for prediction of these two motion classes may generally be divided into those that posit that non-biological motion trajectories are predicted using the same motor planning and/or simulation mechanisms used for human actions, and those that posit distinct mechanisms for each. Using brain lesion patients and healthy controls, this study examined critical neural substrates and behavioral correlates of human and non-bi

The new chimeric chiron genes evolved essential roles in zebrafish embryonic development by regulating NAD+ levels.

The origination of new genes is important for generating genetic novelties for adaptive evolution and biological diversity. However, their potential roles in embryonic development, evolutionary processes into ancient networks, and contributions to adaptive evolution remain poorly investigated. Here, we identified a novel chimeric gene family, the chiron family, and explored its genetic basis and functional evolution underlying the adaptive evolution of Danioninae fishes. The ancestral chiron gene originated through retroposition of nampt in Danioninae 48-54 million years ago (Mya) and expanded

Effects of Occlusion on Periodontal Wound Healing.

Trauma from occlusion remains an important concept in dentistry. Traumatic occlusion affects the biological system through a series of events that induces an aseptic inflammatory response and bone resorption. Clinical evidence of these biological events is present in the form of active traumatic lesions or adaptive mechanisms. If these clinical signs are not identified prior to periodontal surgery, failure of the procedure could result due to an exacerbated leukocyte presence and unresolving wound. However, if traumatic occlusion is adjusted, evidence demonstrates that even large inflammatory

ORBIT: Oncogenic Representation Learning via Bi-Prototype Contrastive Learning in Hyperbolic Space for cancer driver gene identification.

Accurate identification of cancer driver genes is crucial for precision oncology but remains challenging due to the complexity of integrating heterogeneous data and modeling dynamic biological systems. To address these limitations, we propose ORBIT (Oncogenic Representation Learning via Bi-Prototype Contrastive Learning in Hyperbolic Space). Our framework synergistically fuses multi-omics profiles with functional network data using a context-adaptive graph reweighting mechanism to capture cancer-specific dynamics. The model employs a bi-prototype contrastive learning strategy within hyperbolic

The establishment and characteristics of cell-adapted IBV strain H120.

Avian infectious bronchitis virus is an important pathogen in poultry worldwide. Vaccination is the only effective way to prevent and control IBV infection. H120, one of the safest vaccine strains, which has been used worldwide as a primary vaccine, cannot adapt to passaged cells, which severely restricts the quality of the vaccine. Based on the reverse genetics of our previous research work, we constructed the recombinant R-H120-Beaudette-p65(S) strain by replacing the complete spike gene of H120 with the corresponding spike gene from the Beaudette p65 strain. Some biological characteristics,

Macrophages-Key cells in the response to wear debris from joint replacements.

The generation of wear debris is an inevitable result of normal usage of joint replacements. Wear debris particles stimulate local and systemic biological reactions resulting in chronic inflammation, periprosthetic bone destruction, and eventually, implant loosening, and revision surgery. The latter may be indicated in up to 15% patients in the decade following the arthroplasty using conventional polyethylene. Macrophages play multiple roles in both inflammation and in maintaining tissue homeostasis. As sentinels of the innate immune system, they are central to the initiation of this inflammat

Porphyrin pathways as targets for combating antimicrobial resistance: virulence and therapeutic opportunities.

Porphyrins are versatile biomolecules central to the synthesis of many essential molecules across all domains of life. Their ability to absorb light, accept electrons and facilitate redox reactions underpins diverse biological roles. Importantly, porphyrin properties can be easily tailored through substitutions of side chains and chelated metals. This adaptability has enabled porphyrins to play critical roles in bacterial environmental adaptations, most notably in enhancing competitive advantage and virulence. The association between porphyrin adaptations and virulence in pathogenic bacteria p
