The relationship between intestinal microbiota and the central nervous system in normal gastrointestinal function and disease.

Although many people are aware of the communication that occurs between the gastrointestinal (GI) tract and the central nervous system, fewer know about the ability of the central nervous system to influence the microbiota or of the microbiota's influence on the brain and behavior. Within the GI tract, the microbiota have a mutually beneficial relationship with their host that maintains normal mucosal immune function, epithelial barrier integrity, motility, and nutrient absorption. Disruption of this relationship alters GI function and disease susceptibility. Animal studies suggest that pertur

Ablation of connexin43 in uterine smooth muscle cells of the mouse causes delayed parturition.

Gap junctions are characteristically increased in the myometrium during term and preterm delivery and are thought to be essential for the development of uterine contractions during labour. Expression of connexin43 (Cx43), the major myometrial gap junction protein, is increased during delivery. We have generated a mouse mutant (Cx43fl/fl:SM-CreERT2), in which the coding region of Cx43 can be specifically deleted in smooth muscle cells at any given time point by application of tamoxifen. By this approach, we were able to study long-term effects on myometrial functions that are necessary for part

Distribution of 3,4-methylenedioxymethamphetamine (MDMA) and 3,4-methylenedioxyamphetamine (MDA) stereoisomers in a fatal poisoning.

This communication presents the quantitation and differential distribution of the enantiomers of 3,4-methylenedioxymethamphetamine (MDMA) and its physiologically active metabolite 3,4-methylenedioxyamphetamine (MDA) in a fatal poisoning following insufflation of MDMA, cocaine and heroin. Animal studies have demonstrated the stereoselective pharmacokinetics and neurotoxicity of these compounds; however, enantiomeric distributions have not been reported in humans. Quantitation of MDMA and MDA enantiomer was by gas chromatography/mass spectrometry (GC/MS) following chiral derivatization with N-tr

Effects of AFQ056 on language learning in fragile X syndrome.

BACKGROUNDFXLEARN, the first-ever large multisite trial of effects of disease-targeted pharmacotherapy on learning, was designed to explore a paradigm for measuring effects of mechanism-targeted treatment in fragile X syndrome (FXS). In FXLEARN, the effects of metabotropic glutamate receptor type 5 (mGluR5) negative allosteric modulator (NAM) AFQ056 on language learning were evaluated in 3- to 6-year-old children with FXS, expected to have more learning plasticity than adults, for whom prior trials of mGluR5 NAMs have failed.METHODSAfter a 4-month single-blind placebo lead-in, participants wer

Whose Knowledge? Examining the Relationship between the Traditional Medicine Sector and Environmental Conservation Using a Stakeholder Analysis: Perceptions on Warwick Herb Market Durban South Africa.

The South African traditional medicine sector is estimated to accommodate millions of citizens, despite it being informal. The existence of such a healthcare system embodies the dual system of both primary and traditional healthcare, with some preferring one and others utilising both systems. The gathering, harvesting, and selling of medicinal plant and animal species have inevitable environmental effects. The paradox between biodiversity conservation and livelihood sustenance is eminent in South Africa's contemporary environmental legislation. The purpose of the study was to highlight and exa

Disturbed immuno-endocrine communication via the hypothalamo-pituitary-adrenal axis in autoimmune disease.

Previous studies in our laboratory demonstrated an altered immuno-endocrine feedback communication via the hypothalamo-pituitary-adrenal (HPA) axis, which may be an important modulatory factor in the development of spontaneous autoimmune thyroiditis in Obese strain (OS) chickens. These birds show a significantly lower, or even absent, increase in serum glucocorticoid levels in response to an intravenous injection of antigen or conditioned medium (CM) from mitogen-stimulated spleen cells known to contain glucocorticoid-increasing factors (GIFs), notably interleukin-1 (IL-1). The present study w

Advancing Health Security and Disease Eradication Through Peace and Health: A Mali Case Study.

Conflict and violence constitute threats to public health. As levels of conflict increase within and between countries, it is important to explore how conflict resolution initiatives can be adapted to meet the health needs of communities, and how addressing the health needs of communities can assist in conflict resolution and contribute to health security. In conflict-affected central Mali, a Peace through Health Initiative, piloted between 2018 and 2022, used conflict resolution trainings, facilitated community meetings, and human and animal health interventions to negotiate "periods of tranq

Preparation of Ex Vivo Rodent Phantoms for Developing, Testing, and Training MR Imaging of the Kidney and Other Organs.

Here we describe a simple and inexpensive protocol for preparing ex vivo rodent phantoms for use in MR imaging studies. The experimental animals are perfused and fixed with formaldehyde, and then wrapped with gauze and sealed with liquid latex. This yields a phantom that preserves all organs in situ, and which avoids the need to keep fixed animals and organs in containers that have dimensions very different from living animals. This is especially important for loading in MR detectors, and specifically the RF coils, they are usually used with. The phantom can be safely stored and conveniently r

Extended-spectrum beta-lactamase (ESBL)-producing Enterobacteriaceae in cattle production - a threat around the world.

Food producing animal is a global challenge in terms of antimicrobial resistance spread. Extended-spectrum beta-lactamase (ESBL)-producing Enterobacteriaceae are relevant opportunistic pathogens that may spread in many ecological niches of the One Health approach as human, animal and environment due to intestinal selection of antimicrobial resistant commensals in food production animals. Cattle production is a relevant ecological niche for selection of commensal bacteria with antimicrobial resistance from microbiota. Enterobacteriaceae show importance in terms of circulation of resistant-bacte

High prevalence and diversity of Leptospira pathogenic serogroups in pigs, cows and free roaming dogs from undeserved rural communities in the coastal region of Ecuador.

Leptospirosis is a zoonotic disease with global distribution but endemic to tropical regions that affects animal production systems. In Ecuador, leptospirosis is endemic with records of human outbreaks dated from more than one century ago, and where the role of animal reservoirs in the transmission dynamics is not well understood. In this study, we carried out a serological surveillance by Microagglutination Test with a panel of 24 serovars for 545 animals represented by pigs and cows, and free roaming dogs from undeserved rural communities in the province of Guayas (Coastal Region of Ecuador)

The skin microbiome of cow-nose rays (Rhinoptera bonasus) in an aquarium touch-tank exhibit.

Public aquaria offer numerous educational opportunities for visitors while touch-tank exhibits offer guests the ability to directly interact with marine life via physical contact. Despite the popularity of touch-tanks, there is a paucity of research about animal health in these exhibits and, in particular, there is little research on the microbial communities in these highly interactive exhibits. Microbial community structure can have implications for both host health and habitat function. To better understand the microbiome of a touch-tank we used high-throughput sequencing of the 16S rRNA ge

What is needed to achieve effective and sustainable control of African animal trypanosomosis?

A welcome resurgence in African animal trypanosomosis (AAT) research has resulted in advances in capabilities, foundational datasets, and understanding. Additionally, there is the prospect of the first new trypanocide in >60 years. However, it is vital to ensure that advances translate to improved and sustainable control in the field. A recent meeting, the Symposium on African Livestock Trypanosomes - Tanzania, convened stakeholders from across the spectrum of AAT research and control to ask how this can be achieved. Current constraints on progress were defined, as were critical gaps and oppor

The multifaceted resources and microevolution of the successful human and animal pathogen methicillin-resistant Staphylococcus aureus.

Methicillin-resistant Staphylococcus aureus (MRSA) is one of the most important bacterial pathogens based on its incidence and the severity of its associated infections. In addition, severe MRSA infections can occur in hospitalised patients or healthy individuals from the community. Studies have shown the infiltration of MRSA isolates of community origin into hospitals and variants of hospital-associated MRSA have caused infections in the community. These rapid epidemiological changes represent a challenge for the molecular characterisation of such bacteria as a hospital or community-acquired

Lymphovenous Anastomoses Between Thoracic Duct and Azygos Vein in a Human Cadaver: A Case Report.

OBJECTIVE: The study adds valuable information regarding lymphovenous communications between the thoracic duct and the azygos vein, which are very rarely discovered during anatomical dissections and very few cases have been mentioned worldwide. A detailed description of our findings and a brief review of the relevant literature are also provided. CASE REPORST: In the current study, two sizeable obliquely directed lymphovenous anastomoses between the thoracic duct and the azygos vein at the midportion of the mediastinum are described in the same cadaver. CONCLUSION: The existence of such anas

Osteomyelitis related lameness across animal species: pathophysiology, etiology, epidemiology and experimental model approaches.

Osteomyelitis is an inflammatory disorder resulting from the invasion of opportunistic pathogens into bones and bone marrows, causing necrosis and affecting multiple animal groups. It represents one of the most prevalent musculoskeletal diseases, imposing substantial economic losses and raising significant animal welfare concerns across livestock and poultry industries. The pathophysiology of osteomyelitis is characterized by mechanisms of disease development, clinical age, the specific anatomic site in the bone, and structural differences between adults and juveniles. The inability of animals

One Health in Mesoamerica: historical legacies, current challenges, and strategic directions.

BACKGROUND: Mesoamerica, a biodiversity hotspot extending from Mexico to Panama, faces intersecting health challenges rooted in colonial-era transformations and amplified by contemporary environmental change. This perspective paper demonstrates how historical legacies interact with current ecological and social drivers to shape zoonotic disease vulnerability across the region. APPROACH: We integrate historical analysis of colonial disease introduction with an examination of current environmental, climatic, and social determinants of disease transmission. Findings from a structured multisector

The carcinogenicity of lead.

The potential for lead to cause neoplasm in animals and man is reviewed. A multitude of studies indicate that principally renal tumours may be produced by various forms of inorganic lead in small rodents. No human study either of an epidemiological form or of a case report in industrial, agricultural or community medicine has proven that lead may cause cancer in man.

Mitigating antimicrobial resistance in the animal industry: reduction strategies and alternatives to antibiotics.

Antimicrobial resistance (AMR) has become a worldwide issue of human and animal health, food security, and environmental sustainability due to its seriousness and the challenge of resolving it. Widespread utilization of antibiotics in the animal industry for metaphylactic, therapeutic, production, and prophylactic purposes has significantly contributed to the spread and emergence of antibiotic-resistant microorganisms. Food- producing animals are also deemed as good reservoirs of "antimicrobial resistance genes (ARGs) and antimicrobial- resistant bacteria (ARB)" that may contaminate humans thr

Tungiasis in domestic, wild, and synanthropic animals in Brazil.

Tungiasis is an ectoparasitic skin disease of humans and warm-blooded animals caused by fleas of the genus Tunga (Jarocki, 1938). Dogs, cats, pigs, and synanthropic rodents are the main animal reservoirs. Of the 14 known species, 12 are found in Latin America and, among these, eight are found in Brazil, including the two zoonotic species Tunga penetrans and Tunga trimamillata. Tunga penetrans predominates among humans and Brazilian pets, where the dog is the most affected species, and its presence is a risk factor associated with the disease in humans. Tunga spp. are widely distributed through

Antimicrobial Resistance in Farm Animals in Brazil: An Update Overview.

In animal husbandry, antimicrobial agents have been administered as supplements to increase production over the last 60 years. Large-scale animal production has increased the importance of antibiotic management because it may favor the evolution of antimicrobial resistance and select resistant strains. Brazil is a significant producer and exporter of animal-derived food. Although Brazil is still preparing a national surveillance plan, several changes in legislation and timely programs have been implemented. Thus, Brazilian data on antimicrobial resistance in bacteria associated with animals co
