Comparison of impedance-based sensors for cell adhesion monitoring and in vitro methods for detecting cytotoxicity induced by chemicals.

The recent European Commission REACH (Registration, Evaluation and Authorisation of Chemicals) policy outlines a plan for toxicological testing by using alternative non-animal in vitro methods. In this context, there is a need to develop and standardise high-throughput screening (HTS) methods for studying the cytotoxicity induced by chemicals. Electrochemical impedance spectroscopy (EIS) can be considered as a complementary technique to alternative in vitro testing for studying cell adhesion to the substrate, and can give real-time and kinetic information on cell responses to a toxicant. This

Repurposing Cryopreserved Slaughterhouse Waste for Vascular Replacement Therapy.

Current vascular replacement therapy (VRT) standards primarily rely on autografting and allografting. However, autografting is associated with substantial donor site comorbidities, while the limited availability of graft tissues constrains allografting. Although synthetic alternatives are widely accessible, they are prone to long-term failure. This study explores an alternative approach to help address the critical supply-demand mismatch for VRT by repurposing cryopreserved slaughterhouse-derived waste aortas. Using an in vitro perfusion-based model, we demonstrated that these tissues maintain

Combating Antimicrobial Resistance by Employing Antimicrobial Peptides: Immunomodulators and Therapeutic Agents against Infectious Diseases.

The rising prevalence of multiple-drug-resistant pathogens poses a formidable challenge to conventional antimicrobial treatments. The inability of potent antibiotics to combat these "superbugs" underscores the pressing need for alternative therapeutic agents. Antimicrobial peptides (AMPs) represent an alternative class of antibiotics. AMPs are essential immunomodulatory molecules that are found in various organisms. They play a pivotal role in managing microbial ecosystems and bolstering innate immunity by targeting and eliminating invading microorganisms. AMPs also have applications in the ag

Conductive hearing loss results in a decrease in central auditory system activity in the young gerbil.

OBJECTIVES/HYPOTHESIS: The impact of childhood conductive HL (CHL) on development of auditory function has long been debated. The present study was conducted to define and compare the consequences of CHL and cochlear ablation (CA) in young and adult animals, using 2-deoxyglucose (2-DG) uptake as a measure of metabolic activity. It was hypothesized that, for both ages, CHL would result in a decrease in activity in the major ascending central auditory system pathway of the manipulated ear, but that this decrease would be significantly less than that observed with CA. STUDY DESIGN: Sham-controll

A two-year study of Salmonella in four natural watersheds highlights the need for increased environmental Salmonella surveillance to close the One Health loop.

Salmonella enterica is a leading bacterial cause of foodborne illness, often transmitted through contaminated food and water. Improved food handling has led to considerable reductions of Salmonella contamination in meat and poultry products; this does not wholly contribute to decreased salmonellosis incidence, highlighting the need to define alternative reservoirs and transmission pathways. In this study, we collected samples from four distinct watersheds over 24 months to characterize Salmonella serovar diversity and utilized phylogenetic approaches, along with proximal land use analyses, to

Effects of prebiotics and precision biotics on performance, animal welfare and environmental impact. A review.

This review aims to analyze the recent studies about prebiotics and precision biotics, as alternatives to animal growth promoters. These substances improve intestinal health, growth performance and poultry environmental impact. Prebiotics are insoluble fibers, that have no nutritive value, but they promote the growth of positive bacteria, increase the nutrients absorption and modulate the immune response. Instead, precision biotics are carbohydrates with glycosidic linkages, which interact with gut bacteria metabolism, reducing the excretion of nitrogen and consequentially, the poultry environ

Toxicity of a methotrexate metronomic schedule in Wistar rats.

Metronomic chemotherapy is a relevant strategy that uses low doses of antineoplastic drugs for sustained periods to control tumor growth, an alternative frequently utilized in veterinary patients. This work aimed to evaluate the toxic effects of a metronomic oral dose of methotrexate (MTX) for 45 days in tumor-free Wistar rats when compared with control animals. Clinical alterations, body weight, food, and water intake were monitored daily, and bone marrow suppression, hematological, biochemical, and histopathological analyses were performed at three points (days 30, 45, and 60). MTX-treated a

Immunization with Brucella abortus S19Δper Conferred Protection in Water Buffaloes against Virulent Challenge with B. abortus Strain S544.

Vaccination of cattle and buffaloes with Brucella abortus strain 19 has been the mainstay for control of bovine brucellosis. However, vaccination with S19 suffers major drawbacks in terms of its safety and interference with serodiagnosis of clinical infection. Brucella abortus S19∆per, a perosamine synthetase wbkB gene deletion mutant, overcomes the drawbacks of the S19 vaccine strain. The present study aimed to evaluate the potential of Brucella abortus S19Δper vaccine candidate in the natural host, buffaloes. Safety of S19∆per, for animals use, was assessed in guinea pigs. In silico analyses and design of chimeric proteins containing epitopes of Bartonella henselae antigens for the control of cat scratch disease.

Bartonella henselae is a Gram-negative bacterium that causes cat scratch disease (CSD), as well as bacteremia, endocarditis, and other clinical presentations. CSD remains one of the most common infections caused by bacteria in the genus Bartonella, and it is transmitted to humans through a scratch or cat bite. Vaccination and more efficient diagnostic methods would represent a promising and sustainable alternative measure for CSD control in humans and animals. Here, we described the in silico analyses and design of three recombinant chimeric proteins (rC1, rC2, and rC3), for use in the control

Parathyroid xenotransplantation without immunosuppression in experimental hypoparathyroidism: long-term in vivo function following microencapsulation with a clinically suitable alginate.

Permanent hypoparathyroidism is one of the most difficult of all endocrine disorders to treat medically. Because this deficiency syndrome rarely is a life-threatening condition, systemic immunosuppression for recipients of transgenic transplants is not justified. An alternative would be protecting the tissue to be transplanted from the immunologic response (immunoisolation) by coating it with a semipermeable membrane- microencapsulation. Unfortunately, prior to the first clinical use, further analysis of the coating substance (alginate) demonstrated that it has potential cancerogenic propertie

Luminescence spectroscopy as a screening tool for the potential carcinogenicity of asphalt fumes.

A subset of polycyclic aromatic compounds (PACs), which contain 4-6 annulated rings, has been documented as the source of carcinogenicity in animal skin painting studies of petroleum products and asphalt fumes (M. L. Machado, P. W. Beatty, J. C. Fetzer, A. H. Glickman and E. L. McGinnis, Fundam. Appl. Toxicol., 1993, 21, 492; T. A. Roy, S. W. Johnson, G. R. Blackburn and C. R. Mackerer, Fundam. Appl. Comparison of neurotization versus nerve repair in an animal model of chronically denervated muscle.

PURPOSE: Reinnervation of chronically denervated muscle is clinically unpredictable and poorly understood. Current operative strategies include either direct nerve repair, nerve grafting, nerve transfer, or neurotization. The goal of this study is to compare muscle recovery using microneural repair versus neurotization in a rat model of chronic denervation. METHODS: Fifty-eight Sprague-Dawley rats had surgical denervation of the tibialis anterior muscle by transecting the common peroneal nerve. After 0, 8, 12, or 22 weeks of denervation, animals were assigned to either a direct repair or a ne

Nutrition and Metabolism: Foundations for Animal Growth, Development, Reproduction, and Health.

Consumption of high-quality animal protein plays an important role in improving human nutrition, growth, development, and health. With an exponential growth of the global population, demands for animal-sourced protein are expected to increase by 60% between 2021 and 2050. In addition to the production of food protein and fiber (wool), animals are useful models for biomedical research to prevent and treat human diseases and serve as bioreactors to produce therapeutic proteins. For a high efficiency to transform low-quality feedstuffs and forages into high-quality protein and highly bioavailable

Selective control of Rhipicephalus microplus in a dairy cattle herd from different genetic groups.

Alternatives for Rhipicephalus microplus control are needed in the light of its resistance to acaricides. One of the ways to decrease the use of acaricides in a herd is selective control (SC). In the present study, SC was evaluated in a dairy herd consisting of different genetic groups: Holstein, Jersey, crossbreed and Girolando. Ticks were counted in the right anterior third region on around 90 cows, totaling nine evaluations at intervals of 21 days. Commercial pour-on acaricide was applied only when the infestation was greater than or equal to eight ticks larger than 4 mm in the anterior thi

Secretion-based production of prolyl-hydroxylated human type III collagen in scalable Physcomitrella photobioreactors.

Scalable moss bioreactors enable the production of high-quality recombinant prolyl-hydroxylated human collagen without heterologous P4H expression, offering a sustainable and vegan alternative to conventional collagens derived from animals. Collagens are structural proteins of the extracellular matrix essential for skin elasticity and integrity. They are widely used in dietary supplements and cosmetics. Conventional collagens of animal origin raise concerns regarding ethics, safety, and sustainability. As a vegan alternative, we report on the production of a 30 kDa prolyl-hydroxylated human co

In Vivo Acaricidal Effect of Linoleic Acid and Olive Oil Against Psoroptes cuniculi Mites in Experimentally Infested Rabbits.

PURPOSE: Psoroptes cuniculi is the most prevalent mite affecting rabbits worldwide. It is the primary cause of otitis in this species and significantly compromises animal welfare, leading to considerable economic losses. In search of alternative control strategies, several plant-derived oils such as olive, cinnamon, oregano, neem, and lemon oils have been evaluated in vitro for their acaricidal potential. In this study, the in vivo efficacy of extra virgin olive oil (EVOO) and linoleic acid (LA) was assessed as alternative treatments against P. cuniculi. METHODS: Twenty-four rabbits were expe
