A high-throughput integrated biofilm-on-a-chip platform for the investigation of combinatory physicochemical responses to chemical and fluid shear stress.

Physicochemical conditions play a key role in the development of biofilm removal strategies. This study presents an integrated, double-layer, high-throughput microfluidic chip for real-time screening of the combined effect of antibiotic concentration and fluid shear stress (FSS) on biofilms. Biofilms of Escherichia coli LF82 and Pseudomonas aeruginosa were tested against gentamicin and streptomycin to examine the time dependent effects of concentration and FSS on the integrity of the biofilm. A MatLab image analysis method was developed to measure the bacterial surface coverage and total fluor

Comparative efficacy of teat sealants given prepartum for prevention of intramammary infections and clinical mastitis: a systematic review and network meta-analysis.

A systematic review and network meta-analysis were conducted to assess the relative efficacy of internal or external teat sealants given at dry-off in dairy cattle. Controlled trials were eligible if they assessed the use of internal or external teat sealants, with or without concurrent antimicrobial therapy, compared to no treatment or an alternative treatment, and measured one or more of the following outcomes: incidence of intramammary infection (IMI) at calving, IMI during the first 30 days in milk (DIM), or clinical mastitis during the first 30 DIM. Risk of bias was based on the Cochrane

Emerging drugs for gastro-oesophageal reflux disease.

Gastro-oesophageal reflux disease is a common medical problem caused by the exposure of the distal oesophagus to gastric contents. Existing medical therapy is very effective, but symptomatic relief with acid suppressants is often delayed. Treatment focuses on the suppression of gastric acid rather than on the underlying pathophysiological abnormalities, such as transient non-swallow-related lower oesophageal sphincter relaxation. Current pharmacological developments concentrate on drugs with lasting acid suppression and a faster onset of action. Compounds interacting with the complex neuromusc

Review article: gastro-oesophageal reflux disease as a functional gastrointestinal disorder.

Gastro-oesophageal reflux disease encompasses a family of disorders united by a common pathogenesis: the excessive reflux of gastric contents. The best defined disorder is oesophagitis, but it is now evident that proton pump inhibitor therapy can resolve oesophagitis in virtually every case. The same cannot be said for gastro-oesophageal reflux disease symptoms, however, thus moving the frontier to symptom control. Endoscopy-negative reflux disease is a heterogeneous group, no longer bound by the common denominator of endoscopic findings. In fact, three distinct patient subgroups emerge: (i) i

Sleep issues in gastroesophageal reflux disease: beyond simple heartburn control.

There is a burgeoning interest in the relationship between sleep-related gastroesophageal reflux (GER) and the development of esophageal and extra esophageal complications. The physiological changes associated with sleep, such as suspension of the regulation of body temperature, may influence nocturnal GER and esophageal acid clearance. Data indicate that sleep induces considerable risk of prolonged acid mucosal contact and facilitates the occurrence of proximal migration of acid, thereby increasing the probability of pulmonary aspiration. Nocturnal GER can lead to the development of esophagit

Treatment options for functional dyspepsia.

Non-ulcer dyspepsia is a common condition in primary care. The treatment of this condition is still controversial and no single therapy is uniformly effective. In this article we review currently available treatment trials for non-ulcer dyspepsia.

Suppression of acid secretion in peptic ulcer disease.

Until recently, suppression of gastric acid secretion in patients with peptic ulcer was empirical and of unproven value. Anticholinergic drugs had only modest inhibitory effects on acid secretion, many side effects, and uncertain efficacy. Controlled trials using antacids demonstrated the value of reducing gastric acidity for healing duodenal ulcer. The discovery of histamine-2 (H2) receptor antagonists in the 1970s and the introduction of H+,K(+)-ATPase inhibitors in the 1980s made reduction of acid secretion the first-choice modality for healing and preventing recurrences of duodenal and gas…

Effects of acid suppression on microbial flora of upper gut.

Decreased acid secretion, due to therapy or disease, predisposes to increased bacterial counts in gastric juice. As bacterial numbers increase, the number of nitrate-reducing strains and the concentration of luminal nitrite usually also increase. However, there is controversy (mainly because of assay problems) about whether decreased acid increases generation of N-nitroso compounds: these may be produced by acid or by bacterial catalysis, and the relative contributions of each are still uncertain. Other potentially important factors include ascorbate secretion (can prevent nitrite conversion t

Effects of protective drugs on gastric alkaline secretion in man.

This study was designed to determine the effects of sucralfate, De-Nol, and Maalox 70 on gastric HCO3 secretion in 34 healthy humans. Alkaline secretion was measured after pretreatment with ranitidine to abolish H+ secretion, using a constant perfusion-aspiration system and back-titration of the perfusates to the original pH 6.0. Luminal release of PGE2 was also measured in the gastric perfusates. Addition of sucralfate or De-Nol resulted in increments of gastric HCO3 secretion, reaching about 45% and 59%, respectively, of the maximal HCO3 response to 16,16-dimethyl PGE2 (dmPGE2). The highest

[In vitro effect of 2 pharmaceutical preparations containing bismuth on the fermentative capacity of intestinal bacteria].

An investigation was done on the action in vitro of two pharmaceutical preparations containing Bi, De Nol and Pepto Bismol, on the fermentative capacity of intestinal bacteria. For the purpose, using an instrument and a technique previously described, fecal fermentation (FF) was measured in 21 stool samples, to which lactose was added to assure appreciable levels of fermentation. In 11 of the samples, FF was measured without and with addition of De Nol; and in the 10 remaining samples, without and with addition of Pepto Bismol. In 7 of the samples with which the action of De Nol was studied, t

Treatment of gastric ulcer. What is old and what is new.

The rational treatment of gastric ulcer (GU) requires both an understanding of the various causative factors responsible for what is best considered a spectrum of disorders, as well as a familiarity with the newer antisecretory and cytoprotective therapies that are available. Gastric ulcers that fulfill the criteria to be peptic ulcers (ie, occur in the antrum in the presence of excess luminal acid, with or without a coexisting duodenal ulcer) are best treated with a histamine H2-receptor antagonist. For GUs that develop in the setting of normal or reduced acid output, or those that occur as a

A new look at peptic ulcer.

There have been notable advances in knowledge about peptic ulcer recently. Gastrin-producing tumors have been recognized as a rare cause of ulcer, and multiple physiologic defects have been found in duodenal ulcer, including excessive release of gastrin after food intake, increased sensitivity to gastrin, and decreased inhibition by low pH. The tendency of gastric ulcer patients to reflux duodenal contents into the stomach may have pathogenetic significance. Two new classes of drugs strongly inhibit acid secretion in man: chemically modified prostaglandins and histamine analogues which block t

Drugs, diseases and altered gastric emptying.

Drugs are usually given orally. They are not absorbed to any extent from the stomach but may be absorbed very rapidly from the small intestine. Thus factors influencing the rate of gastric emptying may alter the rate of absorption of most if not all orally administered drugs. Food, hormones, posture, peritoneal irritation, severe pain, gastric ulcer, diabetes and other metabolic diseases, as well as drugs such as alcohol, anticholinergics, narcotic analgesics, ganglion blocking drugs, antacids and metoclopramide all influence the rate of gastric emptying and they will, in turn, change the rate…
