Encapsulation of Disease-Causing and Commensal Mitis Group Non-Pneumococcal Streptococci.

Background: Mitis group non-pneumococcal streptococci (MGNPS), specifically Streptococcus mitis, Streptococcus infantis and Streptococcus oralis, have recently been shown to cause pneumonia and/or bacteremia. These organisms often have capsular (cps) operons resembling those in pneumococci, and some express cps-generated polysaccharides that antigenically cross-react with pneumococcal capsular serotypes. But, to date, a series of MGNPS isolates has not been studied by electron microscopy (EM) for the presence of a capsule. Methods: We studied 21 MGNPS; 11 were isolated from sputum and determin

Preparation of TTO/UF resin microcapsule via in situ polymerisation and modelling of its slow release.

For slow release of tea tree oil (TTO), TTO were encapsulated by urea-formaldehyde (UF) resin via in situ polymerisation. The effects of curing time and drying condition on particle size, and TTO loading of the TTO/UF microcapsules were investigated. The results indicated that TTO/UF resin microcapsules with curing time of 80 min had narrow size distribution and good wall cover. Drying at ambient was better to maintain the TTO content than drying at oven. The loading of TTO with curing time of 80 min can be up to 45 wt.% of the mass-proportion to the prepared microcapsules, and more than 90 wt

Mutations in pneumococcal cpsE generated via in vitro serial passaging reveal a potential mechanism of reduced encapsulation utilized by a conjunctival isolate.

UNLABELLED: The polysaccharide capsule of Streptococcus pneumoniae is required for nasopharyngeal colonization and for invasive disease in the lungs, blood, and meninges. In contrast, the vast majority of conjunctival isolates are acapsular. The first serotype-specific gene in the capsule operon, cpsE, encodes the initiating glycosyltransferase and is one of the few serotype-specific genes that can tolerate null mutations. This report characterizes a spontaneously arising TIGR4 mutant exhibiting a reduced capsule, caused by a 6-nucleotide duplication in cpsE which results in duplication of Ala

Sortase-conjugation generates a capsule vaccine that protects guinea pigs against Bacillus anthracis.

Capsules protect bacteria against phagocytic clearance. Capsular polysaccharides or polyglutamates have evolved also to resist antigen presentation by immune cells, thereby interfering with the production of opsonophagocytic antibodies. Linking capsular material to a carrier protein stimulates its presentation to the immune system. For many conjugate vaccines this is achieved by a process of random chemical cross-linking. Here we describe a new technology, designated sortase-conjugation, which generates a single amide bond between the C-terminal end of a carrier protein and the capsular materi

Significant variation in serotype-specific immunogenicity of the seven-valent Streptococcus pneumoniae capsular polysaccharide-CRM197 conjugate vaccine occurs despite vigorous T cell help induced by the carrier protein.

Streptococcus pneumoniae capsular polysaccharides (PnPSs) induce protective antibodies but are T cell-independent type 2 antigens and are poorly immunogenic in infants. Conjugate vaccines of PnPSs linked to proteins like cross-reactive material (CRM(197)) increase PS antibody titer and elicit immunologic memory in infants. Despite being linked to an identical carrier protein, each PS component of the 7-valent PnPS-CRM(197) vaccine has different immunogenicity. To determine whether variations in conjugate-induced memory T cell responses or PnPS-specific antibody-secreting cells (ASCs) were resp

Follicle-Oriented "Acid-Free" Supramolecular Microcapsules Achieve Efficient Acne Therapy via Skin Microbiota Remodeling.

Acne vulgaris is a multifactorial inflammatory disorder involving follicular obstruction, excessive sebum production, Cutibacterium acnes (C. acnes) colonization, immune activation, and barrier dysfunction. Current topical treatments are often limited by cutaneous irritation, antimicrobial resistance, and insufficient follicular deposition. Here, an acid-free, pH-responsive, follicle-oriented supramolecular polycystic microcapsule (SPM) particles system was developed based on a phosphatidylcholine/pterostilbene deep eutectic solvent (PC/PT DES). In this study, "SPM particles" refers to the opt

Wet-Heating-Induced Maillard Conjugation Combined With Rosemary Extract Co-Encapsulation Improves the Stability of Complex-Coacervated Citronella Oil Microcapsules.

A combined treatment integrating black bean protein isolate-gum Arabic (BBPI-GA) wet-heating conjugation and rosemary extract co-encapsulation was employed to improve the physicochemical characteristics and stability of citronella oil microcapsules. Changes in color, turbidity, molecular weight, and protein secondary structure confirmed the formation of BBPI-GA conjugates, with an optimal heating time of 1 h. Wet-heating conjugation increased the encapsulation efficiency (EE) of citronella oil from 91.97% (non-conjugated control) to 97.62% and produced microcapsules with a denser morphology an

Rapid microbiological method for analysis of clindamycin-based product potency and its equivalence to UV and HPLC methods.

Clindamycin is an antimicrobial sold in capsules, injectable solution, or gel for topical use. Despite its wide use, current official compendia and literature lack microbiological methods to assess its potency, relying mainly on physicochemical assays such as high-performance liquid chromatography (HPLC). These techniques, however, do not evaluate the biological activity of the drug. In this context, the present study proposes, for the first time, aimed to develop and validate a rapid microbiological method to determine the potency of clindamycin in capsule form and to compare its results with

Nanoniosome-encapsulated levoflaxicin as an antibacterial agent against Brucella.

A study was conducted to examine the prevalence of brucellosis (in animal farms) in the vicinity of Islamabad and Rawalpindi. A total of 170 milk samples were collected randomly from several farmhouses. The collected milk samples were initially screened by a Brucella selective medium. The bacterial isolates grown on the selective medium were subjected to biochemical identification for further confirmation of Brucella species. Among the tested samples, 28 (16.4%) were found positive for selective medium and 14 (8.2%) were found positive after biochemical confirmation. The antimicrobial suscepti

In vitro interaction of antifungal and antibacterial drugs against Cryptococcus neoformans var. grubii before and after capsular induction.

This study evaluated the synergistic interactions between amphotericin B (AMB) and azithromycin (AZM), daptomycin (DAP), linezolid (LNZ), minocycline (MINO), fluconazole (FLZ), flucytosine (5FC), linezolid (LZD), or tigecycline (TIG) against clinical isolates of Cryptococcus neoformans var. grubii before and after capsule induction. High synergism (>75%) was observed for the combinations, AMB+5FC, AMB+TIG, AMB+AZM, AMB+LZD and AMB+MINO but only in the strains after capsule induction. The results show that the presence of the capsule may lower the minimum inhibitory concentrations (MICs) of ant

Recognition via the class A scavenger receptor modulates cytokine secretion by human dendritic cells after contact with Neisseria meningitidis.

Proinflammatory cytokines play a major role in the pathogenesis of meningococcal disease and their serum levels in patients are correlated with the outcome of infection. Dendritic cells initiate immunity against Neisseria meningitidis and are a major source of proinflammatory cytokines. Here we show that physical interaction of human DC with N. meningitidis via the class A scavenger receptor (SRA) modulates cytokine release by DC. Phagocytosis and uptake via SRA were shown to increase release of TNF-alpha, IL-1 beta and IL-6. In contrast, secretion of IL-8 is enhanced after recognition of N. m

Human neutrophil chemotaxis is modulated by capsule and O antigen from an extraintestinal pathogenic Escherichia coli strain.

Gram-negative enteric bacilli, such as Escherichia coli, are common causes of nosocomial pneumonia. The interaction between pulmonary neutrophils and the infecting pathogen is a critical step in determining the outcome. Previous studies from our laboratory, for which a rat model of pneumonia was used, established that pulmonary neutrophil recruitment was modulated by the E. coli virulence factors capsule and O-specific antigen. To begin to understand the mechanism by which this recruitment occurs, we conducted in vitro and ex vivo chemotaxis assays, for which we used a clinically relevant E. c

The Short-Chain Fatty Acids Propionate and Butyrate Augment Adherent-Invasive Escherichia coli Virulence but Repress Inflammation in a Human Intestinal Enteroid Model of Infection.

Short-chain fatty acids (SCFAs), which consist of six or fewer carbons, are fermentation products of the bacterial community that inhabits the intestine. Due to an immunosuppressive effect on intestinal tissue, they have been touted as a therapeutic for inflammatory conditions of the bowel. Here, we study the impact of acetate, propionate, and butyrate, the three most abundant SCFAs in the intestine, on gene expression in the intestinal pathobiont adherent-invasive Escherichia coli. We pair this with adherence, invasion, and inflammation in Caco-2 and human intestinal enteroid (HIE)-derived mo

Murine Models for Staphylococcal Infection.

Staphylococcus aureus is a Gram-positive bacterium that colonizes almost every organ in humans and mice and is a leading cause of diseases worldwide. S. aureus infections can be challenging to treat due to widespread antibiotic resistance and their ability to cause tissue damage. The primary modes of transmission of S. aureus are via direct contact with a colonized or infected individual or invasive spread from a colonization niche in the same individual. S. aureus can cause a myriad of diseases, including skin and soft tissue infections (SSTIs), osteomyelitis, pneumonia, endocarditis, and sep

A non-invasive in vivo imaging system to study dissemination of bioluminescent Yersinia pestis CO92 in a mouse model of pneumonic plague.

The gold standard in microbiology for monitoring bacterial dissemination in infected animals has always been viable plate counts. This method, despite being quantitative, requires sacrificing the infected animals. Recently, however, an alternative method of in vivo imaging of bioluminescent bacteria (IVIBB) for monitoring microbial dissemination within the host has been employed. Yersinia pestis is a Gram-negative bacterium capable of causing bubonic, septicemic, and pneumonic plague. In this study, we compared the conventional counting of bacterial colony forming units (cfu) in the various in

Characterization of Yersinia pestis Interactions with Human Neutrophils In vitro.

Yersinia pestis is a gram-negative, zoonotic, bacterial pathogen, and the causative agent of plague. The bubonic form of plague occurs subsequent to deposition of bacteria in the skin by the bite of an infected flea. Neutrophils are recruited to the site of infection within the first few hours and interactions between neutrophils and Y. pestis have been demonstrated in vivo. In contrast to macrophages, neutrophils have been considered non-permissive to Y. pestis intracellular survival. Several studies have shown killing of the vast majority of Y. pestis ingested by human neutrophils. However,
