Mitragyna speciosa Korth leaves extracts induced the CYP450 catalyzed aminopyrine-N-demethylase (APND) and UDP-glucuronosyl transferase (UGT) activities in male Sprague-Dawley rat livers.

BACKGROUND: Mitragyna speciosa leaves have been abused by drug addicts as some of the alkaloids (mainly mitragynine) from the plant possess opiate and cocaine-like effects. These bring to its prohibition in Malaysia in 2004 as consumption of M. speciosa leaves has been perceived to lead to the abuse of other drugs such as cannabis and heroin. METHODS: In the current study, the in vitro and in vivo effects of M. speciosa methanolic, aqueous and total alkaloid leaves extracts on drug metabolizing enzymes, namely, cytochrome P450s (CYP450s) and UDP-glucuronosyl transferase (UGT) had been evaluat

Continuous fluorescence anisotropy-based assay of BOCILLIN FL penicillin reaction with penicillin binding protein 3.

We report a simple, rapid, and reproducible fluorescence anisotropy-based method for measuring rate constants for acylation and deacylation of soluble penicillin binding protein (PBP) constructs by compounds in microtiter plates by means of competition with time-dependent acylation by BOCILLIN FL. The method is demonstrated by measuring the acylation rate constants of the PBP3 periplasmic domains from Pseudomonas aeruginosa and Acinetobacter baumannii by BOCILLIN FL, aztreonam, meropenem, and ceftazidime. The new method requires very little protein and can be completed in approximately 1h per

Oxidative tailoring reactions catalyzed by nonheme iron-dependent enzymes: streptorubin B biosynthesis as an example.

Tailoring enzymes catalyze reactions that modify natural product backbone structures before, during, or after their biosynthesis to create a final product with specific biological activities. Such reactions can be catalyzed by a myriad of different enzyme families and are responsible for a wide variety of transformations including regio- and/or stereospecific acylation, alkylation, glycosylation, halogenation, and oxidation. Within a broad group of oxidative tailoring enzymes, there is a rapidly growing family of nonheme iron- and oxygen-dependent enzymes that catalyze a variety of remarkable

Quality control of fungus-specific glucosylceramide in Cryptococcus neoformans by endoglycoceramidase-related protein 1 (EGCrP1).

A fungus-specific glucosylceramide (GlcCer), which contains a unique sphingoid base possessing two double bonds and a methyl substitution, is essential for pathogenicity in fungi. Although the biosynthetic pathway of the GlcCer has been well elucidated, little is known about GlcCer catabolism because a GlcCer-degrading enzyme (glucocerebrosidase) has yet to be identified in fungi. We found a homologue of endoglycoceramidase tentatively designated endoglycoceramidase-related protein 1 (EGCrP1) in several fungal genomic databases. The recombinant EGCrP1 hydrolyzed GlcCer but not other glycosphin

Simultaneous abiotic reduction-biotic oxidation in a microbial-MnO2-catalyzed Fenton-like system.

The possibility of simultaneous activity of superoxide-mediated transformations and heterotrophic aerobic bacterial metabolism was investigated in catalyzed H(2)O(2) propagations (CHP; i.e., modified Fenton's reagent) systems containing Escherichia coli. Two probe compounds were used: glucose for the detection of heterotrophic metabolism of E. coli, and tetrachloromethane (CCl(4)) for the detection of superoxide generated in a MnO(2)-catalyzed CHP system. In the MnO(2)-catalyzed CHP system without bacteria, only CCl(4) loss was observed; in contrast, only glucose degradation occurred E. coli s

Use of microperoxidase-11 to functionalize tin dioxide electrodes for the optical and electrochemical sensing of hydrogen peroxide.

In this paper, we employ microperoxidase MP-11 immobilized on mesoporous SnO(2) electrodes in order to study its peroxidase activity and reaction mechanism. We demonstrate the catalytic redox chemistry of the immobilized MP-11 via direct interfacial electron transfer without the use of electron mediators. By taking advantage from the optical transparency of the SnO(2) electrodes, optical absorbance spectroscopy is used in order to compliment the data information obtained from electrochemical techniques. The catalytic activity of the immobilized MP-11 is found to proceed via the Fenton reaction

Analysis of group I intron splicing in the presence of naturally occurring methylxanthines.

BACKGROUND: Recent advances in the understanding of RNA structure-function, intricate folding and its affinity to bind small molecules have led to the proposal that RNA can be a fastidious target for drug design. The revelation that RNA can act as enzymes as in group I intron and that has been recognized by small molecule ligands targeting the catalytic activity has necessitated our focus on group I intron as target for RNA binders. METHODS: We studied the group I intron splicing of Tetrahymena in the presence of naturally occurring methylxanthines (theophylline, theobromine and caffeine) at

Structural and Functional Insights into Catalytic Efficiency and Stability Trade-Offs Induced by CBM Truncation in a GH5 Endoglucanase from Bacillus subtilis DLG.

Cellulose depolymerization into glucose requires the synergistic action of endoglucanase, exoglucanase, and β-glucosidase. In this study, Bacillus subtilis DLG endoglucanase (BsEGL) and its family 3 carbohydrate-binding module (CBM3)-truncated mutant (BsEGLCD) were heterologously expressed in Escherichia coli BL21(DE3) and characterized. Both enzymes displayed optimal activity at 50 °C and pH 4.5, with BsEGLCD exhibiting superior thermal stability and broader pH stability. Both enzymes maintained over 90% relative activity within 0.5-4.5 M NaCl. Notably, BsEGLCD exhibited superior ethanol tole

Expanding Aldopentose Catalysis of a Novel PQQ-Dependent Glucose Dehydrogenase without Compromising Native d-Glucose Activity.

Lignocellulosic biomass is an abundant renewable resource that is rich in aldohexoses and aldopentoses. Efficient biocatalysts for aldopentose oxidation remain limited, and few enzymes exhibit high catalytic efficiency toward both aldohexoses and aldopentoses. Here, a naturally promiscuous pyrroloquinoline quinone-dependent glucose dehydrogenase (PQQ-GDH) from Pseudomonas fragi NL20W was selected as an engineering template and tailored by semirational design to expand aldopentose oxidation while preserving native d-glucose activity. The V706P/L723 M mutant exhibited markedly enhanced catalytic

Synergistic engineering of Nattokinase terminal and calcium-binding regions for enhanced catalytic performance and Oral stability.

Nattokinase,a potent thrombolytic enzyme, is hindered by low activity and poor thermal stability, limiting its therapeutic and industrial applications. This study tackles these issues using an AI-driven approach to boost nattokinase's catalytic performance, based on the hypothesis of synergistic regulation between the terminal structure and calcium-binding region, we utilized the DeepDDG and MPEK servers to predict mutations in the Q275G mutant, focusing on improving both stability and catalytic activity. Our approach identified the Q275G-T174C (M2) mutant, which successfully mitigated the typ

Two novel β-galactosidases from Aeromonas caviae with potential industrial applications in milk and catalytic mechanism analysis using molecular docking.

Lactose intolerance has been a significant global health concern, as it is caused by the absence of lactase, leading to the inability of the human body to absorb lactose. This study investigated two genes encoding maltose O-acetyltransferase with β-galactosidase activity from Aeromonas caviae to evaluate their potential application value for lactose degradation in bovine and human milk. The two novel β-galactosidases (AcGal25: 22.0 kDa and AcGal31: 21.3 kDa) were heterologously expressed and biochemically characterized. The optimal pH of both enzymes was 8.0, and the optimal temperature of AcG

Unveiling six novel CALB-like lipases using genome-centric and patent-driven prospection.

Lipases present biotechnological applications in various industrial sectors due to their ability to perform multiple biochemical reactions. However, the high cost sometimes discourages their potential uses, besides the extensive number of patents involving them. One of the most utilized and researched lipases is Candida antarctica lipase B (CALB), known for its versatility, encompassing enantioselectivity, thermostability, and a wide range of substrates. Therefore, finding new CALB-like lipases is an interesting strategy to enable the implementation of biocatalysts, especially if intellectual

Tailor-made novel electrospun polystyrene/poly(d,l-lactide-co-glycolide) for oxidoreductases immobilization: Improvement of catalytic properties under extreme reaction conditions.

Immobilized enzymes find applications in many areas such as pharmacy, medicine, food production and environmental protection. However, protecting these biocatalysts against harsh reaction conditions and retaining their enzymatic activity even after several biocatalytic cycles are major challenges. Properly selected supports and type of surface modifier therefore seem to be crucial for achieving high retention of catalytic activity of immobilized biomolecules. Here we propose production of novel composite electrospun fibers from polystyrene/poly(d,l-lactide-co-glycolide) (PS/PDLG) and its appli

Computational Investigation of the Mechanism of Diels-Alderase PyrI4.

We studied the mechanisms of activation and stereoselectivity of a monofunctional Diels-Alderase (PyrI4)-catalyzed intramolecular Diels-Alder reaction that leads to formation of the key spiro-tetramate moiety in the biosynthesis of the pyrroindomycin family of natural products. Key activation effects of PyrI4 include acid catalysis and an induced-fit mechanism that cooperate with the unique "lid" feature of PyrI4 to stabilize the Diels-Alder transition state. PyrI4 enhances the intrinsic Diels-Alder stereoselectivity of the substrate and leads to stereospecific formation of the product.

Rosette-shaped graphitic carbon nitride acts as a peroxidase mimic in a wide pH range for fluorescence-based determination of glucose with glucose oxidase.

Rosette-shaped graphitic carbon nitride (rosette-GCN) is described as a promising alternative to natural peroxidase for its application to fluorescence-based glucose assays. Rosette-GCN was synthesized via a rapid reaction between melamine and cyanuric acid for 10 min at 35 °C, followed by thermal calcination for 4 h. Importantly, rosette-GCN possesses a peroxidase-like activity, producing intense fluorescence from the oxidation of Amplex UltraRed in the presence of H2O2 over a broad pH-range of, including neutral pH; the peroxidase activity of rosette-GCN was ~ 10-fold higher than that of con
