Molecular characterization of Cytauxzoon brasiliensis in domestic cats (Felis catus) from Rio de Janeiro state, Brazil: hematological findings.

Cytauxzoon spp. are tick-borne haemoprotozoans that infect domestic and wild felids and are of increasing relevance in veterinary medicine and epidemiology of vector-borne pathogens. In Brazil, the genetic diversity of these agents in wild felids has been progressively expanded, culminating in the description of Cytauxzoon brasiliensis. However, the occurrence of this species in domestic cats is unknown. Therefore, the present study aimed to detect and molecularly characterize C. brasiliensis in domestic cats from the state of Rio de Janeiro, Brazil, and to describe the main hematological find

Humans infested with Ixodes ricinus are exposed to a diverse array of tick-borne pathogens in Serbia.

Tick-borne pathogens (TBPs) pose a major threat to human health in Europe and the whole northern hemisphere. Despite a high prevalence of TBPs in Ixodes ricinus ticks, knowledge on the incidence of tick-borne diseases in humans infested by this tick species is limited. This study was conducted in the year 2019 on patients who presented themselves to the Pasteur Institute Novi Sad with tick infestations. Ticks (n = 31) feeding on human (n = 30) and blood samples from the same individuals were collected by physicians and a microfluidic real-time high-throughput PCR system was used to test the ge

Haemoparasitic agents associated with ovine babesiosis: A possible negative interaction between Babesia ovis and Theileria ovis.

Babesiosis, theileriosis, and anaplasmosis are the most common tick-borne diseases in sheep. The majority of anaplasmosis and theileriosis are subclinical; however, babesiosis causes severe infections in small ruminants. Although there are many reports of co-infections with the agents of these diseases, their clinical severity compared with either of the infections alone is unknown. Within the host, interactions between co-infecting species may cause variations in clinical presentation and response to therapy. The aim of this study was to determine the tick-borne agents in sheep located at sit

A rapid isothermal RPA-CRISPR/Cas12a assay for detection of Rickettsia rickettsii.

INTRODUCTION: Rocky Mountain spotted fever (RMSF) resulting from the tick-borne Rickettsia rickettsii infections is a potentially fatal tick-borne disease affecting humans and dogs in the Americas. It is difficult to confirm the diagnosis early in the laboratory on account of low-level and inconsistent rickettsemia in whole blood. METHODS: Here we established a fast, fully isothermal RPA-CRISPR/Cas12a assay that targeted the vitamin uptake transporter (vut) gene of R. rickettsii. Two independent crRNA primer sets (104 bp and 92 bp) were developed to independently amplify the gene target to en

Mosquito-based detection of retroviruses and arboviruses in Senegal: expanding the scope of xenosurveillance.

BACKGROUND: Mosquitoes are well-known vectors for arthropod-borne viruses, yet their role as passive carriers of non-arthropod-borne viruses remains underexplored. Xenosurveillance, a method that utilizes blood-feeding arthropods to sample host and pathogen genetic material, has emerged as a valuable tool in viral ecology. In this study, we investigated the viral landscape of blood-fed mosquitoes from Senegal and report the first detection of Jaagsiekte Sheep Retrovirus (JSRV)-related and Enzootic Nasal Tumor Virus 2 (ENTV-2)-related sequences, alongside endemic arboviruses. Our study aimed to

Assessment of HMGB-1 concentration in tick-borne encephalitis and neuroborreliosis.

BACKGROUND: The aim of this study was to determine the concentration of HMGB-1 (high mobility group box 1) in the serum and cerebrospinal fluid (CSF) of patients suffering from tick-borne encephalitis (TBE) and neuroborreliosis (NB). Focus was placed on HMGB-1 measurement in the CSF or serum in order to establish whether this could help to differentiate between NB and TBE. METHODS: Eighty patients with meningitis and meningoencephalitis were enrolled in the study. The patients were divided into two groups: group I comprised patients with NB (n=40) and group II comprised patients with TBE (n=4

Pseudomonas mendocina native valve infective endocarditis: a case report.

BACKGROUND: Gram-negative microorganisms are uncommon pathogens responsible for infective endocarditis. Pseudomonas mendocina, a Gram-negative water-borne and soil-borne bacterium, was first reported to cause human infection in 1992. Since then, it has rarely been reported as a human pathogen in the literature. We describe the first case of native valve infective endocarditis due to P. mendocina in the USA. CASE PRESENTATION: A 57-year-old white man presented with bilateral large leg ulcers, fever, and marked leukocytosis. His past medical history included gout and chronic alcohol use. P. men

Diagnosis of canine vector-borne diseases in young dogs: a longitudinal study.

Canine vector-borne diseases (CVBDs) pose a diagnostic challenge, particularly when a dog is coinfected with more than one pathogen. The purpose of this study was to generate information about the diagnosis of CVBDs in young dogs following their first exposure to flea, tick, sand fly, louse, and mosquito vectors. From March 2008 to May 2009, 10 purpose-bred young naive beagle dogs and a cohort of 48 mixed-breed dogs living in an area to which CVBD is endemic in southern Italy were monitored using different diagnostic tests (cytology, serology, and PCR). Overall, PCR detected the highest number

Louse- and flea-borne rickettsioses: biological and genomic analyses.

In contrast to 15 or more validated and/or proposed tick-borne spotted fever group species, only three named medically important rickettsial species are associated with insects. These insect-borne rickettsiae are comprised of two highly pathogenic species, Rickettsia prowazekii (the agent of epidemic typhus) and R. typhi (the agent of murine typhus), as well as R. felis, a species with unconfirmed pathogenicity. Rickettsial association with obligate hematophagous insects such as the human body louse (R. prowazekii transmitted by Pediculus h. humanus) and several flea species (R. typhi and R. f

Natural Mosquito-Pathogen Hybrid IgG4 Antibodies in Vector-Borne Diseases: A Hypothesis.

Chronic exposure to antigens may favor the production of IgG4 antibodies over other antibody types. Recent studies have shown that up to a 30% of normal human IgG4 is bi-specific and is able to recognize two antigens of different nature. A requirement for this specificity is the presence of both eliciting antigens in the same time and at the same place where the immune response is induced. During transmission of most vector-borne diseases, the pathogen is delivered to the vertebrate host along with the arthropod saliva during blood feeding and previous studies have shown the existence of IgG4

Molecular detection of vector-borne and hemotropic pathogens in raccoons (Procyon lotor) from a tropical ecotourism area in Costa Rica.

Interactions between wildlife, domestic animals, and humans in ecotourism settings could facilitate the circulation of pathogens with zoonotic potential. Raccoons (Procyon lotor), due to their synanthropic behavior and adaptability, may serve as hosts for several infectious agents at these interfaces. This study aimed to investigate the presence of vector-borne and hemotropic pathogens in free-ranging raccoons inhabiting Manuel Antonio National Park (MANP) and its surrounding communities, a major tourist destination in Costa Rica. Between 2021 and 2022, nineteen raccoons were captured using To

Borrelia miyamotoi FbpA and FbpB Are Immunomodulatory Outer Surface Lipoproteins With Distinct Structures and Functions.

Pathogens that traffic in the blood of their hosts must employ mechanisms to evade the host innate immune system, including the complement cascade. The Lyme disease spirochete, Borreliella burgdorferi, has evolved numerous outer membrane lipoproteins that interact directly with host proteins. Compared to Lyme disease-associated spirochetes, relatively little is known about how an emerging tick-borne spirochetal pathogen, Borrelia miyamotoi, utilizes surface lipoproteins to interact with a human host. B. burgdorferi expresses the multifunctional lipoprotein, BBK32, that inhibits the classical p

First nationwide survey of the seroprevalence of Toxoplasma gondii in wild boars in Japan.

The increase in some wildlife species is of global concern. The utilization of wildlife meat as food and feed represents a potential source of food-borne pathogens; this is particularly a potential concern for the use of wild board as a food source in Japan. Regarding food safety and an animal infectious disease control, however, little is known about the infection level of zoonotic pathogens including Toxoplasma gondii (T. gondii) in wild boar populations in their natural habitats in Japan. A total of 1279 blood samples from 41 prefectures were collected from apparently healthy wild boars dur

Region-specific transcriptomic and functional signatures of mononuclear phagocytes in the epididymis.

In the epididymis, prevention of autoimmune responses against spermatozoa and simultaneous protection against pathogens is important for mal … before they reached the epithelial lining. Cauda MPs may thus confer a stronger protection against blood-borne pathogens compared to proximal regions. By identifying immunoregulatory mechanisms in the epididymis, our study may lead to new therapies for male infertility and epididymitis and identify potential targets for immunocontraception.

Emergence of bovine ehrlichiosis in Belgian cattle herds.

Bovine ehrlichiosis is a tick-borne rickettsial disease caused by Anaplasma phagocytophilum. The disease can also be transmitted to humans. Outbreaks in cattle have been described in many European countries. In Belgium, infections caused by A. phagocytophilum have been reported in humans and dogs; however, this paper details the first report of ehrlichiosis in cattle herds in Belgium. The first case described was in a dairy herd located in eastern Belgium. Clinical signs included hyperthermia, polypnea, and swelling of the limbs. The other case was diagnosed in a second, mixed purpose herd in
