Permeability of bone and cartilage, and stiffness of collagen within cartilage, influence osteochondral fluid transport during cyclic compression: A study in finite elements.

Osteochondral fluid transport likely plays a critical role in joint health and disease, yet the mechanical factors influencing this transport remain incompletely understood. This study established a finite element model of osteochondral fluid transport under cyclic compression, incorporating depth-dependent material properties and osmotic swelling. Using biphasic constitutive models for bone and cartilage, we simulated fluid flux across the osteochondral interface and performed a parametric sensitivity analysis of seven different mechanical properties. Results demonstrate that bone and cartila…

In vivo study on the short-term effect of radiofrequency energy on chondromalacic patellar cartilage and its correlation with calcified cartilage pathology in an equine model.

Chondromalacia can cause joint pain and synovial effusion with the potential for developing into osteoarthritis. Thermal chondroplasty using radiofrequency energy (RFE) has been reported to be superior to mechanical debridement for treating chondromalacia. We compared short-term changes in biomechanical properties of articular cartilage after treatment with monopolar (mRFE) or bipolar RFE (bRFE) or mechanical debridement (MD) on experimentally created grade II chondromalacia patellae. Chondromalacia patellae was created arthroscopically in both patellae of 15 ponies. Ten months after surgery, …

Hidden in Plain Sight: Triticeal Cartilage Calcification Causing Odynophagia.

The triticeal cartilage is a small cartilage found within the thyrohyoid membrane present in approximately 33%-53% of individuals. Rarely, calcification/ossification of this cartilage can occur, leading to symptoms of dysphagia, odynophagia, or globus sensation. Despite its clinical relevance, symptomatic triticeal cartilage calcification remains poorly characterized in the literature, with only isolated case reports and no established diagnostic or management framework. We present a case of a 44-year-old woman who presented with seven months of atypical throat and neck pain and globus sensati…

Procyanidins and Their Therapeutic Potential Against Bone/Cartilage Diseases.

Bone and cartilage diseases impair labor capacity and cause significant economic losses. As a dietary flavonoid, procyanidins have a wide range of sources and possess antioxidant, anti-inflammatory, and anti-tumor properties. They are used to treat various diseases such as tumors, cardiovascular diseases, and Alzheimer's disease. Recent studies have shown that procyanidins inhibit osteoclast activation, ameliorate the inflammatory environment, protect cartilage from damage by inflammatory factors, and reduce bone loss. Larger polymer procyanidins extracted from plants are oligomers that cannot…

Perinatal outcomes in 35 children with cartilage hair hypoplasia.

BACKGROUND: Cartilage-hair hypoplasia (CHH), an autosomal recessive skeletal dysplasia due to RMRP mutations, is characterized by short stature, immunodeficiency, anemia, and increased malignancies. Growth failure has its onset prenatally. Birth and neonatal care in pregnancies with fetal skeletal dysplasia have been studied in some other skeletal dysplasias but there have been no previous studies on these aspects in CHH. In this retrospective cohort study, we reviewed patient records for 35 Finnish children with CHH born in 2000–2023 to evaluate the course and management of pregnancies and de…

Prenatal Diagnosis of Cartilage-Hair Hypoplasia: A Narrative Review.

Cartilage-hair hypoplasia is a rare autosomal recessive skeletal dysplasia. It is particularly prevalent in the Finnish and Amish populations but increasing reports have been documented worldwide. It is caused by pathogenic variants in the RMRP gene. The clinical presentation is highly variable and may include short-limbed short stature, metaphyseal abnormalities, hypotrichosis, and immune deficiency, among other features. Some of the manifestations may present early in the prenatal period and ultrasound assessment is often the tool that raises suspicion for this condition. This review aims to…

Biological Reinforced Concrete for Cartilage Repair With 3D Printing.

The development of biomimetic cartilage constructs (BCCs) with natural extracellular matrix (ECM) microenvironments and topological cues to accelerate the reconstruction of natural articular cartilage (NAC) after injury is challenging due to its complex structure, low cellular content, and less vascularity. Inspired by concrete rebar structure, a biomimetic cartilage named "biological reinforced concrete" is fabricated, with collagen fiber orientation transitioning from parallel to perpendicular, replicating the ECM microenvironments and complex construct of NAC. 3D-printed ultrafine fiber net…

ER O-glycosylation in synovial fibroblasts drives cartilage degradation.

How arthritic synovial fibroblasts (SFs) activate cartilage ECM degradation remains unclear. GALNT enzymes initiate O-glycosylation in the Golgi; when relocated to the ER, their activity stimulates ECM degradation. Here, we show that in human rheumatoid and osteoarthritic synovial SFs, GALNTs are relocated to the ER. In an RA mouse model, GALNTs relocation occurs shortly before arthritis symptoms and abates as the animal recovers. An ER GALNTs inhibitor prevents cartilage ECM degradation in vitro and expression of this chimeric protein in SFs results in the protection of cartilage. One of the …

Inhibition of 15-hydroxy prostaglandin dehydrogenase promotes cartilage regeneration.

Aging or injury to the joints can lead to cartilage degeneration and osteoarthritis (OA), for which there are limited effective treatments. We found that expression of 15-hydroxy prostaglandin dehydrogenase (15-PGDH) is increased in the articular cartilage of aged or injured mice. Both systemic and local inhibition of 15-PGDH with a small-molecule inhibitor (PGDHi) led to regeneration of articular cartilage and reduction in OA-associated pain. Using single-cell RNA-sequencing and multiplexed immunofluorescence imaging of cartilage, we identified the major chondrocyte subpopulations. Inhibition…
