Data-independent fluorescence molecular imaging analysis: a two-stage clustering framework for intraoperative tumor imaging.

BACKGROUND: Intraoperative fluorescence molecular imaging (FMI) is increasingly used to distinguish benign from malignant tumors; however, existing quantitative methods are predominantly data-driven, requiring large datasets and exhibiting limited adaptability in rare diseases or low-incidence tumor surgeries. Furthermore, high false-positive rates and substantial inter-operator variability remain significant challenges, hindering further application of FMI-guided surgery. This study introduces a novel, self-referenced clustering framework that overcomes these limitations by leveraging interna

Transmission patterns of pertussis in school-based clustered outbreaks: A retrospective analysis in Zhejiang Province, China, 2024.

We conducted a retrospective study using data from Zhejiang Province's pertussis surveillance system between February 1 and May 1, 2024, defining school-based clusters as ≥5 cases occurring within 21 d in the same class. We identified 124 outbreaks involving 825 confirmed cases, with cluster sizes ranging from 5 to 16 cases (median: 7). Most outbreaks occurred in primary schools (79.03%, 98/124), followed by kindergartens (19.35%, 24/124) and middle schools (1.61%, 2/124). Among cases, 19.3% (159/825) were index cases (predominantly aged 7-8 y) and 80.7% (666/825) were secondary infections. Th

Label-Free Clustering Analysis Platform Drives Cascaded Workflow for Scalable Production of Therapeutic Extracellular Vesicles.

Extracellular vesicles (EVs) have emerged as highly promising natural nanomedicines and nanocarriers, holding transformative potential for the treatment of various diseases. However, the lack of rapid and comprehensive characterization techniques for EV preparation analysis, coupled with the absence of efficient quality control methods, significantly hinders process optimization and large-scale production. To address these challenges, we developed a label-free clustering analysis (LFCA) platform that integrates nano-flow cytometry for particle size distribution analysis with a clustering algor

Identifying distinct clinical phenotypes and outcomes in adult-onset IgA vasculitis using unsupervised clustering analysis.

OBJECTIVE: IgA vasculitis (IgAV) is a small-vessel vasculitis. The diversity of clinical presentations and outcomes requires the identification of groups with a distinct phenotype for management and follow-up. METHODS: The French retrospective IGAVAS database (n = 260) includes adult-onset IgAV. Agglomerative hierarchical clustering was performed, using k-means and Ward's method, based on 7 parameters (sex, age, constitutional symptoms, skin necrosis, joint and digestive involvement, abnormal kidney function). These results were validated on an independent cohort of Slovenian patients (n = 20

Analysis of the genetic and phylogenetic context of Escherichia coli O77g:H18 associated with clustered cases of HUS in France in 2025.

In January 2025, Santé publique France detected several cases of hemolytic uremic syndrome (HUS) in adults caused by a rare serotype of Shiga toxin-producing Escherichia coli, STEC O77g:H18. Epidemiological and traceability investigations linked the consumption of raw-milk cheese to these cases. The outbreak strain belonged to sequence type 69 (ST69) and harbored a Shiga toxin (stx) subtype stx2d located on a unique prophage. Putative virulence genes cdt-VABC, hlyE, fimH, and lpfA were detected; however, STEC virulence genes (eae and aggR) were absent. Phylogenetic analysis of E. coli O77g str

The relationship between the phenotype of long COVID symptoms and one-year psychosocial outcomes: an exploratory clustering analysis.

The impact of Coronavirus Disease 2019 (COVID-19) is not limited to acute symptoms; it also extends to post-infection sequelae, such as long COVID and post-COVID conditions. These conditions are characterized by various symptoms, such as malaise, fatigue, and cognitive dysfunction, and are considered to reflect different underlying pathologies. Using a cluster analysis, we hypothesized that long COVID may have different psychosocial outcomes depending on the phenotype. This study is based on the COVID-19 RECOVERY STUDY II (CORES II) conducted in 20 centers in Japan. CORES II included patients …

Identification of energy metabolism and m6A-related gene clusters in Kashin-Beck disease based on bioinformatics analysis.

Kashin-Beck disease (KBD) is a chronic and debilitating osteoarthropathy predominantly affecting populations in certain endemic regions. Despite extensive research, the underlying mechanisms of KBD remain poorly understood. Here we analyzed gene expression profiles from KBD patients and healthy controls, identifying 16 differentially expressed genes. Key pathways related to oxidative stress, apoptosis, and epigenetic regulation were implicated in KBD's development. Notably, GPX4 upregulation and differential m6A RNA methylation suggest potential therapeutic targets. Additionally, distinct meta

Machine Learning Analysis of Post-Acute COVID Symptoms Identifies Distinct Clusters, Severity Groups, and Trajectories.

Questionnaires that capture patient-reported symptomatology provide low-cost but potentially high-value data for the de novo discovery of disease phenotype, severity, and responsiveness to intervention groupings within an umbrella condition. The availability of comprehensive electronic health records (EHRs) has nonetheless overshadowed the use of questionnaires data for symptom analysis in the context of COVID-19. We analyzed de-identified questionnaires from post-acute COVID-19 cohorts at the University of California, San Francisco (UCSF, n = 669), Icahn School of Medicine at Mount Sinai (ISM
