BACKGROUND: Beginning at birth, the microbes in the gut perform essential duties related to the digestion and metabolism of food, the development and activation of the immune system, and the production of neurotransmitters that affect behavior and cognitive function.
OBJECTIVES: The objectives of this review are to (a) provide a brief overview of the microbiome and the "microbiome-gut-brain axis"; (b) discuss factors known to affect the composition of the infant microbiome: mode of delivery, antibiotic exposure, and infant-feeding patterns; and (c) present research priorities for nursing scie

The majority of the enteric nervous system is formed by vagal neural crest cells which enter the foregut and migrate rostrocaudally to colonise the entire length of the gastrointestinal tract. Absence of enteric ganglia from the distal colon are the hallmark of Hirschsprung disease, a congenital disorder characterised by severe intestinal dysmotility. Mutations in the receptor tyrosine kinase RET have been identified in approximately 50% of familial cases of Hirschsprung disease but the cellular processes misregulated in this condition remain unclear. By lineage tracing neural crest cells in m

Regular exercise as part of one's lifestyle is well-recognized for its beneficial effect on several diseases such as cardiovascular disease and obesity; however, many questions remain unanswered regarding the effects of exercise on the gut environment. This study aimed to investigate the impact of long-term endurance exercise on modulating inflammation and endoplasmic reticulum (ER) stress. Fifteen-week-old male Sprague-Dawley (SD) rats were subjected to six months of endurance treadmill training, while age-matched controls remained sedentary. Results showed that IL-6 mRNA levels in colon tiss

Campylobacter jejuni is a leading cause of diarrheal disease in humans and an intestinal commensal in poultry and other agriculturally important animals. These zoonotic infections result in significant amounts of C. jejuni present in the food supply to contribute to disease in humans. We previously found that a transposon insertion in Cjj81176_1038, encoding a homolog of the Escherichia coli LivJ periplasmic binding protein of the leucine, isoleucine, and valine (LIV) branched-chain amino acid transport system, reduced the commensal colonization capacity of C. jejuni 81-176 in chicks. Inflammatory bowel disease (IBD) is characterized by gut dysbiosis and impaired microbial metabolite signaling. Emerging evidence suggests that gut microbial metabolites, such as trace amines (tryptamine, phenethylamine, and tyramine), function as endogenous TAAR1 agonists and may contribute to IBD pathogenesis. Our study identified elevated fecal trace amine levels in ulcerative colitis (UC) patients and DSS-induced colitis mice. In vitro, exposure to these trace amines enhanced 5-HT secretion in QGP-1 cells and ex vivo mouse colonic tissues, and this effect could be blocked by the TAAR1 anta

Perturbations of the colonic microbiota can contribute to the initiation and progression of colorectal cancer, leading to an increase in pathogenic bacteria at the expense of protective bacteria. This can contribute to disease through increasing carcinogenic metabolite/toxin production, inducing inflammation, and activating oncogenic signaling. To limit disease progression, external factors that may influence the colonic microbiota need to be considered in patients with colorectal cancer. One major factor that can influence the colonic microbiota is iron. Iron is an essential micronutrient tha

Inflammatory bowel disease (IBD) is driven by a breakdown in immune regulation and epithelial barrier function, yet the contribution of eosinophils to this process has remained poorly defined and controversial. While eosinophils infiltrate the intestinal mucosa during both flares and remission, their role in shaping disease outcomes is unclear. Our RNA-seq analyses of colonic eosinophils isolated from dextran sulfate sodium (DSS)-treated mice revealed a significant upregulation of cyclooxygenase (COX)-2 (gene name, Ptgs2). Eosinophil-specific deletion of COX-2 (Ptgs2fl/fleoCre+/-) reduced IL-2

PURPOSE: Surgical options in metachronous or recurrent rectal cancer after anterior or low anterior resection are limited and frequently result in abdominoperineal rectal extirpation sacrificing the sphincter or in straight coloanal reconstruction. Decreased capacity and distensibility in straight coloanal reconstruction after proctectomy correlate well with increased daily stool frequency, urgency, and incontinence. A new technique for coloanal pouch reconstruction using the ileocecal segment is proposed.
METHODS: A pedunculated ileocecal segment was rotated 180 degrees counterclockwise and

Despite their relatedness, Salmonella and Shigella enteropathogens differ in infectious dose, pathogenesis, and disease kinetics. The prototype strains Salmonella enterica serovar Typhimurium (Salmonella) and Shigella flexneri (Shigella) use Type-3-secretion-systems (T3SSs) to colonize intestinal epithelial cells (IECs). However, they have evolved unique sets of T3SS effectors and accessory virulence factors. A synthesis of how these differences impact the temporal progression of infection in non-transformed human epithelia is missing. Here, we followed Salmonella and Shigella infections of hu

INTRODUCTION: Adenosquamous carcinoma (ASC) of the colon is an exceedingly rare and aggressive subtype, with limited data available on long-term survival, particularly among patients with ulcerative colitis (UC).
CASE PRESENTATION: We present a 39-year-old male with a 9-year history of UC who experienced severe anemia and hematochezia. Imaging and histopathological analysis confirmed a diagnosis of stage IIIB ASC of the right colon. The patient underwent total colectomy followed by adjuvant chemotherapy (XELOX), ileoanal pouch reconstruction, and stoma closure. After 56 months of follow-up, h

The purpose of this study was to explore the protective effects of selenium containing soybean peptides (SePPs) on inflammatory bowel disease in colitis mice. During the experimental period, the mice were administered with SePPs for 14 days, and then treated with drinking water containing 2.5% dextran sodium sulfate (DSS) for 9 days, while the intervention of SePPs was continued. The results showed that low-dose SePPs (15 μg Se per kg per d bw) could effectively alleviate DSS-induced inflammatory bowel disease through the improvement of the antioxidant levels, reduction of inflammatory factor

BACKGROUND: Intestinal barrier dysfunction plays a central role in the pathological onset of Crohn's disease. We identify the cadherin superfamily member protocadherin 20 (PCDH20) as a crucial factor in Crohn's disease. Here we describe the function of PCDH20 and its mechanisms in gut homeostasis, barrier integrity, and Crohn's disease development.
RESULTS: PCDH20 mRNA and protein expression is significantly downregulated in the colonic epithelium of Crohn's disease patients and mice with induced colitis compared with controls. In mice, intestinal-specific Pcdh20 knockout causes defects in en

How the microbiota affects health and disease is a crucial question. In mice, gut Clostridium bacteria are potent inducers of colonic interleukin (IL)-10-producing Foxp3 regulatory T cells (Treg), which play key roles in the prevention of colitis and in systemic immunity. In humans, although gut microbiota dysbiosis is associated with immune disorders, the underlying mechanism remains unknown. In contrast with mice, the contribution of Foxp3 Treg in colitis prevention has been questioned, suggesting that other compensatory regulatory cells or mechanisms may exist. Here we addressed the regulat

BACKGROUND: Inflammatory bowel disease (IBD) has a remitting and relapsing disease course; however, relatively little is understood regarding how inflammatory damage in acute colitis influences the microbiota, epithelial barrier, and immune function in subsequent colitis.
METHODS: Mice were administered trinitrobenzene sulphonic acid (TNBS) via enema, and inflammation was assessed 2 days (d2) or 28 days (d28) later. Colitis was reactivated in some mice by re-treating at 28 days with TNBS and assessing 2 days later (d30). Epithelial responsiveness to secretagogues, microbiota composition, colo

G protein-coupled receptors (GPRs), including pro-inflammatory GPR4 and ovarian cancer GPR1 (OGR1/GPR68), are involved in the pH sensing of the extracellular space and have been implicated in inflammatory bowel disease (IBD). Previous data show that a loss of GPR4 or OGR1 independently is associated with reduced intestinal inflammation in mouse models of experimental colitis. In the present manuscript, we investigated the impact of the simultaneous loss of GPR4 and OGR1 in animal models of IBD. To study the effects of combined loss of Gpr4 Ogr1 in IBD we used the well-established acute dextran

BACKGROUND: Serum Amyloid A (SAA) is a major acute phase protein of unknown function. SAA is mostly expressed in the liver, but also in other tissues including the intestinal epithelium. SAA reportedly has anti-bacterial effects, and because inflammatory bowel diseases (IBD) result from a breakdown in homeostatic interactions between intestinal epithelia and bacteria, we hypothesized that SAA is protective during experimental colitis.
METHODS: Intestinal SAA expression was measured in mouse and human samples. Dextran sodium sulfate (DSS) colitis was induced in SAA 1/2 double knockout (DKO) mi

INTRODUCTION: Minimal invasive surgical approach can achieve quick functional recovery. However, the oncologic outcome for cancer is still a concern. This study aims to compare the oncologic outcome between laparoscopic and open methods in the curative resection of Stage II or III left-sided colon cancers.
METHODS: In consideration of statistical power up to 90%, 286 eligible patients with curable left-sided colon cancer (Tumor-Node-Metastasis Stage II and Stage III disease) requiring the takedown of colonic splenic flexure to facilitate a curative left hemicolectomy were recruited randomly a

Microbial taxa in the soil microbiome with specific ecological functions play crucial roles in plant health. Flavobacterium is found closely associated with physiological functions that promote plant growth and enhance disease resistance. While cooperative interactions mediated by metabolite exchange can stabilize microbial consortia, the specific interactions between these consortia and plants remain understudied, particularly regarding their application in plant disease control. Here, we isolate a natural microbial consortium comprising Achromobacter, Flavobacterium, and Bacillus strains fro

We present the case of a 43-year-old woman with complete esophageal and gastric stenosis after caustic ingestion. Following initial emergency surgery for gastric perforation, she remained dependent on a jejunostomy for over a year, developing severe malnutrition, dysphagia, and psychological decline. Definitive reconstruction was achieved with a one-stage subtotal esophagectomy and total gastrectomy. A right colon interposition graft, routed through the posterior mediastinum, was used after confirming adequate perfusion despite the absence of the right colic artery. The procedure included a ce
