Spatial autophagy failure as a pharmacodynamic endpoint for autophagy-targeted dermatological interventions.

Autophagy-modulating dermatological interventions include topical, intralesional and systemic therapies, defined bioactive molecules and nutraceutical candidates. Their effects are commonly evaluated using tissue-averaged LC3-II, p62/SQSTM1 and canonical pathway markers. Although these measures support assessment of autophagy pathway engagement, they may miss spatially restricted pharmacodynamic non-response and cannot determine whether persistent local dysfunction reflects inadequate exposure or biology-limited non-response. Here we propose spatial autophagy failure (SAF) as a spatial pharmac

Social media data and its potential for pharmacovigilance: a comparative analysis of reported prevalences regarding drug-induced gingival overgrowth (DIGO).

Drug-induced gingival overgrowth (DIGO) is an increasingly relevant topic among dentists, physicians and patients as well. Due to high prescription rates of calcium channel blockers, anticonvulsants and immunosuppressants, the prevalences of this adverse effect should not be underestimated. To identify differences in the prevalence rates presented, but also possible similarities in the available data, we compared information from package leaflets, social media platforms, current literature and summaries of product characteristics. To obtain a comprehensive overview of the current situation reg

Phenotype-Based Selection of Cyclosporine A Formulations in Dry Eye Disease: Integration of Molecular, Rheological, and Clinical Outcomes.

PURPOSE: Cyclosporine A (CsA) is widely used for dry eye disease (DED), yet therapeutic outcomes vary substantially across available formulations. Growing evidence suggests that formulation properties, rather than CsA concentration alone, determine ocular surface bioavailability and clinical response. This study compared the molecular, rheological, and clinical characteristics of CsA formulations to establish a phenotype-based therapeutic selection strategy for DED management. METHODS: Human conjunctival epithelial cells under hyperosmotic stress were treated with 0.05% CsA nanoemulsion, 0.1%

GutMove: a new educational game for active learning and assessment of competencies on gastrointestinal motility.

We described our first-ever gastrointestinal (GI) motility game (GutMove), designed to enhance teaching and assessment of knowledge on motility across the GI tract. The game has seven sections: 1 (pregastric regions), 2 (upper stomach), 3 (lower stomach), 4 (small intestine and its transition), 5 (large intestine), 6 (stimulating hormones), and 7 (inhibiting hormones). Sections 1-4 have 8 cells each on the left side, and 12 e-tokens each scattered on the right side, although only 8 e-tokens are correctly matchable to the corresponding left cells, while the remaining 4 e-tokens are distractors

Stiffening of soft silicone upon deep UV treatment as characterized using nanoindentation.

Silicones are elastomers that have a wide variety of uses, including biomedical applications such as the coating of biomedical devices and as implants. Soft silicones with mechanical properties similar to those of biological tissues have particularly gained use as substrates for cell culture in mechanobiology studies. In this context, it would be desirable to be able to alter their surface mechanical properties with a relatively simple physical treatment. While deep ultraviolet (deep UV) or ultraviolet C (UV-C) treatment has been previously used as a surface treatment method for stiffer silico

Homogeneous catalytic hydrogenation of CO2 - amino acid-based capture and utilization.

In this review, we provide an overview of research efforts to integrate carbon dioxide capture specifically using amino acid-based sorbents with its thermocatalytic hydrogenation promoted by homogeneous metal complexes. Carbon capture and utilization (CCU) is a promising strategy for the production of fuels, chemicals and materials using CO2 scrubbed from point sources and the atmosphere as a C1 feedstock while mitigating CO2 emissions. Compared to established (alkanol)amines, amino acids offer some advantages as CO2 capture agents due to their lower volatility, higher oxygen stability and low

[Shortfalls of new data 2017-2020 in the German transplant registry].

INTRODUCTION: Recently, considerable deficiencies of legacy data (2006–2016) of the newly established German Transplant Registry were described. Current data (2017–2020) of the registry are subject to the present assessment. MATERIAL AND METHODS: The registry is divided in 80 elements containing different topics. From those elements the required dataset needs to be extracted by the investigator. We analyzed data on heart, liver, lung and kidney transplantation, focusing on the comparison of data published in the Eurotransplant (ET) statistics library with registry data. Moreover, data provide

Proposal for a Severity Score (DeASI, Dermatophytosis Area and Severity Index) for the Evaluation of Dermatophyte Infections - A Delphi Consensus Study.

BACKGROUND: There has been a significant increase in the incidence of recurrent, resistant, and extensive dermatophyte infections worldwide recently. This menace has spurred the need for more well-designed randomized controlled trials to optimize the treatment of dermatophyte skin infections. One of the limitations in designing such studies is the limited availability of standard and validated score, to measure the severity of dermatophyte infections. AIMS: To create a severity score for the evaluation of dermatophyte infections. MATERIALS AND METHODS: A Delphi consensus model was used to fr

Sarcopenic obesity and health outcomes: An umbrella review of systematic reviews with meta-analysis.

Many studies support the idea that sarcopenic obesity (SO) could be considered a potential risk factor for negative health outcomes. These results have been inconsistent, and no umbrella reviews exist regarding this topic. Several databases until November 2023 were searched for systematic reviews with meta-analysis of observational studies (cross-sectional, case-control and prospective). For each association, random-effects summary effect sizes with correspondent 95% confidence intervals (CIs) were evaluated using the GRADE tool. Among the 213 papers initially screened, nine systematic reviews

Variability in surgical approaches to hernias in patients with ascites.

BACKGROUND: Hernias in patients with ascites are common, however we know very little about the surgical repair of hernias within this population. The study of these repairs has largely remained limited to single center and case studies, lacking a population-based study on the topic. STUDY DESIGN: The Michigan Surgical Quality Collaborative and its corresponding Core Optimization Hernia Registry (MSQC-COHR) which captures specific patient, hernia, and operative characteristics at a population level within the state was used to conduct a retrospective review of patients with ascites undergoing

PROMETHAZINE-INDUCED THROMBOPHLEBITIS IN A NIGERIAN MAN: A CASE REPORT.

INTRODUCTION: Promethazine is a phenothiazine derivative that possesses antihistamine, anti-dopaminergic and anticholinergic properties. It is commonly used to treat motion sickness, allergic conditions, nausea and vomiting, in addition to its use as a sedative. Promethazine has vesicant properties and is highly caustic to the intima of blood vessels and surrounding tissues. Intravenous administration may result in thrombophlebitis, unintentional intra-arterial administration, perivascular extravasation and tissue necrosis. To the best of our knowledge there is no previous published report of

Promoted Abutment-Soft Tissue Integration Around Self-Glazed Zirconia Surfaces with Nanotopography Fabricated by Additive 3D Gel Deposition.

INTRODUCTION: Improving the biological sealing around dental abutments could promote the long-term success of implants. Although titanium abutments have a wide range of clinical applications, they incur esthetic risks due to their color, especially in the esthetic zone. Currently, zirconia has been applied as an esthetic alternative material for implant abutments; however, zirconia is purported to be an inert biomaterial. How to improve the biological activities of zirconia has thus become a popular research topic. In this study, we presented a novel self-glazed zirconia (SZ) surface with nano

Can Serum Drops Containing Doxycycline Provide a Supplemental Anti-Bacterial Effect in the Treatment of Bacterial Keratitis?

PURPOSE: Systemic doxycycline has been prescribed to reduce inflammation and enhance corneal healing in bacterial keratitis. Topical autologous serum drops (ASD) containing doxycycline following oral supplementation may additionally confer an anti-bacterial effect. The potential of this supplementation was evaluated by determining the in vitro susceptibility of bacterial keratitis isolates to doxycycline. METHODS: The minimum inhibitory concentrations (MICs) of doxycycline against 100 bacterial keratitis isolates were determined using Etests. Twenty-seven Staphylococcus aureus, ten coagulase-

Effect of Intraoperative Neuromonitoring on the Risks of Recurrent Laryngeal Nerve Injury During Thyroidectomy: A Doubly Robust Approach.

OBJECTIVE: To evaluate the relationship between the use intraoperative neuromonitoring (IONM) during thyroidectomy and the risk of recurrent laryngeal nerve (RLN) injury. BACKGROUND: The role of IONM in reducing RLN injury during thyroidectomy remains controversial. Several studies on this topic apply conventional multivariable regression to adjust for confounding. However, estimates from this method may be biased due to model misspecification, especially with a rare outcome such as RLN injury. METHODS: We used a pooled dataset created by linking the 2016-2019 National Surgical Quality Impro

Drivers of zoonotic disease risk in the Indian subcontinent: A scoping review.

Literature on potential anthropogenic drivers of zoonotic disease risk in the Indian subcontinent is sparse. We conducted a scoping review to identify primary sources, published 2000-2020, to clarify what research exists and on which areas future research should focus. We summarised findings thematically by disease. Of 80 sources included, 78 (98%) were original research articles and two were conference abstracts. Study designs and methods were not always clearly described, but 74 (93%) were quantitative (including one randomised trial), five (6%) were mixed-methods, and one was qualitative. M
