A simple in vitro assay for assessing the efficacy, mechanisms and kinetics of anti-prion fibril compounds.

Prion diseases are caused by the conversion of normal cellular prion proteins (PrP) into lethal prion aggregates. These prion aggregates are composed of proteinase K (PK) resistant fibrils and comparatively PK-sensitive oligomers. Currently there are no anti-prion pharmaceuticals available to treat or prevent prion disease. Methods of discovering anti-prion molecules rely primarily on relatively complex cell-based, tissue slice or animal-model assays that measure the effects of small molecules on the formation of PK-resistant prion fibrils. These assays are difficult to perform and do not dete

Natural product-inspired esters and amides of ferulic and caffeic acid as dual inhibitors of HIV-1 reverse transcriptase.

Using an HIV-1 Reverse Transcriptase (RT)-associated RNase H inhibition assay as lead, bioguided fractionation of the dichloromethane extract of the Ocimum sanctum leaves led to the isolation of five triterpenes (1-5) along with three 3-methoxy-4-hydroxy phenyl derivatives (6-8). The structure of this isolates were determined by 1D and 2D NMR experiments as well as ESI-MS. Tetradecyl ferulate (8) showed an interesting RNase H IC50 value of 12.4 μM and due to the synthetic accessibility of this secondary metabolite, a structure-activity relationship study was carried out. A series of esters and

Corn bran fiber valorization via feruloylated xylooligosaccharide production: Evaluation of antioxidant and α-amylase and α-glucosidase inhibitory effects.

Feruloylated xylooligosaccharides (FXOS) were alkali-solubilized from corn bran followed by enzymatic treatments (4 enzyme treatments were compared). The FXOS were ultrafiltrated using molecular weight cut-offs of 1 kDa and 10 kDa. Antioxidant activities, α-amylase and α-glucosidase inhibitory effects were assessed for the different fractions. The lower molecular weight fraction (<1 kDa), enzymatically hydrolyzed for 24 h (FXOS1-24), exhibited the strongest in vitro antioxidant activity, with IC50 values of 0.14 mg/mL (DPPH) and 1.0 mg/mL (ABTS•+). In addition, FXOS1-24 inhibited porcine pancr

Volatile formation by combined cultures of yeast and homofermentative or heterofermentative lactobacilli in plant-based fermented foods.

Despite increasing interest of consumers in plant-based products, sales of the latter have been subtly decreasing mainly due to organoleptic challenges. Fermentation offers a sustainable option for improving flavour production in plant-based products. Yeast and lactobacilli are common food fermenting microbes, but a proper understanding of the interactions between the two microorganisms during flavour formation is lacking. This study explored the impact of a co-cultures of yeast and lactobacilli on aroma formation during fermentation of wheat and pulse-based food systems. Volatile analysis usi

Genome-wide CRISPRi screen and proteomic profiling identify key genes related to ferulic acid's antifungal activity.

Fungal pathogens of plants must overcome host-imposed stressors, including antimicrobial small molecules. Ferulic acid (FA), a plant-derived phenolic compound, induces fungal stress and cell death. To uncover genetic determinants of FA sensitivity, we performed a genome-wide CRISPR interference (CRISPRi) screen in Saccharomyces cerevisiae. We confirmed that FA impairs yeast growth and triggers stress granule marker sequestration, establishing a relevant selection condition. The CRISPRi screen identified 194 genes involved in the FA-induced stress response and 12 whose repression enhanced resis

Interaction character between ferulic acid derivatives and bitter taste receptors regulated the bitterness perception via combined assessment of electronic tongue and optimal protein model.

The study of interaction between phenolic compounds and the bitter receptor is rare. Therefore, the current study employed an electronic tongue for investigating the association of the phenolic structure with its corresponding bitterness suppression. Following the preparation of ferulic acid (FA) derivatives, computational simulation techniques are integrated to analyze the interaction mechanisms between ferulic acid and its derivatives with bitter taste receptors at the biomolecular level, thereby establishing a preliminary structure-activity relationship concerning bitterness suppression. Ferulic Acid Alleviates Traumatic Brain Injury and Gastrointestinal Disorders by Promoting Ghrelin to Regulate the Microbiota-Brain-Gut Axis Inflammation and Pyroptosis.

Traumatic brain injury (TBI) is a severe condition with a high mortality rate, affecting multiple organs, including the gastrointestinal (GI) tract. Ghrelin is a brain-gut peptide that regulates the microbiota-brain-gut axis, facilitating communication between the GI tract and the central nervous system. This study aimed to investigate the role of ferulic acid (FA) in regulating Ghrelin to improve TBI and GI disorders (GID) induced by controlled cortical impact (CCI). This study used CCI as the in vivo TBI model and scratch-induced injury of primary astrocytes as the in vitro TBI model. The ro

Multi-Modal Therapy for Diabetic Ulcers: A Hydrogel Platform Combining Antibacterial, Oxygenating, ROS-Scavenging, and Pro-Healing Functions.

Diabetic ulcers, characterized by excessive accumulation of reactive oxygen species (ROS), local hypoxia, persistent inflammation, and secondary bacterial infection, pose a serious threat to human health. To address these challenges, this study developed an injectable hydrogel based on covalent cross-linking between polyethylene glycol derivatives and ε-polylysine. This hydrogel is further loaded with recombinant Milk fat globule-epidermal growth factor-VIII (rMFG-E8) and ferulic acid (FA)-loaded mesoporous manganese dioxide (HM-FA), forming a "single application, multiple benefits" platform.

Rice Bran Extract Alleviates Inflammation and Promotes Wound Healing in Radiation Dermatitis.

Radiation dermatitis is a common side effect of radiotherapy, affecting up to 95% of cancer patients receiving radiation therapy and often leading to skin damage, inflammation, and ulceration. The pathogenesis of radiation dermatitis involves complex mechanisms, such as the production of reactive oxygen species (ROS) and sustained inflammatory responses. Current treatments, including topical steroids, moisturisers, and non-steroidal anti-inflammatory drugs (NSAIDs), often provide limited efficacy, primarily addressing symptoms rather than the underlying pathophysiological processes. In this st

Ferulic acid alleviates retinal neovascularization by modulating microglia/macrophage polarization through the ROS/NF-κB axis.

Inflammation plays a pivotal role in ischemia-induced retinal neovascularization. Targeting microglia/macrophage-based neuroinflammation presents a promising therapeutic strategy. Ferulic acid (FA), a natural and active ingredient in plants, exerts favorable anti-oxidative and anti-inflammatory activities. In this study, we investigated the inhibitory effect of FA against hypoxia-induced retinal angiogenesis using cultured retinal vascular endothelial cells and an oxygen-induced retinopathy mouse (OIR) model. The immunoregulatory effect of FA on microglia/macrophage polarization was evaluated

MicroRNA-9 mediated the protective effect of ferulic acid on hypoxic-ischemic brain damage in neonatal rats.

Neonatal hypoxic-ischemic brain damage (HIBD) is prone to cognitive and memory impairments, and there is no effective clinical treatment until now. Ferulic acid (FA) is found within members of the genus Angelica, reportedly shows protective effects on neuronal damage. However, the protective effects of FA on HIBD remains unclear. In this study, using the Morris water maze task, we herein found that the impairment of spatial memory formation in adult rats exposed to HIBD was significantly reversed by FA treatment and the administration of LNA-miR-9. The expression of miRNA-9 was detected by RT-

Exogenous and endogenous therapeutic effects of combination Sodium Ferulate and bone marrow stromal cells (BMSCs) treatment enhance neurogenesis after rat focal cerebral ischemia.

Combining bone marrow stromal cells (BMSCs) with pharmacological therapy is an attractive approach for neurological function recovery of stroke. Our previous reports demonstrated that Sodium Ferulate (SF) combined with BMSCs administration could facilitate BMSCs migration into the ischemic brain by up-regulation of stromal cell-derived factor-1 alpha (SDF-1α)/chemokine (CXC motif) receptor-4 axis after stroke. To further investigate whether combination treatment could enhance neurogenesis through exogenous and endogenous therapeutic effects, we established rat permanent middle cerebral artery

In vivo protective effect of ferulic acid against noise-induced hearing loss in the guinea-pig.

Ferulic acid (FA) is a phenolic compound whose neuroprotective activity was extensively studied in vitro. In this study, we provided functional in vivo evidence that FA limits noise-induced hearing loss. Guinea-pigs exposed to acoustic trauma for 1 h exhibited a significant impairment in auditory function; this injury was evident as early as 1 day from noise exposure and persisted over 21 days. Ferulic acid (150 mg/kg i.p. for 4 days) counteracted noise-induced hearing loss at days 1, 3, 7 and 21 from noise exposure. The improvement of auditory function by FA was paralleled by a significant re

Genome-wide analysis of the structural genes regulating defense phenylpropanoid metabolism in Populus.

Salicin-based phenolic glycosides, hydroxycinnamate derivatives and flavonoid-derived condensed tannins comprise up to one-third of Populus leaf dry mass. Genes regulating the abundance and chemical diversity of these substances have not been comprehensively analysed in tree species exhibiting this metabolically demanding level of phenolic metabolism. Here, shikimate-phenylpropanoid pathway genes thought to give rise to these phenolic products were annotated from the Populus genome, their expression assessed by semiquantitative or quantitative reverse transcription polymerase chain reaction (P
