Atomic Structures of Minor Proteins VI and VII in Human Adenovirus.

Human adenoviruses (Ad) are double-stranded DNA (dsDNA) viruses associated with infectious diseases, but they are better known as tools for gene delivery and oncolytic anticancer therapy. Atomic structures of Ad provide the basis for the development of antivirals and for engineering efforts toward more effective applications. Since 2010, atomic models of human Ad5 have been derived independently from photographic film cryo-electron microscopy (cryo-EM) and X-ray crystallography studies, but discrepancies exist concerning the assignment of cement proteins IIIa, VIII, and IX. To clarify these di

Structural transition of the immature poxvirus scaffold from preassembly to the scaffold lattice.

During poxvirus morphogenesis, the external scaffold defines the shape and size of immature virions. However, its structural characterization has been hindered by the pleomorphic nature of authentic virus particles. Here, we present single-particle cryo-electron microscopy and cryo-electron tomography structures of two distinct in vitro assemblies of the vaccinia virus scaffold protein D13 that recapitulate oligomeric states previously observed in situ. These structures reveal a dramatic transition from rod-like oligomers, representing a pre-assembly state, to a fully formed scaffold, triggere

Stress-induced membrane insertion at the β-barrel assembly machinery complex regulates BepA metalloprotease activity.

Proteases must be tightly regulated to prevent uncontrolled degradation, yet the mechanisms ensuring such control remain poorly understood. Members of the widespread M48 metalloprotease family are kept inactive by an autoinhibitory plug that blocks catalytic water activation, but how this plug is released remained elusive. Here, using genetic, biochemical and cryo-EM approaches, we discover the activation mechanism of BepA, a quality-control protease that preserves outer membrane integrity by surveilling the β-barrel assembly machinery (BAM) in Gram-negative bacteria. Our cryo-EM analysis of B

DNA-PK driven nucleosome unwrapping enables NHEJ in chromatin.

DNA double-strand breaks, one of the most cytotoxic forms of DNA damage, are primarily repaired by non-homologous end joining (NHEJ) in human cells. NHEJ is initiated by the Ku70/80 heterodimer (Ku) and DNA-dependent protein kinase catalytic subunit (DNA-PKcs), followed by factors that bridge and ligate DNA ends. Ku and DNA-PKcs require ~28 base pairs of free duplex DNA, which is often unavailable in chromatin with nucleosomes acting as barriers. The role of DNA-PKcs remains unclear, as previous in vitro studies mainly used naked DNA. Here, in vitro ligation assays show that DNA-PKcs promotes

The molecular fidelity of Aβ pathology in 5xFAD and AppNL-FPsen1P117L mice revealed by cryo-EM.

Animal models, particularly mouse models, are indispensable for understanding the pathogenic mechanisms and developing therapeutic and diagnostic drugs for Alzheimer’s diseases (AD). While most AD mouse models aim to mimic amyloid-β (Aβ) pathology in their brains, previous research has demonstrated that not all models succeed in reproducing Aβ fibril structures observed in the brains of AD patients. In this study, we determined cryo-electron microscopy (cryo-EM) structures of Aβ fibrils extracted from the brains of two AD models, 5xFAD and AppNL−FPsen1P117L. The former represents one of the mo

Structural basis for the contribution of latent TGFβ binding protein to TGFβ latency and activation.

Transforming growth factor-β (TGFβ) is a potent cytokine that controls all aspects of cellular behavior. TGFβ is secreted in complex with its prodomain and latent TGFβ-binding protein-1 (LTBP1), forming the large latent complex (LLC), which through interaction with the extracellular matrix enables integrin-mediated activation. Although TGFβ structures are known, the influence of LTBP1 on the structure and activity of TGFβ is unknown. Here, we report the LLC cryo-EM structure comprising the LTBP1 eight-cysteine domain covalently bound to TGFβ, revealing a hydrophobic interface between TGFβ and

Structure of the IgY-FcRY complex and its comparison with IgG-FcRn.

Immunoglobulin Y (IgY), the avian counterpart to mammalian IgG and IgE, is transported across yolk sac membranes via the FcRY receptor, providing passive immunity to embryos. Although functionally analogous to the mammalian FcRn-IgG system, the molecular basis of IgY-FcRY interactions remains unclear. Here, we resolved the cryo-electron microscopy structure of the chicken IgY-FcRY complex at 3.06 Å resolution, revealing a distinctive binding interface. FcRY forms a double-ring architecture similar to other mannose receptor family members including PLA2R and DEC-205, yet its CTLD5 domain adopts

A Highly Sensitive Water-Soluble Donor-Acceptor Dye for Early-Stage Amyloid Aggregation Kinetics.

Protein aggregation into amyloid fibrils underlies numerous human diseases, yet the most widely used fluorescent probe, Thioflavin T (ThT), offers an incomplete picture of the process and fails to detect certain fibril structures. Here, we introduce and characterize the photophysical properties of DANIR-2b(2OH), a water-soluble push-pull dye that overcomes these limitations. It successfully binds early prefibrillar aggregates and small fibrils of the human Islet Amyloid Polypeptide that elude detection by ThT, which we confirm by time-resolved cryo-electron microscopy of aliquots taken during

Structural insights into spectral tuning and retinal exchange in cone visual pigments.

Color vision in catarrhine primates relies on red-, green-, and blue-sensitive cone pigments that share an 11-cis-retinal chromophore but differ in absorption maxima. Red and green pigments arose by recent gene duplication and differ at only a few residues. Here, we report cryo-electron microscopy structures of red and green cone pigments from the cynomolgus macaque (Macaca fascicularis) integrated with low-temperature vibrational spectroscopy and quantum mechanical and molecular mechanical modeling. The red-green spectral shift is dominated by threonine 285, the hydroxyl dipole of which modul

Cryo-EM structures of GPR75 reveal an occluded orthosteric pocket challenging conventional drug discovery paradigms for an anti-obesity target.

The global obesity epidemic, affecting over 650 million adults, demands innovative therapeutics. GPR75 has emerged as a promising anti-obesity target, with genetic evidence linking loss-of-function variants to protection against obesity and type 2 diabetes. However, structural insights have remained elusive due to GPR75's inherent expression and stabilization challenges. Here we present the cryo-EM structures of human GPR75 in apo and Gq-coupled states, achieved through advanced stabilization techniques including NanoBiT and molecular glue approaches. Our structures reveal unique architectural

Structures of Ostα/β reveal a unique fold and bile acid transport mechanism.

Bile acid and steroid hormone homeostasis are critical for human health, with disruptions linked to metabolic and endocrine disorders1,2. The organic solute transporter Ostα/β, essential for bile acid efflux in enterohepatic circulation3, has long defied mechanistic elucidation. Here we present cryogenic electron microscopy structures of human Ostα/β in apo and substrate-bound states at 2.6-3.1 Å resolution, revealing a distinctive membrane protein architecture that defines a new transporter class. Ostα/β forms a symmetric tetramer of heterodimers, with each Ostα subunit showing a new seven-tr

Structures of LPOR-Chlide complexes reveal the structural basis of membrane remodeling and photocatalysis.

Light-dependent protochlorophyllide oxidoreductase (LPOR) is a light-driven enzyme in flowering plants. It is involved in chlorophyll biosynthesis while also reorganizing membrane lipids into the cubic membrane network that supports chloroplast development. However, the structural basis of these two activities and their relationship have remained unclear. Here, cryo-electron microscopy of chlorophyllide-bound LPOR oligomers reveals nine distinct assembly states, including helical filaments, stacked rings and segmented strings of dimers. We find that strings of LPOR dimers reshape lipid bilayer

Cryo-EM structures of heteromeric Kir4.1/5.1 channel suggest mechanisms of inward rectification and channel blockage.

Inwardly rectifying potassium (Kir) channels play key roles in regulating membrane potential and potassium transport through voltage-dependent inhibition by cytoplasmic Mg²⁺ and polyamines. Despite decades of extensive studies, the structural basis for polyamine-mediated inward rectification remains unclear. Here, we present cryo-EM structures of the heteromeric Kir4.1/5.1 channel-which is critical for brain and kidney function and whose dysfunction causes EAST/SeSAME syndrome-in its apo state and in complex with spermine, a channel blocker named VU0134992, and EHop-016, an inhibitor identifie

A plant pathogen effector blocks stepwise assembly of a helper NLR resistosome.

Helper NLRs function as central nodes in plant immune networks. Upon activation, they oligomerize into inflammasome-like resistosomes to initiate immune signaling, yet the dynamics of resistosome assembly remain poorly understood. Here, we show that the virulence effector AVRcap1b from the Irish potato famine pathogen Phytophthora infestans suppresses immune activation by directly engaging oligomerization intermediates of the tomato helper NLR SlNRC3. Cryo-EM structures of SlNRC3 in AVRcap1b-bound and unbound states reveal that AVRcap1b bridges multiple protomers, stabilizing a stalled interme

Bioactive lipid-mediated structural and functional regulation of the essential human potassium channel Kir7.1.

The inwardly rectifying potassium channel Kir7.1 is essential for the physiological function of diverse tissues, including the retinal pigment epithelium and the gestational myometrium. Loss-of-function mutations in KCNJ13, which encodes Kir7.1, or conditional ablation of Kir7.1 in the retinal pigment epithelium, lead to early-onset vision loss. Despite strong genetic evidence supporting Kir7.1 as a therapeutic target, its regulation by endogenous ligands-beyond phosphoinositides-remains poorly understood. Here, we report cryo-electron microscopy structures of human Kir7.1 in multiple function

Anti-malaria antibody engineering broadens recognition motifs and reveals new homotypic interactions that enhance protective breadth.

The monoclonal antibody L9 mediates high-level protection against malaria in children for up to 6 months in Africa. L9 preferentially binds with high affinity to a triplicate of NVDP-minor repeats on the P. falciparum circumsporozoite protein (PfCSP). Here, we sought to improve the affinity of L9 to enhance protection against rare strains with two spatially separated minor repeats or a single minor repeat. Site saturation mutagenesis and yeast display-screening identified a panel of affinity-improved variants. In vivo challenge showed one variant, L9_yd19, to be modestly more potent against a

Engineering the ADDomer Nanoparticle Vaccine Scaffold for Improved Assembly and Enhanced Stability.

Virus-like particles (VLPs) are promising platforms for next-generation vaccines due to their ability to present antigens in highly ordered, repetitive geometries emulating pathogen-associated patterns to elicit potent immune responses. The ADDomer is a synthetic dodecahedral VLP scaffold derived from the penton base protein (PBP) of human adenovirus serotype 3 (Ad3). PBP tolerates insertion of multiple antigenic epitopes in flexible surface-exposed loops, and spontaneously self-assembles in vitro into ADDomer nanoparticles, but faces limitations including incomplete assembly and susceptibilit

Structural characterization of plum pox virus by cryo-electron microscopy.

Plum pox virus (PPV), a significant member of the genus Potyvirus, represents a global agricultural challenge, causing significant economic losses and threatening fruit farming due to its easy transmission to most Prunus species. Here, we present the high-resolution structural characterization of PPV using cryo-electron microscopy (cryo-EM). The reconstructed structure at 2.9 Å reveals a filamentous virion with a helical assembly formed by the coat protein (CP), which encapsidates a single-stranded RNA (ssRNA) genome. The structure of the CP core shows remarkable conservation with other potyvi

Structure and organization of AMPA receptor-TARP complexes in the mammalian cerebellum.

AMPA receptors (AMPARs) are multimodal transducers of glutamatergic signals throughout the brain. Their diversity is exemplified in the cerebellum: At afferent synapses, AMPARs mediate high-frequency excitation, whereas in Bergmann glia (BG) they support calcium transients that modulate synaptic transmission. This spectrum arises from different combinations of core subunits (GluA1-4), auxiliary proteins, and posttranscriptional modifications. Using mass spectrometry, cryo-electron microscopy, and electrophysiology, we characterize major cerebellar AMPARs in pigs: calcium-impermeable GluA2/A4 h

Directed evolution of phages in biofilms enhances Pseudomonas aeruginosa control through improved lipopolysaccharide recognition.

Pseudomonas aeruginosa is a leading cause of chronic lung infections in cystic fibrosis (CF) patients. While bacteriophages hold potential as a treatment for antibiotic-resistant infections, the complex structure and heterogeneity of P. aeruginosa biofilms pose significant challenges to phage therapy. In this study, we investigate the adaptive evolution of the Pbunavirus phage PE1 to biofilms formed by a CF-derived P. aeruginosa isolate. Our findings reveal that biofilm-adapted PE1 mutants exhibit enhanced efficacy in controlling biofilms in vitro under conditions mimicking the CF lung environ
