Gut microbiota-derived indole-3-acetic acid ameliorates necrotizing enterocolitis-induced adolescent-stage cognitive impairment via regulating AHR-mediated neuroinflammation and synaptic structural integrity.

BACKGROUND: Necrotizing enterocolitis (NEC) is the most severe acute gastrointestinal disease in preterm infants and is an independent risk factor for adolescent-stage cognitive deficits, especially persistent cognitive impairment. Accumulating evidence indicates that gut-brain axis dysfunction mediates NEC-related brain injury. However, the precise metabolic mechanism linking gut microbiota to sustained cognitive deficits remains unclear. This study aimed to explore the effects of gut microbiota-derived indole-3-acetic acid (IAA) and its downstream AHR signaling on NEC-induced neuroinflammati

I-FABP protein/mRNA and IL-6 as biomarkers of intestinal barrier dysfunction in neonates with necrotizing enterocolitis and SPF BALB/c mouse models.

OBJECTIVE: Neonatal necrotizing enterocolitis (NEC) is a serious intestinal inflammatory disease. We investigated intestinal fatty acid binding protein (I-FABP), I-FABP mRNA, and interleukin-6 (IL-6) as potential diagnostic biomarkers in NEC. METHODS: Forty mice were subjected to hypoxic-ischemic intestinal injury, and then serum I-FABP protein and mRNA levels were quantified. Ileal tissue pathological scores were determined by hematoxylin and eosin staining. I-FABP expression levels and translocation in these tissues were detected using western blotting and immunofluorescence, respectively.

Prophylactic antenatal N-Acetyl Cysteine administration combined with postnatal administration can decrease mortality and injury markers associated with necrotizing enterocolitis in a rat model.

BACKGROUND: Necrotizing enterocolitis (NEC) is a devastating gastrointestinal disease of neonates, especially premature neonates. To date, there is no prophylactic treatment against NEC, except breast milk and slow increase in enteral feeding, and there is no antenatal prophylaxis. AIMS: To assess possible protective effects of antenatal N-Acetyl Cysteine (NAC) against the intestinal pathophysiological changes associated with NEC in a rat model of NEC and against its associated mortality. METHODS: Newborn Sprague-Dawley rats were divided into 5 groups: control (n = 33); NEC (n = 32)-subjecte

3-Fucosyllactose and 3'-sialyllactose combined with Lacticaseibacillus rhamnosus 1.0320 alleviate necrotizing enterocolitis by modulating the gut microbiota and regulating NF-κB/Nrf2 pathways.

Necrotizing enterocolitis (NEC) is a severe gastrointestinal disease that primarily affects premature infants. In recent years, increasing attention has been paid to the potential roles of human milk oligosaccharides (HMOs) and probiotics in the prevention and treatment of NEC. In this study, a neonatal rat NEC model was established to evaluate the protective effects of Lacticaseibacillus rhamnosus 1.0320 combined with 3-FL and 3'-SL. The results demonstrated that 1.0320FS improved the macroscopic manifestations of NEC, reduced clinical disease scores, and improved histopathological alteration

Validation of a Predictive Nomogram for Mortality Among Neonates With Necrotizing Enterocolitis in Tertiary Care Hospitals at Bahir Dar City, Bahir Dar, Northwest Ethiopia.

BACKGROUND: Necrotizing Enterocolitis (NEC) is a serious gastrointestinal disease primarily affecting preterm neonates. Despite improvements in neonatal care, NEC continues to contribute significantly to neonatal mortality, particularly in low-resource settings. In Ethiopia, NEC-related mortality rates vary from 45% to 89%, reflecting both the severity of the disease and inconsistencies in existing evidence. Moreover, little is known about the predictors of NEC-related mortality in the local context. This underscores the need for a locally derived predictive model to support early risk stratif

Routine contrast enema prior to stoma reversal seems only required following treatment for necrotizing enterocolitis: An evaluation of the diagnostic accuracy of the contrast enema.

INTRODUCTION: Contrast enemas are often made prior to stoma reversal in order to detect distal intestinal strictures distal of the stoma. If untreated these strictures can cause obstruction which might necessitate redo-surgery. However, the value of contrast enemas is unclear. Therefore, we aim to evaluate the contrast enema's diagnostic accuracy in detecting strictures in children with a stoma. METHODS: Young children (≤3 years) treated with a stoma between 1998 and 2018 were retrospectively included. The STARD criteria were followed. Patients treated for anorectal malformations and those th

Antibacterial and anti-inflammatory effects of Lactobacillus reuteri in its biofilm state contribute to its beneficial effects in a rat model of experimental necrotizing enterocolitis.

INTRODUCTION: Necrotizing enterocolitis (NEC) remains a significant surgical emergency in neonates. We have demonstrated the efficacy of Lactobacillus reuteri (Lr) in protecting against experimental NEC when administered as a biofilm by incubation with maltose loaded dextranomer microspheres. Lr possesses antimicrobial and anti-inflammatory properties. We developed mutant strains of Lr to examine the importance of its antimicrobial and anti-inflammatory properties in protecting the intestines from NEC. METHODS: Premature rat pups were exposed to hypoxia/hypothermia/hypertonic feeds to induce

Routine Administration of a Multispecies Probiotic Containing Bifidobacterium and Lactobacillus to Very Low Birth Weight Infants Had No Significant Impact on the Incidence of Necrotizing Enterocolitis.

Background: Necrotizing enterocolitis (NEC) is the leading cause of gastrointestinal morbidity in preterm infants, and prevention and treatment strategies have remained largely unchanged over the past several decades. As understanding of the microbiome has increased, probiotics have been hypothesized as a possible strategy for decreasing rates of NEC, and several studies have noted significant decreases in rates of NEC after initiation of probiotics in preterm infants. However, a recent AAP report cited caution on the use of probiotic use in part because studies of probiotic use in ELBW infant

[Effects of glutamine supplemented parenteral nutrition on the incidence of necrotizing enterocolitis, nosocomial sepsis and length of hospital stay in very low birth weight infants].

INTRODUCTION: Parenteral feeding is the basic way of nutrition in the first day of life in infants with very low birth weight. Due to its instability glutamine is not included in aminoacid solutions used for parenteral nutrition. Meanwhile glutamine is an important aminoacid, which plays a major role in the maturation of the gastrointestinal tract as well as the immunological system. AIM OF THE STUDY: The aim of our study was to estimate if glutamine supplementation of parenteral nutrition in neonates with the very low birth weight can decrease the incidence of necrotizing enterocolitis -- NE

Effect of Probiotic Supplementation in Preventing Necrotizing Enterocolitis Sepsis and Mortality Among Preterm and Very Low Birth Weight Infants: A Systematic Review and Meta-Analysis.

Preterm and very low birth weight (VLBW) infants are highly vulnerable to gastrointestinal and systemic complications due to immature gut barriers, dysbiosis, and underdeveloped immunity, with necrotizing enterocolitis (NEC) and late-onset sepsis contributing significantly to morbidity and mortality. Probiotic supplementation has been proposed as a strategy to restore gut microbial balance, enhance gastrointestinal maturation, and reduce disease risk. This study systematically reviews the impact of probiotics on NEC, sepsis, mortality, feeding tolerance, hospitalization, and related physiologi
