Geographic and genetic diversity in gallbladder cancer mutation profiles: insights from a worldwide exome analysis.

BACKGROUND: Gallbladder cancer (GBC), an aggressive disease with limited treatment options, occurs mainly in low-income and middle-income regions of Asia and Latin America. We analysed whole-exome sequencing data from 262 GBC tumour-normal sample pairs from Chilean, Chinese, Indian, Japanese and South Korean patients to investigate differences in GBC mutation profiles according to geographic location and genetic ancestry. METHODS: We used a unified analysis pipeline to ascertain gene mutations and mutational signatures, estimated the individual proportions of major ancestry subtypes in the in…

Exome Sequencing Resolving a Complex Pediatric Neurodevelopmental Disorder After Inconclusive Prenatal Testing: A Case Report.

BACKGROUND: Prenatal detection of fetal structural anomalies often prompts chromosomal analysis; however, chromosomal microarray analysis (CMA) has limited diagnostic yield for monogenic disorders. Whole-exome sequencing (WES) has emerged as a powerful tool for identifying single-gene etiologies, particularly in cases with complex neurodevelopmental phenotypes. CASE PRESENTATION: We report a female infant presenting with prenatally detected ventriculomegaly and inconclusive chromosomal testing. Prenatal investigations, including karyotyping and genome-wide chromosomal sequencing, identified s…

Whole exome sequencing of sequential, ipsilateral HPV-positive conjunctival, and lacrimal sac squamous cell carcinomas.

A 71-year-old Caucasian female with a history of a right conjunctival intraepithelial neoplasia (CIN) presented with an expanding right lacrimal sac mass. Pathology from the lacrimal sac biopsy revealed invasive, moderately differentiated, non-keratinizing squamous cell carcinoma (SCC) that stained positive for p63 and p40 and negatively for CEA. The conjunctival and lacrimal sac masses were noncontiguous and found to be HPV-positive SCCs after definitive surgery. Whole exome sequencing demonstrated distinct, shared deletions and gains in both tumors, with the lacrimal sac SCC having more gene…
