Glutamate transporter blockade affects Ca(2+) responses in astrocytes.

Brief pretreatment of astrocytes in culture with glutamate (500 microM for 20 min), was earlier shown to significantly enhance the Ca(2+) re … in astrocytes, which may be important in regulating the neurotoxic accumulation of glutamate in the extracellular space and the synapse.

Proteomic profiling of the extracellular matrix in skeletal muscle.

A dynamic extracellular matrix (ECM) surrounds individual cellular units and fills the intercellular space to provide physical support, structural organization and a medium for signaling mechanisms. This article focuses on the ECM in skeletal muscles, which exhibits a diverse and dynamic protein composition in the endomysium, perimysium and epimysium. The muscle matrisome plays a key role in providing tissue integrity during embryonic myogenesis, muscle repair and myofiber regeneration. Mass spectrometry-based proteomics has been instrumental in the systematic cataloguing of the muscle ECM, in

Tracheobronchial amyloidosis following COVID-19 infection.

Deposition of amyloid proteins in extracellular space can occur due to uncontrolled inflammatory processes. Tracheobronchial amyloidosis (TBA) is a rare form of such disease. A 68-year-old woman was evaluated for chronic cough following a COVID-19 infection. Although post-viral sequelae were suspected, further investigation revealed TBA. This condition involves amyloid deposits within the airways. The temporal association with COVID-19 raises a potential etiological link, suggesting viral-induced inflammatory or immune-mediated processes may play a role. This case underlines the need to consid…

Gastric Siderosis Due to Oral Ferrous Sulfate Supplements.

Gastric siderosis is the deposition of excess amount of iron from oral ferrous sulfate supplements to the gastric mucosa. It is an often overlooked entity in the literature and can be related to symptoms such as dyspepsia, nausea, and melena through mucosal injury. Different etiologies of gastric siderosis display distinct histopathological patterns. Pattern B, which is most commonly associated with oral iron supplements, is seen when iron is deposited in the extracellular space of the lamina propria. It is crucial to consider gastric siderosis as a potential diagnosis in symptomatic patients …

In Vivo EPR Assessment of pH, pO2, Redox Status, and Concentrations of Phosphate and Glutathione in the Tumor Microenvironment.

This protocol demonstrates the capability of low-field electron paramagnetic resonance (EPR)-based techniques in combination with functional paramagnetic probes to provide quantitative information on the chemical tumor microenvironment (TME), including pO2, pH, redox status, concentrations of interstitial inorganic phosphate (Pi), and intracellular glutathione (GSH). In particular, an application of a recently developed soluble multifunctional trityl probe provides unsurpassed opportunity for in vivo concurrent measurements of pH, pO2 and Pi in Extracellular space (HOPE probe). The measurement…

How should hypomagnesaemia be investigated and treated?

Hypomagnesaemia is relatively common, with an estimated prevalence in the general population ranging from 2·5% to 15%. It may result from inadequate magnesium intake, increased gastrointestinal or renal loss or redistribution from extracellular to intracellular space. Drug-induced hypomagnesaemia, particularly related to proton pump inhibitor (PPI) therapy, is being increasingly recognized. Most patients with hypomagnesaemia are asymptomatic; symptomatic magnesium depletion is often associated with multiple other biochemical abnormalities, including hypokalaemia, hypocalcaemia and metabolic ac

Kinetics of intravenously administered carnitine in haemodialysed children.

The pharmacokinetics of low-dose bolus L-carnitine (5 mg kg-1 body wt) in five haemodialysed children were investigated. Kinetic variables were obtained by applying a two-compartment open model. The elimination half-life was very short, 2.43 +/- 0.35 h, despite the reduced plasma clearance of 41.2 +/- 5.7 ml min-1, compared with healthy adults. The apparent volume of distribution, 0.27 +/- 0.07 1 kg-1 body wt, corresponds well to the size of the extracellular space. The kinetic behaviour of intravenously supplied carnitine may assist in future evaluations of the therapeutic application of this…

CD9 protects the sperm from cytotoxic factors in the epididymis as extracellular components.

The mechanism by which seemingly normal sperm cause infertility is still under debate. Although CD9 is expressed in male reproductive tissues, its role in male fertility remains unclear. To address this, we investigated the role of CD9 in analyzing Cd9 -deficient ( Cd9 -KO) male mice. The litter size of Cd9 -KO males was comparable, regardless of mating experience. When Cd9 -KO males experienced their first mating chance, a considerable number of neonates died 48 hours after birth. Electron microscopy reveals the presence of CD9 in the epididymal space. Our results suggest that CD9 contributes

HMGB1 and its membrane receptors as therapeutic targets in an intravesical substance P-induced bladder pain syndrome mouse model.

HMGB1, a nuclear protein, once released to the extracellular space, promotes somatic and visceral pain signals. We thus analyzed the role of HMGB1 in an intravesical substance P-induced bladder pain syndrome (BPS) mouse model. Intravesical administration of substance P caused referred hyperalgesia/allodynia in the lower abdomen and hindpaw without producing severe urothelial damage, which was prevented by an anti-HMGB1-neutralizing antibody, thrombomodulin α capable of inactivating HMGB1 and antagonists of RAGE or CXCR4. The HMGB1 inactivation or RAGE blockade also reversed the established bla…

The intracellular and extracellular changes associated with ischaemia--effects of catecholamines in arrhythmogenesis.

Beta sympathetic stimulation and the beta-adrenergic agonists exert a variety of electrophysiologic effects which contribute to the genesis of ischaemia-related arrhythmias. These are both primary, most likely related to enhancement of the slow calcium current, and secondary, resulting from changes in the metabolism of the ischaemic myocardium as reflected in the accumulation of ions and other end products of metabolism in the extracellular space. In addition, the regional influence of these changes may contribute to the ionic and electrical inhomogeneities. Further studies are needed to defin…

Immediately surrounding the lesions were activated fibroblasts and fine fibrillar material in the extracellular space between them. The sclerotic foci often were apposed to normal modiolar bone that never appeared degraded. CONCLUSIONS: The similarities between these bony lesions and known human otic capsule diseases suggests parallel processes are involved. Thus, …

Ozonated water attenuates nucleus pulposus cell apoptosis and matrix degradation by suppressing NF-κB signaling in a rat model of Staphylococcus aureus-induced discitis.

PURPOSE: Discitis is a severe suppurative infection of the intervertebral disc (IVD) posing significant clinical challenges. This study evaluated the therapeutic efficacy of ozonated water (OW) in a rat model of Staphylococcus aureus-induced discitis and elucidated its underlying mechanisms, focusing on NF-κB signaling, extracellular matrix homeostasis, and nucleus pulposus (NP) cell apoptosis. METHODS: A rat tail coccygeal IVD discitis model was established by injecting S. aureus (102 cfu) into the disc space. From day 3 post-infection, animals received transdiscal lavage with normal saline,

A novel collagenase from Vibrio Alginolyticus: experimental study for Dupuytren's disease.

OBJECTIVE: Dupuytren contracture (DC) is a highly prevalent hand affection in which contracted fingers compromise hand function. It is a benign fibroproliferative condition affecting the hand palmar fascia with a deposition of excess matrix proteins in the extracellular space of the palmar aponeurosis. In particular type III over type I collagen V. Alginolyticus collagenase (CVA), is a new enzyme that is fully active on the collagen filaments and inactive on other components of the dermal extracellular matrix. The aim of this study is to evaluate the safety and effectiveness of an intra-lesion…

Ocular P2 receptors and glaucoma.

Adenosine triphosphate (ATP), an energy source currency in cells, is released or leaked to the extracellular space under both physiological and pathological conditions. Extracellular ATP functions as an intercellular signaling molecule through activation of purinergic P2 receptors. Ocular tissue and cells release ATP in response to physiological stimuli such as intraocular pressure (IOP), and P2 receptor activation regulates IOP elevation or reduction. Dysregulated purinergic signaling may cause abnormally elevated IOP, which is one of the major risk factors for glaucoma. Glaucoma, a leading c…

Chronic exposure to aluminium impairs the glutamate-nitric oxide-cyclic GMP pathway in the rat in vivo.

Aluminium is neurotoxic and is considered a possible etiologic factor in Alzheimer's disease, dialysis syndrome and other neurological disor … mulation with S-nitroso-N-acetylpenicillamine was similar to controls. Basal cGMP in the cerebellar extracellular space was decreased by 50% in aluminium-treated rats. These results indicate that chronic exposure to aluminium reduces the basal activity of guanylate cyclase and impairs the glutamate-nitric oxide-cGMP pathway in the animal in vivo.

Trophic effects of purines in neurons and glial cells.

In addition to their well known roles within cells, purine nucleotides such as adenosine 5' triphosphate (ATP) and guanosine 5' triphosphate (GTP), nucleosides such as adenosine and guanosine and bases, such as adenine and guanine and their metabolic products xanthine and hypoxanthine are released into the extracellular space where they act as intercellular signaling molecules. In the nervous system they mediate both immediate effects, such as neurotransmission, and trophic effects which induce changes in cell metabolism, structure and function and therefore have a longer time course. Some tro…

Recombinant human soluble thrombomodulin is associated with attenuation of sepsis-induced renal impairment by inhibition of extracellular histone release.

Multiple organ dysfunction induced by sepsis often involves kidney injury. Extracellular histones released in response to damage-associated molecular patterns are known to facilitate sepsis-induced organ dysfunction. Recombinant human soluble thrombomodulin (rhTM) and its lectin-like domain (D1) exert anti-inflammatory effects and neutralize damage-associated molecular patterns. However, the effects of rhTM and D1 on extracellular histone H3 levels and kidney injury remain poorly understood. Our purpose was to investigate the association between extracellular histone H3 levels and kidney injur
