Clostridioides difficile is an anaerobic, toxin-producing pathogen that colonizes the host gastrointestinal tract. Within this hostile environment, the bacterium encounters stressors such as reactive nitrogen species (RNS), which impose nitrosative stress that must be mitigated for survival. This study aimed to elucidate the molecular mechanisms by which C. difficile defends against nitrosative stress. We screened an unordered transposon mutant library of the epidemic strain R20291 using a nitric oxide (NO) donor and identified a nitrosative stress-sensitive mutant with an inactivated hcpR, a

AIMS: Glycerol dehydratase, encoded by dhaB, has been identified as the rate-limiting enzyme for biosynthesis of 1,3-propanediol (1,3-PDO). This study aimed to enhance the production of 1,3-PDO in Klebsiella pneumoniae by increasing the expression of dhaB.
METHODS AND RESULTS: Plasmid-based and genomic integration systems for increasing the expression of dhaB were compared. The results showed that plasmid-promoted overexpression of dhaB resulted in growth inhibition, and no inhibition of cell growth was observed by overexpression of dhaB through genomic integration. However, overexpression by

Erwinia amylovora, the causative agent of fire blight, poses a threat to pome fruit production worldwide. Disease initiation involves a transition from an epiphytic to an endophytic growth stage, with early activation of the type III secretion system (T3SS) on floral stigmas enabling host tissue invasion. Among the diverse arsenals of E. amylovora, the T3SS and its effectors (T3Es), along with the exopolysaccharide amylovoran, represent core pathogenicity determinants. Systemic colonisation and persistence within the host are orchestrated by a complex regulatory network that coordinates transi

Leptospirosis is a zoonotic disease that affects humans, companion animals, livestock, and wildlife. There are over 60 species that are established pathogens. Leptospires must rapidly adapt to changing environmental conditions as they pass between the environment and vertebrates. Bioinformatic analyses have identified a putative CdaA-type diadenylate cyclase (DAC) in Leptospira interrogans Fiocruz L1-130 (lic10844). DACs catalyze the synthesis of cyclic di-adenosine monophosphate (c-di-AMP) from two ATP molecules. The potential regulatory roles and effector mechanisms of c-di-AMP among pathoge

Environmental metabolites and metabolic pathways significantly influence bacterial pathogenesis and interspecies competition. We previously discovered that a mutation in the triosephosphate isomerase gene, tpiA, in Pseudomonas aeruginosa led to defective type III secretion and increased susceptibility to aminoglycoside antibiotics. In this study, we found that the tpiA mutation enhances the Las quorum sensing system due to reduced translation of the negative regulator RsaL. Further investigations demonstrated an upregulation of CspC, a CspA family protein that represses rsaL translation. DNA p

NAD+ and its derivatives, which act as redox coenzymes, are crucial for cellular metabolism and energy production. Nevertheless, the processes by which Streptococcus mutans, a bacterium known for causing dental caries, synthesizes NAD+ are not well elucidated. Through a genome-wide screen, we identified the nicotinic acid salvage pathway and the evolutionarily incomplete PnuC-NadR pathway involved in NAD+ biosynthesis in S. mutans UA159. The nicotinic acid pathway is regulated by SmNiaR, a nicotinic acid-responsive transcription regulator featuring an N-terminal DNA-binding winged helix-turn-h

Metabolic homeostasis is a central organizing principle of physiology whereby dynamic processes work to maintain a balanced internal state. Highly reactive essential metabolites are ideally maintained at equilibrium to prevent cellular damage. In the facultative methylotrophic bacterium Methylobacterium extorquens PA1, the utilization of one-carbon growth substrates, including methanol, generates formaldehyde as an obligate intermediate. Formaldehyde is highly chemically reactive and capable of damaging various biomolecules, making formaldehyde homeostasis critical during methylotrophic growth

The tier 1 bioterrorism agent Burkholderia pseudomallei causes melioidosis, a tropical disease with fatality rates that can exceed 40% despite antibiotic therapy. Antibiotic failure is likely to be, at least in part, due to biofilm-dwelling B. pseudomallei, and therefore, an improved understanding of how this pathogen regulates biofilm formation could reveal new opportunities for clinical intervention. The antimicrobial radical nitric oxide (NO) plays a key role in host immune defenses against bacteria, and the ability of B. pseudomallei to sense and mitigate NO toxicity is vital for establish

Yersinia pestis is a gram-negative bacterium and the causative agent of plague. The Y. pestis virulence factor plasminogen activator protease (Pla) is an outer membrane aspartic protease that facilitates the dissemination of bacteria from the site of inoculation to deeper tissue during bubonic plague. During pneumonic plague, Pla acts as an adhesin, which contributes to the suppression of early innate immune responses in the lungs, and as a protease that aids in resisting bacterial killing by neutrophils. Two-component regulatory systems (TCSs) are involved in bacterial adaptation to environme

PhoU proteins are negative regulators of the phosphate response, regulate virulence, and contribute to antibiotic resistance. Staphylococcus aureus has multiple genes encoding PhoU homologs that regulate persister formation and potentially virulence, but the molecular mechanisms of this regulation are not fully understood. We used a bacterial adenylate cyclase two-hybrid system to assess interactions between PhoU homologs and other proteins known to interact with PhoU from Escherichia coli. S. aureus PhoU (also referred to as PhoU1) interacted with PhoU itself; PitR (also referred to as PhoU2)

Nucleotide secondary messengers regulate various processes in bacteria allowing them to rapidly respond to changes in environmental conditions. c-di-AMP is an essential second messenger required for the growth of the human pathogen Staphylococcus aureus, regulating potassium, osmolyte uptake, and beta-lactam resistance. Cellular concentrations of c-di-AMP are regulated by the activities of two enzymes, DacA and GdpP, which synthesize and hydrolyze c-di-AMP, respectively. Besides these, only a limited number of other factors are known to regulate c-di-AMP levels. Using a c-di-AMP biosensor cons

UNLABELLED: Rickettsia rickettsii is an obligate intracellular, tick-borne bacterium that causes Rocky Mountain spotted fever. The demanding nature of cultivating these bacteria within host cells and the labor involved in obtaining clonal isolates have severely limited progress regarding the development of compatible genetic tools to study this pathogen. Conditional expression of genes that might be toxic or have an otherwise undesirable effect is the next logical goal to expand upon the constitutive expression plasmids generated thus far. We describe the construction of an inducible promoter

UNLABELLED: Streptococcus pneumoniae is an important human pathogen that normally resides in the human nasopharynx. Competence-mediated bacteriocin expression by S. pneumoniae plays a major role in both the establishment and persistence of colonization on this polymicrobial surface. Over 20 distinct bacteriocin loci have been identified in pneumococcal genomes, but only a small number have been characterized phenotypically. In this work, we demonstrate that three-fourths of S. pneumoniae strains contain a highly conserved scb locus that encodes an active lactococcin 972-like bacteriocin called

Transcription regulation is a critical means by which microorganisms sense and adapt to their environments. Bacteria contain a wide range of highly conserved families of transcription factors that have evolved to regulate diverse sets of genes. It is increasingly apparent that structural similarities between transcription factors do not always equate to analogous transcription regulatory networks. For example, transcription factors within the copper-sensing operon repressor (CsoR)-resistance to cobalt and nickel repressor family have been found to repress a wide range of gene targets, includin

Iron is an essential element required for all organisms. Iron response regulator (Irr) is a crucial transcriptional regulator and can affect the growth and iron uptake of Brucella. The growth rate of Brucella melitensis M5-90 irr mutant was significantly lower than that of B. melitensis M5-90 under normal or iron-sufficient conditions, however, the growth rate of the B. melitensis M5-90 irr mutant was significantly higher than that of B. melitensis M5-90 under iron-limited conditions. In addition, irr mutation significantly reduced iron uptake under iron-limited conditions. Previous studies su

Hallmarks of the developmental cycle of the obligate intracellular pathogenic bacterium Chlamydia are the primary differentiation of the infectious elementary body (EB) into the proliferative reticulate body (RB) and the secondary differentiation of RBs back into EBs. The mechanisms regulating these transitions remain unclear. In this report, we developed an effective novel strategy termed dependence on plasmid-mediated expression (DOPE) that allows for the knockdown of essential genes in Chlamydia. We demonstrate that GrgA, a Chlamydia-specific transcription factor, is essential for the secon

Transcription termination is an essential and dynamic process that can tune gene expression in response to diverse molecular signals. Yet, the genomic positions, molecular mechanisms, and regulatory consequences of termination have only been studied thoroughly in model bacteria. Here, we use several RNA-seq approaches to map RNA ends for the transcriptome of the spirochete Borrelia burgdorferi - the etiological agent of Lyme disease. We identify complex gene arrangements and operons, untranslated regions and small RNAs. We predict intrinsic terminators and experimentally test examples of Rho-d

Vibrio cholerae is a waterborne bacterium that primarily infects the human intestine and causes cholera fatality. Quorum sensing (QS) negatively regulates the expression of V. cholerae virulence gene. However, the primary associated mechanisms remain undetermined. This investigation identified a new QS regulator from the TetR family, LuxT, which increases V. cholerae virulence by directly inhibiting hapR expression. HapR is a master QS regulator that suppresses virulence cascade expression. The expression of luxT increased 4.8-fold in the small intestine of infant mice than in Luria-Bertani br

Heterogeneous environments such as the chronically infected cystic fibrosis lung drive the diversification of Pseudomonas aeruginosa populations into, e.g., mucoid, alginate-overproducing isolates or small-colony variants (SCVs). In this study, we performed extensive genome and transcriptome profiling on a clinical SCV isolate that exhibited high cyclic diguanylate (c-di-GMP) levels and a mucoid phenotype. We observed a delayed, stepwise decrease of the high levels of c-di-GMP as well as alginate gene expression upon passaging the SCV under noninducing, rich medium growth conditions over 7 day
