Neuropeptide Y (NPY) and galanin (GAL), two of the most abundant neuropeptides in the mammalian nervous system, are consistently associated with brain disorders, yet their therapeutic potential remains unrealized. Both are state-dependent modulators that are released under physiological load, acting as adaptive circuit stabilizers. Both have generated expensive drug programs that failed, not because the biology was wrong, but because the development strategy was. This manuscript uses NPY and GAL as paired case studies to argue that inconsistent translation reflects strategy mismatch in target

A four-year-old female who was born term via spontaneous vaginal delivery (SVD) with a birth weight of 3.4 kg had an onset of persistent hypoglycaemia at the 6th hour of life. She was diagnosed with congenital hyperinsulinism based on high glucose load, negative ketone and a good response to glucagon. Genetic workup revealed the presence of ATP Binding Cassette Subfamily C Member 8 (ABCC8 genes) mutation which indicated a focal form of congenital hyperinsulinism. She was resistant to the standard dose of oral diazoxide but responded to subcutaneous somatostatin. At the age of 3 years and 6 mon

Hidradenitis suppurativa usually develops in the third decade of life and is infrequent in children and adolescents. This article reviews the scant epidemiological clinical and therapeutic data that are available for this age group. From the epidemiological point of view, reports have been more numerous in children, which has been related to hormone alterations and with a possibly higher genetic load than when this disease develops in adults. Clinically, it seems that when hidradenitis suppurativa develops in young patients, there is greater disease extension. The therapeutic approach is espec…

Three children with acrodermatitis enteropathica (AE) were treated with oral zinc dipicolinate (zinc-PA). The daily dose of zinc required to prevent exacerbations, when administered as the dipicolinate complex, was one-third the minimum amount of zinc required as the sulfate salt. The concentration of picolinic acid in the plasma of asymptomatic children with AE was significantly less than that of normal children. However, oral treatment with PA alone was ineffective. The plasma of the three AE children contained a measurable quantity of kynurenine which was undetectable in plasma from normal

Increased arterial pressure is obviously the major stimulus to cardiac hypertrophy in hypertension. However, different studies suggest that in addition to the pressure load, other factors could play participating roles in determining the degree of ventricular hypertrophy in response to the hypertensive disease as well as the degree of its reversal after control of arterial pressure. These other mechanisms include genetic factors and concimitant processes such as aging and the presence of cardiomyopathy or other disease. Two neurohumoral influences, namely, the adrenergic and the renin-angioten

The course of infection with Leishmania donovani was followed in seven strains of laboratory mice with measurement of the liver parasite burdens over 20 weeks. The acute parasite population growth rate varied greatly between, but not within, strains. Four strains were relatively resistant with less than an eight-fold increase while the three acutely susceptible strains showed over an eight-fold increase in the first month. Thereafter, one initially susceptible strain showed a dramatic fall in parasite numbers with histological liver damage while another strain maintained an immense parasite lo

Iron overload is an uncommon complication of untransfused chronic hemolytic anemias. This paper describes only the fourth reported case of erythrocyte pyruvate kinase deficiency and iron overload. Important aspects of this case are the presence of the HLA genotype commonly associated with genetic (idiopathic) hemochromatosis and a history of ingestion of large doses of ascorbic acid. Their roles in the development of iron loading and toxicity are discussed. A beneficial response to treatment with desferrioxamine was observed before significant iron removal. A mechanism for this action of desfe

Three of six members of one family have a renal glycosuria of variable degree. In addition to this they have a mild loss of phosphate under normal conditions. In one patient the phosphate-loss becomes very high after a loading dose. After describing the different data the mechanism of renal glucose transport and its relationship to phosphate transport as well as the genetic of renal glucosuria are discussed. The combination of renal glycosuria and renal loss of phosphate is very rare. The difference in regard to gluco-phosphaturia is mentioned.

Chagas disease (CD) exhibits marked clinical heterogeneity during its chronic phase, which has been partially attributed to the genetic diversity of Trypanosoma cruzi [which is currently classified into six Discrete Typing Units (DTUs, Tc I - Tc VI)]. However, establishing a direct genotype-phenotype correlation remains challenging. We conducted a cross-sectional study in 202 chronic CD patients from Córdoba, Argentina, combining molecular typing (mini-exon, 24Sα rRNA, 18S rRNA), quantitative PCR-based parasite load estimation, and clinical stratification according to cardiomyopathy presence a

BACKGROUND: Human immunodeficiency virus 1 (HIV-1) remains a global health challenge, emphasizing the need to identify new host factors that influence pathogenesis to serve as drug targets. Identifying genetic determinants of HIV-1 control across diverse populations remains a top priority.
METHODS: We conducted a multiancestry genome-wide association study (GWAS) of HIV-1 set-point viral load (spVL) leveraging genotype and viral load data from 10 723 individuals with HIV-1. To prioritize candidate genes and mechanisms, we integrated GWAS findings with transcriptome-wide association study (TWA

In the midst of a biodiversity crisis, it is important to conserve diversity at all levels, including species, genetic, and behavioural diversity. Animals with different behavioural traits can interact with their environment in distinct ways; hence, the distribution of behavioural traits within a population can influence the efficacy and outcome of conservation interventions. On the Galápagos Islands, Darwin’s finches are threatened by an invasive parasite, the avian vampire fly, Philornis downsi. Avian vampire fly larvae develop in the base of the nest, where they feed on the blood and tissue

The fading efficacy of antibiotics is a growing global health concern due to its life-threatening consequences and increased healthcare costs. Non-genetic mechanisms of antimicrobial resistance, such as those employed by Chlamydia pneumoniae and Chlamydia trachomatis, complicate treatment as these bacteria can enter a non-replicative, persistent state under stress, evading antibiotics and linking to inflammatory conditions. Understanding chlamydial persistence at the molecular level is challenging, and new models for studying Chlamydia-host interactions in vivo are urgently needed. Caenorhabdi

Infections due to human herpesvirus 6 (HHV-6) are frequent during early childhood. Usually, they have a favorable clinical course. Conversely, HHV-6 congenital infections occur in about 1% of neonates and may present with more severe clinical pictures. HHV-6 can be found in lung tissues and bronchoalveolar lavage (BAL) samples from patients with pneumonia and in immunocompromised patients can cause mild to severe pneumonia. In neonates, the role of HHV-6 in the genesis of severe pneumonia is poorly defined still now. We describe a healthy infant with a late-onset (15 days of life) severe inter

Most of the loci identified by genome-wide association studies (GWAS) for late-onset Alzheimer's disease (LOAD) are in strong linkage disequilibrium (LD) with nearby variants all of which could be the actual functional variants, often in non-protein-coding regions and implicating underlying gene regulatory mechanisms. We set out to characterize the causal variants, regulatory mechanisms, tissue contexts, and target genes underlying these associations. We applied our INFERNO algorithm to the top 19 non-APOE loci from the IGAP GWAS study. INFERNO annotated all LD-expanded variants at each locus

Single nucleotide polymorphisms in PICALM, a key component of clathrin-mediated endocytosis machinery, have been identified as genetic susceptibility loci for late onset Alzheimer's disease (LOAD). We previously reported that PICALM protein levels were decreased in AD brains and that PICALM was co-localised with neurofibrillary tangles in LOAD, familial AD with PSEN1 mutations and Down syndrome. In the present study, we analysed PICALM expression, cell localisation and association with pathological cellular inclusions in other tauopathies and in non-tau related neurodegenerative diseases. Late onset Alzheimer's disease (LOAD) etiology is influenced by complex interactions between genetic and environmental risk factors. Large-scale genome wide association studies (GWAS) for LOAD have identified 10 novel risk genes: ABCA7, BIN1, CD2AP, CD33, CLU, CR1, EPHA1, MS4A6A, MS4A6E, and PICALM. We sought to measure the influence of GWAS single nucleotide polymorphisms (SNPs) and gene expression levels on clinical and pathological measures of AD in brain tissue from the parietal lobe of AD cases and age-matched, cognitively normal controls. We found that ABCA7, CD33, and CR1 expression lev

The efficacy of Bacillus Calmette-Guérin (BCG) vaccine in preventing adult tuberculosis (TB) is highly variable. Genetic differences between BCG vaccine substrains, which can be divided into early strains and late strains based on the loss of region of difference two (RD2), may result in the variability and BCG substrains. The effect of lack of RD2 on the protective efficacy of BCG substrains against TB remains unknown. In this study, we demonstrated that CFP21 and MPT64(rCM) fusion protein, encoded by RD2 of Mycobacterium tuberculosis, could stimulate higher level of interferon (IFN)-γ in tub

The majority of pathogenic mitochondrial DNA (mtDNA) mutations are heteroplasmic, with both mutant and wild-type alleles present within the same individual. MtDNA is transmitted only from females to their offspring but a single female can bear offspring who harbour different levels of mutant mtDNA and have a variable phenotype. In single families, this complex genetic and phenotypic variability has confounded the identification of any relationship between the level of mutant mtDNA (mutation load) in the mother and the clinical features of her offspring. To obtain a more accurate description of

BACKGROUND: Tuberous sclerosis complex (TSC) is a rare genetic disorder with multisystem involvement. A magnetic-resonance (MRI) based classification of tubers into types A, B and C has been proposed. However, the relationship between different tuber types and their quantitative characteristics, also the non-neurological manifestations of TSC remains unknown.
AIMS: To quantitatively evaluate different MRI-defined tuber types and to explore their relationships with major disease manifestations in patients with tuberous sclerosis complex.
METHODS: We performed quantitative manual assessment of
