Acanthamoeba induces cell-cycle arrest in host cells.

Acanthamoeba can cause fatal granulomatous amoebic encephalitis (GAE) and eye keratitis. However, the pathogenesis and pathophysiology of these emerging diseases remain unclear. In this study, the effects of Acanthamoeba on the host cell cycle using human brain microvascular endothelial cells (HBMEC) and human corneal epithelial cells (HCEC) were determined. Two isolates of Acanthamoeba belonging to the T1 genotype (GAE isolate) and T4 genotype (keratitis isolate) were used, which showed severe cytotoxicity on HBMEC and HCEC, respectively. No tissue specificity was observed in their ability to

Host NK cells are required for the growth of the human filarial parasite Brugia malayi in mice.

Human lymphatic filariasis, which afflicts an estimated 120 million people worldwide, is caused by the large nematode parasites Wuchereria bancrofti and Brugia malayi. Filarial nematodes require both an arthropod vector and a mammalian host to complete their life cycle. Within the definitive (mammalian) host, the lymphatic filarial parasites reside in the lymph nodes and lymphatics, a seemingly hostile environment for infectious agents, since the location exposes them to the immune defenses of the host. We present data here that suggest that the growth of B. malayi in the mammalian host is dep

Variability in infection dynamics emerges from the interplay between unique host and pathogen characteristics.

Variation in infection dynamics across host species can profoundly influence parasite epidemiology, ecology, and evolution. However, because experiments involving multiple hosts and multiple parasite species that capture the full infection dynamics within hosts are rare, the underlying mechanisms of host heterogeneity effects across different parasite species remain largely unknown. We dissected the specific roles of host and pathogen effects in shaping within-host infection dynamics using a model system comprising three coexisting rodent species from Israel's northwestern Negev Desert and the

An impossible journey? The development of Plasmodium falciparum NF54 in Culex quinquefasciatus.

Although Anopheles mosquitoes are the vectors for human Plasmodium spp., there are also other mosquito species-among them culicines (Culex spp., Aedes spp.)-present in malaria-endemic areas. Culicine mosquitoes transmit arboviruses and filarial worms to humans and are vectors for avian Plasmodium spp., but have never been observed to transmit human Plasmodium spp. When ingested by a culicine mosquito, parasites could either face an environment that does not allow development due to biologic incompatibility or be actively killed by the mosquito's immune system. In the latter case, the molecular

[Relationships of the plague pathogen and vector from different parasitic systems].

The specific features of interaction of the strains of the plague microbe of the main subspecies, which circulate in the area of natural foci of Mongolia and China, with Citellophilus tesquorum sungaris fleas, the major vector of the plague pathogen in a Transbaikalian natural focus, as well as with Xenopsylla cheopis ones, the classical vector, were revealed. Experiments used virulent Yersinia pestis strains, such as I-3230 isolated from C.tesquorum in Mongolia in 1998 and 2155 isolated from humans in Manchuria (China) in 1947. They established that ectoparasites from other parasitic systems

Olfaction shapes host-parasite interactions in parasitic nematodes.

Many parasitic nematodes actively seek out hosts in which to complete their lifecycles. Olfaction is thought to play an important role in the host-seeking process, with parasites following a chemical trail toward host-associated odors. However, little is known about the olfactory cues that attract parasitic nematodes to hosts or the behavioral responses these cues elicit. Moreover, what little is known focuses on easily obtainable laboratory hosts rather than on natural or other ecologically relevant hosts. Here we investigate the olfactory responses of six diverse species of entomopathogenic

Within-host dynamics of mycoplasma infections: conjunctivitis in wild passerine birds.

The host-pathogen interaction drives infectious disease dynamics at the individual, population and community levels. Here I present and analyze a model of the vertebrate immune response to mycoplasma infections, and use it to identify which pathogen and host immune characteristics drive patterns of Mycoplasma gallisepticum (MG) infections in the house finch (Carpodacus mexicanus) and other passerine birds. I also address which host and pathogen characteristics most affect host infectiousness and survival. These results imply that much of the observed variation in the house finch likely arises

Diversity and biology of Spirometra tapeworms (Cestoda: Diphyllobothriidea), zoonotic parasites of wildlife: A review.

Tapeworms of the genus Spirometra Faust, Campbell et Kellogg, 1929 have long been known as intestinal parasites of carnivores and their larv … osts, and their distribution. The existing taxonomic problems and the insufficient knowledge of the host specificity and distribution of Spirometra tapeworms can only be solved by combining molecular and morphological data, i.e. by comparing genetically characterised specimens with corresponding morphological vouchers (hologenophores). Redescription and molecular characterization of two species of Pauciconfibula (Monogenea, Microcotylidae) from trachinid fishes in the Mediterranean Sea.

Many Pauciconfibula spp. have a long and complicated taxonomic history. The remaining unsolved taxonomic confusion in this genus is impelled by the host range and status of Pauciconfibula spp. from trachinid fishes: Pauciconfibula trachini and Pauciconfibula draconis, from Trachinus radiatus and Trachinus draco (Trachinidae), respectively. Pauciconfibula trachini was reported on Trachinus draco, type host of Pauciconfibula draconis suggesting thus a stenoxenic specificity for the former monogenean and the occurrence of two congeneric polyopisthocotyleans on a single host. Moreover, the validit

What adaptive changes in hemagglutinin and neuraminidase are necessary for emergence of pandemic influenza virus from its avian precursor?

Wild ducks serve as the primary host for numerous and various influenza type A viruses. Occasionally, viruses from this reservoir can be transferred to other host species and cause outbreaks of influenza in fowl, swine, and horses, as well as result in novel human pandemics. Cellular tropism and range of susceptible host species are determined by interaction between virus and receptor molecules on cells. Here we discuss modern data regarding molecular features underlying interactions of influenza viruses with cellular receptors as well as a role for receptor specificity in interspecies transmi

Evidence for a human-specific Escherichia coli clone.

Escherichia coli is a widespread commensal of the vertebrate intestinal tract. Until recently, no strong association between a particular clone and a given host species has been found. However, members of the B2 subgroup VIII clone with an O81 serotype appear to be human host specific. To determine the degree of host specificity exhibited by this clone, a PCR-based assay was used to screen 723 faecal and clinical isolates from humans, and 904 faecal isolates from animals. This clone was not detected among the animal isolates, but was discovered in people living in Africa, Europe and South Amer…

Neuraminidase-mediated augmentation of in vitro immune response of patients with solid tumors.

Host blood lymphocytes undergo accentuated blastic transformation when cultured with tumor cells pretreated with neuraminidase. The effect has been observed in 38 patients with such common solid tumors as bronchus carcinoma, skin melanoma, hypernephroma, or adenocarcinoma of the breast, lung, colon, or rectum. Individual response varied but often exceeded response to allogeneic cells. Three patients with glioblastoma of the brain did not respond. Lymphoblastic transformation was not observed in three of four cultures containing benign tumor or in any cultures containing normal tissue analogues

Outer surface protein polymorphisms linked to host-spirochete association in Lyme borreliae.

Lyme borreliosis is caused by multiple species of the spirochete bacteria Borrelia burgdorferi sensu lato. The spirochetes are transmitted by ticks to vertebrate hosts, including small- and medium-sized mammals, birds, reptiles, and humans. Strain-to-strain variation in host-specific infectivity has been documented, but the molecular basis that drives this differentiation is still unclear. Spirochetes possess the ability to evade host immune responses and colonize host tissues to establish infection in vertebrate hosts. In turn, hosts have developed distinct levels of immune responses when inv

Secreted effectors in Toxoplasma gondii and related species: determinants of host range and pathogenesis?

Recent years have witnessed the discovery of a number of secreted proteins in Toxoplasma gondii that play important roles in host-pathogen interactions and parasite virulence, particularly in the mouse model. However, the role that these proteins play in driving the unique features of T. gondii compared to some of its nearest apicomplexan relatives (Hammondia hammondi and Neospora caninum) is unknown. These unique features include distinct dissemination characteristics in vivo and a vast host range. In this review we comprehensively survey what is known about disease outcome, the host response

Female Mouse Model of Neisseria gonorrhoeae Infection.

Mouse models of infection are important tools in the study of infectious disease or host the development of products to prevent or treat infections. The estradiol-treated mouse model of Neisseria gonorrhoeae genital tract infection has proved to be a valuable system for determining the importance of gonococcal factors that mediate evasion of host innate effectors in vivo or host gonococcal adaptation to hormonally driven host factors in females. Examination of mechanisms that Neisseria gonorrhoeae uses to subvert the host immune response also has been greatly aided by this whole model system,
