Association of Blastocystis subtype 3 with elevated IL-8-mediated inflammation in patients with irritable bowel syndrome.

BACKGROUND: The role of Blastocystis in the pathophysiology of irritable bowel syndrome (IBS) remains controversial, with emerging evidence suggesting subtype-specific differences in pathogenicity. Interleukin-8 (IL-8), a key pro-inflammatory chemokine, has been implicated in mucosal immune activation in both IBS and parasitic infections. OBJECTIVE: This study aimed to investigate the association between Blastocystis subtypes and clinical manifestations of IBS, with a particular focus IL-8 as part of the systemic pro-inflammatory immune response associated with functional GIT disorders and in

Resolution Medicine: Steps to Uncovering the Specialized Pro-resolving Mediator Superfamily and Their Potent Functions in Resolving Inflammation.

Surgery, barrier breaks, injury, and microbial invasions initiate the cardinal signs of inflammation: heat, redness, swelling, and pain mediated by the classic prostaglandins. Tissue histology defined the acute inflammatory response, its initiation with leukocyte infiltration, and normally self-limited resolution phase. Resolution, once thought to be passive, is an active program accompanied by temporal biosynthesis of novel lipid mediators that activate clearance of apoptotic neutrophils (polymorphonuclear neutrophil-PMN) and cellular debris and limit the further PMN infiltration. Structural

Micro- and nanoplastics-mediated immunometabolic reprogramming: a systemic nexus for chronic inflammation and multi-organ disease risk.

Micro- and nanoplastics (MNP) are ubiquitous environmental stressors increasingly detected in human tissues and directly linked to clinical cardiovascular events. This review proposes immunometabolic reprogramming as the central nexus through which MNP drive systemic immune dysregulation and chronic inflammation. Upon entering via multiple routes, MNP selectively sequester in immune organs, disrupting mitochondrial quality control via Drp1-mediated fission and activating the cGAS-STING pathway. Furthermore, MNP induce a pseudohypoxic state that stabilizes HIF-1a, driving a glycolytic "Warburg-

Skeletal muscle health and the development of prediabetes in middle-aged and older adults: mediated by inflammation and lipid indices.

AIMS: This study aims to examine the effect of relative muscle strength (RMS) on the progression to diabetes or regression to normoglycemia in individuals with prediabetes. METHODS: Data were sourced from the China Health and Retirement Longitudinal Study (CHARLS), which followed 3250 individuals aged ≥45 years with prediabetes. RMS was calculated as the ratio of grip to appendicular skeletal muscle mass (ASM). Multinomial logistic regression was used to assess the association of RMS with subsequent glycemic transitions, including regression to normoglycemia, persistence of prediabetes, and p

Cardiomyocyte-Derived USP20 Attenuates Diabetic Cardiomyopathy by Facilitating the Degradation of STING and Mitigating STING-Mediated Inflammation.

Although extensive clinical and basic research has been conducted on diabetic cardiomyopathy (DbCM), the therapeutic efficacy for this condition remains significantly limited. Ubiquitin-specific peptidase 20 (USP20), a deubiquitinating enzyme, plays an essential role in regulating protein ubiquitination and modulating various cellular processes. In this study, we aimed to investigate the effect of USP20 on the pathogenesis of DbCM, which may provide a novel therapeutic target for its treatment. The cardiomyocyte-specific USP20 conditional knockout (USP20CKO) mice were employed in this study. T

Simvastatin Ameliorates Lipid Metabolism-Mediated Endothelial Inflammation and Tight Junction Impairment in Chronic Cerebral Hypoperfusion.

Chronic cerebral hypoperfusion (CCH) is a key pathological hallmark observable in multiple subtypes of cerebral small vessel disease (CSVD). This condition causes both structural and functional changes within the brain's vascular system, and is particularly damaging to brain microvascular endothelial cells (BMECs). The exact molecular mechanisms underlying BMEC impairment in CCH remain insufficiently defined despite their clinical importance. Emerging evidence indicates that disturbances in intracellular lipid metabolism might contribute substantially to promoting endothelial inflammation and

Microbiota-Mediated Conversion of Asiaticoside to Asiatic Acid Restores Intestinal Homeostasis in Mice with Chronic Intestinal Inflammation.

Asiaticoside, a triterpenoid saponin abundant in Centella asiatica, exhibits notable anti-inflammatory activity but suffers from poor oral bioavailability. Here, we reveal a gut microbiota-dependent mechanism underlying its therapeutic efficacy against chronic intestinal inflammation. Using a DSS-induced mouse model and antibiotic-mediated microbiota depletion, we demonstrate that asiaticoside requires microbial metabolism to alleviate epithelial injury, oxidative stress, and inflammatory cytokine production. HPLC analysis showed that asiaticoside is hydrolyzed by gut microbes into asiatic aci

Tributyl phosphate disrupts hepatic lipid metabolism via cGAS-STING signal pathway mediated inflammation.

Tributyl phosphate (TBP) can lead to abnormal hepatic lipid metabolism. Inflammation triggered by the cGAS-STING may be involved in it. Within this research, we investigated the involved mechanisms of TBP-induced lipid metabolic disorders. Rats and BRL-3A cells are used as models to determine the liver toxicity of TBP. Aspirin was used to alleviate the inflammation induced by TBP. RU.521 and H151 were employed to inhibit the activation of cGAS-STING. HE staining was applied to observe liver injury. The mitochondrial structure was observed with transmission electron microscopy. The quantificati

Resveratrol Attenuates Liver Inflammation in Non-Alcoholic Fatty Liver Disease by Activating PINK1-Mediated Mitophagy.

Resveratrol (RES) has been shown to exhibit therapeutic efficacy against fatty liver disease. Yet, the molecular mechanisms by which RES ameliorates liver injury remain unclear. The aim of this study was to investigate the therapeutic effect and mechanism of resveratrol in fatty liver disease. It was found that dairy cows with fatty liver exhibit characteristic hepatic pathologies, including ballooning degeneration, lipid accumulation and elevated serum AST and ALT levels. Parallel to these changes, we observed significant upregulation of the NLRP3 inflammasome alongside suppression of mitopha

Few-shot learning-driven discovery of Lutein suppresses Th1-mediated inflammation via glucose metabolism.

The discovery of selective immunosuppressants for T cell-mediated diseases like Ulcerative Colitis (UC) is a significant challenge. While traditional screening is inefficient, Artificial intelligence (AI)-driven approaches are often hindered by the scarcity of high-quality labeled data, challenging the accurate identification of functional molecules. In this study, we leveraged a transfer learning strategy to compensate for the lack of high-quality data, establishing a screening platform to identify potential T cell inhibitors. Using this approach, we identified Lutein as a novel, specific imm

Cluster of Differentiation 36-Mediated Inflammation and Lipid Metabolism in Cardiovascular Diseases: From Mechanisms to Novel Therapies.

Cardiovascular diseases (CVDs) continue to pose a key challenge to public health because of their high prevalence, mortality, and disability rates, together with a trend toward younger age at onset. Chronic inflammation and disordered lipid metabolism are key pathological mechanisms underlying the development and progression of CVDs. Although numerous pharmacological agents have been developed to intervene in CVDs, current therapies are still limited by adverse effects, suboptimal efficacy, and insufficient anti-inflammatory properties. Consequently, effective drug-based strategies for the pre

Association between systemic inflammation and overactive bladder: The potential mediating role of obesity-related anthropometric indices.

Systemic inflammation is increasingly implicated in the development of chronic diseases, yet the specific association between the Aggregate Index of Systemic Inflammation (AISI) - a novel composite inflammatory marker - and overactive bladder (OAB) remains poorly understood. This study aimed to elucidate the association between AISI levels and OAB prevalence and to explore the statistical mediating effect of anthropometric indicators within the potential inflammation-metabolism pathway. This cross-sectional study utilized data from the National Health and Nutrition Examination Survey (NHANES)

Extracellular Vesicles of Streptococcus anginosus Mediate Gastritis via Epithelial Barrier Disruption and Macrophage-driven Inflammation.

Recent studies suggest Streptococcus anginosus (SA) contributes to gastric disease beyond Helicobacter pylori, yet its pathogenic mechanisms remain unclear. This study demonstrates that SA-derived extracellular vesicles (SA-EVs) accumulate in gastric tissue, enter epithelial cells, and induce acute gastritis characterized by neutrophil infiltration and elevated cytokines (TNF-α, IL-6, IL-17A). Chronic exposure leads to sustained inflammation, tight junction disruption (Claudin-18, Occludin, ZO-1), and mucosal damage. Proteomic analysis identified TMPC and FBP62 as virulence-associated proteins

Nesfatin-1 Ameliorates Acute Pancreatitis by Inhibiting the Pyroptosis of Acinar Cells and Inflammation Mediated by Macrophages and Neutrophils.

Acute pancreatitis (AP) is an inflammatory disorder with no efficient therapy. Here we demonstrate that the anorexigenic peptide nesfatin-1 exerts potent and dose-dependent protection against both caerulein-induced and hypertriglyceridemic AP. Intraperitoneal administration of nesfatin-1 to restore its serum levels significantly reduced pancreatic necrosis, edema, and infiltration of immune cells, as well as circulating levels of amylase, lipase, and pro-inflammatory cytokines. RNA-seq revealed that nesfatin-1 down-regulated the ER-stress signature (Ddit3, Atf3, Ppp1r15a) and the NF-κB/NLRP3 s

Inflammation-mediated function enhancement of KNa channels promotes network hyperactivity in the peripheral cortex following TBI.

BACKGROUND: Post-traumatic epilepsy (PTE) is a common complication of traumatic brain injury (TBI). Studies have indicated that functional abnormalities of the sodium-activated potassium (KNa) channels are closely related to infantile "refractory epilepsy". However, whether and how the KNa channels are involved in the occurrence of PTE remains unknown. METHODS: We used adult male C57BL/6 J mice to establish a controlled cortical impact (CCI) mice model with different severity levels. We implanted intracranial electrodes 7 days after injury to observe the spontaneous seizures in the moderately

Epidermal METTL1-Mediated m7G Modification Drives Psoriatic Inflammation by Stabilizing Bdkrb1 and Orchestrating Neutrophil Recruitment.

The functional significance of RNA modifications, specifically N7-methylguanosine (m7G), in inflammatory conditions such as psoriasis remains not fully elucidated. This study demonstrates that methyltransferase-like 1 (METTL1), an m7G methyltransferase, is significantly upregulated in epidermal keratinocytes of human psoriatic lesions and imiquimod (IMQ)-induced murine models. Utilizing mice with an inducible keratinocyte-specific Mettl1 deletion (Mettl1fl/flKrt14-CreERT2), the research reveals significantly attenuated psoriasiform inflammation and decreased neutrophil infiltration relative to

Carnosic acid and carnosol promote lipid mediator class-switching toward inflammation resolution in human macrophages.

Lipid mediators (LM), formed from polyunsaturated fatty acids (PUFA) through cyclooxygenase- and lipoxygenase (LOX)-dependent pathways, contribute to all stages of inflammation and are thus valuable targets for drugs that intervene with inflammatory diseases. Novel pharmacotherapeutic concepts attempt to support LM class-switching from pro-inflammatory 5-LOX-derived leukotrienes and cyclooxygenase-derived prostaglandins towards omega-3-PUFA-based specialized pro-resolving mediators (SPM), which accomplishes resolution of inflammation.
