Exploring the Scope and Status of Biological Interventions in Malaria Control.

Malaria is a major health concern worldwide, with an estimated 282 million infections and 610,000 deaths in 2024. Plasmodium parasites, spread by female Anopheles mosquitoes, are the cause. Several interventions, such as indoor residual spraying, insecticide-treated bed nets, and antimalarial drugs, have reduced transmission; however, challenges such as insecticide resistance, environmental concerns, and climate change have led to the exploration of alternative approaches. To address these limitations, biological control, which involves the use of natural organisms and biological agents to tar…

Genetic survey of Ivermectin-resistance markers in head lice from Jazan, Saudi Arabia.

Insecticide resistance in head lice is a widespread problem and has been intensively studied in recent years. Whereas numerous investigations have examined pyrethroid resistance, ivermectin (IVM) resistance has not been assessed in Saudi Arabia and has been addressed in few studies worldwide. IVM is the second most commonly used drug to treat head-louse infestations, and resistance to it is associated with five mutations in the glutamate-gated chloride channel (GluCl) gene. This study aimed to estimate the prevalence of GluCl mutations in the Jazan region. Sanger sequencing and PCR-restriction…

Genetic diversity and occurrence of permethrin resistance-associated mutations in a population of head lice (Pediculus humanus capitis) from Seville, Spain.

Human head lice (Pediculus humanus capitis De Geer, 1767) remain a public health concern, largely due to their widespread resistance to commonly used pediculicides. This study assessed genetic diversity and permethrin resistance in head lice collected from individuals in Seville, Spain. A total of 402 lice were sampled, of which 27 specimens were selected for detailed morphological, molecular, and phylogenetic analyses. Molecular characterization based on mitochondrial markers (cox1, cob and 12S) revealed a specific pattern of genetic diversity, identifying two distinct clades (A and B), with

Fixation of knockdown resistance mutations in Cimex hemipterus (Hemiptera: Cimicidae) from east Iran.

The tropical bed bug, Cimex hemipterus (F.), is an obligate blood-feeding ectoparasite and a well-established pest in urban settings, particularly in tropical and subtropical regions. Insecticide resistance is one of the main challenges in bed bug management worldwide, with pyrethroid resistance being especially widespread. Iran's diverse geography and climate support the presence of both Cimex lectularius L. and C. hemipterus, with the latter being commonly found in the warmer, arid eastern provinces. In this study, we investigated the target site mutations that underlie pyrethroid resistance…

Efficacy of phosphine (PH3) under hypoxia: pilot and industry-scale trials for controlling strongly resistant stored product pests in rice.

BACKGROUND: Phosphine (PH3) is a slow-acting fumigant, therefore control of resistant insect pests requires high concentrations of 540-1080 ppm (0.75-1.5 g m-3) over long exposure periods of up to 21 days. This is difficult to achieve under many practical storage conditions; hence, there is a need to enhance the toxicity of PH3, so that resistant insects can be eradicated in a shorter fumigation time. Co-fumigation with PH3, along with other atmospheric gases (carbon dioxide (CO2) and nitrogen (N2)), enhances PH3 toxicity in laboratory-scale experiments; however, this information has not been

Measuring local genetic variation in permethrin-resistant head lice (Phthiraptera: Pediculidae) from Buenos Aires, Argentina.

The cosmopolitan ectoparasite human head louse, Pediculus humanus capitis (De Geer)(Phthiraptera:Pediculidae), affects mostly school-aged children, with infestations reported every year mainly due to louse resistance to pyrethroids. One of the main resistance mechanisms of pyrethroids is the target site insensitivity (kdr), which is caused by single-nucleotide point mutations (SNPs) located in the voltage-sensitive sodium channel gene. In this study, we analyzed individual head lice toxicologically via the description of their susceptibility profile to permethrin and genetically through the ge

ABCH2 transporter mediates deltamethrin uptake and toxicity in the malaria vector Anopheles coluzzii.

Contact insecticides are primarily used for the control of Anopheles malaria vectors. These chemicals penetrate mosquito legs and other appendages; the first barriers to reaching their neuronal targets. An ATP-Binding Cassette transporter from the H family (ABCH2) is highly expressed in Anopheles coluzzii legs, and further induced upon insecticide exposure. RNAi-mediated silencing of the ABCH2 caused a significant increase in deltamethrin mortality compared to control mosquitoes, coincident with a corresponding increase in 14C-deltamethrin penetration. RT-qPCR analysis and immunolocalization r

Geographical distribution of pyrethroid resistance allele frequency in head lice (Phthiraptera: Pediculidae) from Argentina.

The human head louse, Pediculus humanus capitis De Geer (Phthiraptera: Pediculidae), is an obligate ectoparasite that causes pediculosis capitis and has parasitized humans since the beginning of humankind. Head louse infestations are widespread throughout the world and have been increasing since the early 1990s partially because of ineffective pediculicides. In Argentina, the overuse of products containing pyrethroids has led to the development of resistant louse populations. Pyrethroid insecticides act on the nervous system affecting voltage-sensitive sodium channels. Three point mutations at

Diagnoses of fipronil resistance in Brazilian cattle ticks (Rhipicephalus (Boophilus) microplus) using in vitro larval bioassays.

Fipronil is a phenylpyrazolic insecticide that is widely used in agriculture and has been recently used to control the cattle tick, Rhipicephalus (Boophilus) microplus. Because of the serious problems associated with resistance to the available acaricides, this product has been used as an important alternative to control acaricide-resistant ticks. The objective of this work was to analyse the fipronil sensitivity of ticks that were collected from farms with a history of fipronil use by larval bioassays. A total of 11 Brazilian tick populations were studied: one population from Rio Grande do Su

Efficacy of coumaphos applied as a dip for control of an organophosphorus-resistant strain of Boophilus microplus (Acari: Ixodidae) on cattle.

The efficacy of coumaphos, an organophosphorus acaricide, was tested on a strain of Boophilus microplus (Canestrini) that was 9.5 times more resistant to the chemical than a susceptible strain. Cattle infested with larval, nymphal, and adult ticks were dipped into a vat charged at concentrations of 0.031, 0.083, 0.160, and 0.279% (AI). Assessment of efficacy was based on surviving tick numbers, female weight, egg mass weight, egg hatchability, and index of reproduction. At 0.031% (AI), the level of control was lower than the other coumaphos treatments, and the weight, fecundity, and fertility

Design and synthesis of mesoionic compounds containing benzoxacycle for the control of piercing-sucking pests.

Piercing-sucking pests cause significant agricultural losses, and widespread neonicotinoid use has led to resistance and ecological concerns. Mesoionic compounds show broad-spectrum insecticidal activity with low cross-resistance, yet the mechanism and potential of tetrahydrothiazolo[3,2-a]pyrimidine mesoionic scaffolds against these pests remain largely unexplored. Here, a series of novel tetrahydrothiazolo[3,2-a]pyrimidine mesoionics containing a benzoxacycle moiety were designed, synthesized, and evaluated for insecticidal activity against Megoura crassicauda (M. crassicauda) and Sogatella

Larvicidal Activity of Hemp Extracts and Cannabidiol against the Yellow Fever Mosquito Aedes aegypti.

To mitigate pyrethroid resistance in mosquito vectors of emerging and re-emerging human pathogens, there is an urgent need to discover insecticides with novel modes of action. Natural alternatives, such as extracts derived from plants, may serve as substitutes for traditional synthetic insecticides if they prove to be sustainable, cost-effective, and safe for non-target organisms. Hemp (Cannabis sativa) is a sustainable plant known to produce various secondary metabolites with insecticidal properties, including terpenoids and flavonoids. The goal of this study was to assess the larvicidal acti

Multiple combinations of RDL subunits diversify the repertoire of GABA receptors in the honey bee parasite Varroa destructor.

In insects, γ-aminobutyric acid (GABA) is the major inhibitory neurotransmitter, and GABA-gated ion channels are the target of different classes of insecticides, including fipronil. We report here the cloning of six subunits (four RDL, one LCCH3, and one GRD) that constitute the repertoire of the GABA-gated ion channel family of the Varroa mite (Varroa destructor), a honey bee ectoparasite. We also isolated a truncated GRD subunit with a premature stop codon. We found that when expressed in Xenopus laevis oocytes, three of the four RDL subunits (VdesRDL1, VdesRDL2, and VdesRDL3) formed functio

A natural agonist of mosquito TRPA1 from the medicinal plant Cinnamosma fragrans that is toxic, antifeedant, and repellent to the yellow fever mosquito Aedes aegypti.

Plants produce various secondary metabolites that offer a potential source of novel insecticides and repellents for the control of mosquito vectors. Plants of the genus Cinnamosma are endemic to, and widely-distributed throughout, the island of Madagascar. The barks of these species are commonly used in traditional medicines for treating a wide range of maladies. The therapeutic nature of the bark is thought to be associated with its enrichment of pungent drimane sesquiterpenes, which elicit antifeedant and toxic effects in some insects. Here we test the hypothesis that a bark extract of Cinna

Pesticide Chemical Research in Toxicology: Lessons from Nature.

Pesticide researchers are students of nature, and each new compound and mechanism turns a page in the ever-expanding encyclopedia of life. Pesticides are both probes to learn about life processes and tools for pest management to facilitate food production and enhance health. In contrast to some household and industrial chemicals, pesticides are assumed to be hazardous to health and the environment until proven otherwise. About a thousand current pesticides working by more than 100 different mechanisms have helped understand many processes and coupled events. Pesticide chemical research is a ma
