Reactive oxygen species mediate IL-8 expression in Down syndrome candidate region-1-overexpressed cells.

Reactive oxygen species (ROS) have been considered to mediate inflammation in Down syndrome (DS). The present study is purposed to examine the mechanism of increased ROS levels and inflammatory cytokine IL-8 expression in Down syndrome candidate region-1 (DSCR1)-transfected cells, by determining ROS levels, IL-8 expression, NF-κB activation, and SOD1 levels in human embryonic kidney (HEK) 293 cells. The cells were treated with an antioxidant N-acetyl cysteine (NAC) or a calcium chelator BAPTA and stimulated with or without IL-1β. As a result, basal levels of ROS, IL-8, and NF-κB-DNA binding ac

High efficiency biosynthesis of 1α,25-Dihydroxyvitamin D3 via engineered dihydroxylase and deep eutectic solvent based resting cell bioprocess.

1α,25-dihydroxyvitamin D3 (1α,25(OH)2VD3), the biologically active form of Vitamin D3 (VD3), plays a crucial role in a wide range of physiological processes. However, the efficient biosynthesis of 1α,25(OH)2VD3 is hindered by the low activity of natural enzymes, poor solubility of substrate VD3, and its intracellular metabolic loss. Herein, we developed an integrated bioprocess by synergizing engineered dihydroxylase (CYP109A2-H) with a deep eutectic solvent (DES) based resting cell system to overcome these limitations. A semi-rational design strategy was employed to enhance the performance of

Modeling Facilitated Diffusion of Proteins in Crowded Environment.

The interaction between proteins and DNA is fundamental to numerous enzymatic processes, relying on the precise recognition of specific DNA bases by DNA-binding proteins (DBPs). This interaction occurs within the densely packed environment of the cell nucleus, which contains high concentrations of biomolecules. Consequently, the limited intracellular space can impact the diffusion and association of large macromolecules. This scenario prompts the question of how the complex milieu of the cell nucleus influences the ability of DBPs to locate and bind to their target DNA sites within a large pol…

Air-Mediated Biomimetic Synthesis of Polyhydroxyalkanoate with C4 Diol.

Poly(4-hydroxybutyrate) (P4HB) is a high-performance, well-recyclable, and biodegradable polyhydroxyalkanoate (PHA). However, conventional bioproduction of homopolymeric P4HB involves complex and costly processes with C4 feedstocks, particularly 1,4-butanediol (BDO), and enzyme-coenzyme systems in genetically engineered bacteria. An alternative extracellular chemical route utilizing aerial oxidation of BDO offers cost and energy benefits but struggle with conversion efficiency. Inspired by efficient intracellular oxidation of primary alcohols, we propose a ruthenium-phosphine synergistic catal

Extracellular matrix guidance of autophagy: a mechanism regulating cancer growth.

The extracellular matrix (ECM) exists as a dynamic network of biophysical and biochemical factors that maintain tissue homeostasis. Given its sensitivity to changes in the intra- and extracellular space, the plasticity of the ECM can be pathological in driving disease through aberrant matrix remodelling. In particular, cancer uses the matrix for its proliferation, angiogenesis, cellular reprogramming and metastatic spread. An emerging field of matrix biology focuses on proteoglycans that regulate autophagy, an intracellular process that plays both critical and contextual roles in cancer. Here,

Glial contribution to cyclodextrin-mediated reversal of cholesterol accumulation in murine NPC1-deficient neurons in vivo.

Niemann-Pick type C disease is a rare and fatal lysosomal storage disorder presenting severe neurovisceral symptoms. Disease-causing mutations in genes encoding either NPC1 or NPC2 protein provoke accumulation of cholesterol and other lipids in specific structures of the endosomal-lysosomal system and degeneration of specific cells, notably neurons in the central nervous system (CNS). 2-hydroxypropyl-beta-cyclodextrin (CD) emerged as potential therapeutic approach based on animal studies and clinical data, but the mechanism of action in neurons has remained unclear. To address this topic in vi

New insights into the pathogenesis of Peyronie's disease: A narrative review.

Peyronie's disease (PD) is a benign, progressive fibrotic disorder characterized by scar or plaques within the tunica albuginea (TA) of the penis. This study provides new insights into the pathogenesis of PD based on data from different studies regarding the roles of cytokines, cell signaling pathways, biochemical mechanisms, genetic factors responsible for fibrogenesis. A growing body of literature has shown that PD is a chronically impaired, localized, wound healing process within the TA and the Smith space. It is caused by the influence of different pathological stimuli, most often the effe

The molecular structure and function of sorting nexin 10 in skeletal disorders, cancers, and other pathological conditions.

SNX10 is a member of the phox homology domain-containing family of phosphoinositide-binding proteins. Intracellularly, SNX10 localizes to endosomes where it mediates intracellular trafficking, endosome organization, and protein localization to the centrosome and cilium. It is highly expressed in bone and the gut where it participates in bone mineral and calcium homeostasis through the regulation of osteoclastic bone resorption and gastric acid secretion, respectively. Not surprisingly, patients harboring mutations in SNX10 mutation manifest a phenotype of autosomal recessive osteopetrosis or m

Uropathogenic enterobacteria use the yersiniabactin metallophore system to acquire nickel.

Invasive Gram-negative bacteria often express multiple virulence-associated metal ion chelators to combat host-mediated metal deficiencies. Escherichia coli, Klebsiella, and Yersinia pestis isolates encoding the Yersinia high pathogenicity island (HPI) secrete yersiniabactin (Ybt), a metallophore originally shown to chelate iron ions during infection. However, our recent demonstration that Ybt also scavenges copper ions during infection led us to question whether it might be capable of retrieving other metals as well. Here, we find that uropathogenic E. coli also use Ybt to bind extracellular

Correlation Between the Severity of Subcutaneous Echo-Free Space and the Amount of Extracellular Fluid Determined by Bioelectrical Impedance Analysis of Leg Edema.

BACKGROUND: Both subcutaneous echo-free space (SEFS) observed by ultrasonography and the ratio of intracellular fluid resistance to extracellular fluid resistance (Ri/Re) calculated from bioelectrical impedance analysis (BIA) represent extracellular edema status in the extremities. We tried to correlate these parameters. METHODS AND RESULTS: In 70 legs with venous edema (VE) and 68 legs with lymphedema (LE), subcutaneous tissue ultrasonography to determine SEFS severity (range, 0-2) and BIA to calculate Ri/Re were performed. In the calf, SEFS severity showed good linear correlation with Ri/Re

Immunohistochemical localization of collagen type III and type IV, laminin, tenascin and alpha-smooth muscle actin (alphaSMA) in the human liver in peliosis.

The expression of collagen types III and IV, laminin, tenascin, and hepatic stellate cells (HSCs) activation marker alphaSMA was evaluated immunohistochemically in the liver of three patients with non-bacilar peliosis. Peliosis was attributed to tuberculosis, endometriosis treated with anabolic androgenic steroids, and to pheochromocytoma. Ultrastructural examination of the lesions of the liver revealed cavities that were sometimes lined with sinusoidal endothelial cells or hepatocytic microvilli. In liver sinusoids around cavities, cystic dilatation of the space of Disse and an abundance of a

Mechanism of apoptosis induced by zinc deficiency in peripheral blood T lymphocytes.

Alterations in intracellular Zn(2+) concentrations are believed to play a crucial role in modulating apoptosis. The observation that Zn(2+) deficiency can induce cell death both in vivo and in vitro has been attributed to the fact that exchange of Zn(2+) for Ca(2+) and Mg(2+) within the nuclei may directly activate endogenous endonucleases therefore inducing DNA fragmentation independent of cytoplasmic factors. Here we show that the membrane-permeable zinc chelator, N,N',N'-tetrakis(2-pyridylmethyl) ethylenediamine (TPEN) induces translocation of cytochrome c from the mitochondrial intramembra

Enzymatic defenses against the toxicity of oxygen and of streptonigrin in Escherichia coli.

Anaerobically grown Escherichia coli K-12 contain only one superoxide dismutase and that is the iron-containing isozyme found in the periplasmic space. Exposure to oxygen caused the induction of a manganese-containing superoxide dismutase and of another, previously undescribed, superoxide dismutase, as well as of catalase and peroxidase. These inductions differed in their responsiveness towards oxygen. Thus the very low levels of oxygen present in deep, static, aerobic cultures were enough for nearly maximal induction of the manganese-superoxide dismutase. In contrast, induction of the new sup

Morphogenesis of aura virus.

Aura virus, a member of the Western equine-encephalitis-Whataroa subgroup of group A arboviruses, was studied by electron microscopy in suckling mouse brain and chick embryo cultured cells. Virus precursors, budding particles, and complete virus particles were first detected 10 hr after infection in chick embryo cells and 24 hr after inoculation in mouse brain. Virus precursors were generally seen aligned along cytomembranes, and were less frequently seen closely associated with viroplasm-like foci, tubular aggregates, or scattered in the cytoplasmic matrix without an apparent connection to an

A new class of indole-based HDAC8 inhibitors as potential anti-lung cancer agents: in silico design, synthesis, biological assessment and binding interaction analysis.

A novel series of indole-based hydroxamic acids was designed, synthesized, and evaluated as promising HDAC8 inhibitors for lung cancer therapy. In silico analyses, including classification-based QSAR, chemical space networks, and scaffold diversity analysis on 2078 HDAC8 inhibitors uncovered essential molecular features for strong HDAC8 potency, informing the design of 12 derivatives. Docking studies ranked the top two candidates, which were subsequently derivatized, synthesized and evaluated through HDAC8 enzymatic and cytotoxicity assays. Several compounds proved highly active, with 6c exhib

Functional insights into nucleoside diphosphate kinases encoded by two ndk paralogs in Waddlia chondrophila.

The Chlamydiota phylum consists of obligate intracellular bacteria, including well-known pathogens and emerging environmental species, with diverse host ranges and metabolic capabilities. Among these bacteria, the gene which encodes nucleoside diphosphate kinase, ndk is present in variable numbers. While most chlamydial species carry a single paralog of ndk, some species have two paralogs. In Waddlia chondrophila, the two Ndk proteins encoded by ndk paralogs retain conserved kinase motifs but differ in subcellular localization, suggesting divergent functional roles. According to localization s

The secreted host-cell protein clusterin interacts with PmpD and promotes Chlamydia trachomatis infection.

Attachment and uptake into host cells are pivotal steps in the life cycle of the Chlamydiaceae, a family of obligate intracellular pathogens. Chlamydia trachomatis (Ctr) possesses a family of nine polymorphic membrane proteins (Pmps), which have been shown to be crucial for adhesion and internalization. However, the host-cell molecules involved have so far remained unknown. Here, we show that a fragment of Ctr PmpD, which forms high-molecular-weight oligomers in solution and adheres to epithelial cells, also binds to secreted clusterin (sCLU), a chaperone-like protein that is secreted into the

Damage-Associated Molecular Patterns (DAMPs) in Retinal Disorders.

Damage-associated molecular patterns (DAMPs) are endogenous danger molecules released from the extracellular and intracellular space of damaged tissue or dead cells. Recent evidence indicates that DAMPs are associated with the sterile inflammation caused by aging, increased ocular pressure, high glucose, oxidative stress, ischemia, mechanical trauma, stress, or environmental conditions, in retinal diseases. DAMPs activate the innate immune system, suggesting their role to be protective, but may promote pathological inflammation and angiogenesis in response to the chronic insult or injury. DAMP…

Impact of Cholesterol Metabolism in Immune Cell Function and Atherosclerosis.

Cholesterol, the most important sterol in mammals, helps maintain plasma membrane fluidity and is a precursor of bile acids, oxysterols, and steroid hormones. Cholesterol in the body is obtained from the diet or can be de novo synthetized. Cholesterol homeostasis is mainly regulated by the liver, where cholesterol is packed in lipoproteins for transport through a tightly regulated process. Changes in circulating lipoprotein cholesterol levels lead to atherosclerosis development, which is initiated by an accumulation of modified lipoproteins in the subendothelial space; this induces significant

Mechanisms of virus dissemination in bone marrow of HIV-1-infected humanized BLT mice.

Immune progenitor cells differentiate in bone marrow (BM) and then migrate to tissues. HIV-1 infects multiple BM cell types, but virus dissemination within BM has been poorly understood. We used light microscopy and electron tomography to elucidate mechanisms of HIV-1 dissemination within BM of HIV-1-infected BM/liver/thymus (BLT) mice. Tissue clearing combined with confocal and light sheet fluorescence microscopy revealed distinct populations of HIV-1 p24-producing cells in BM early after infection, and quantification of these populations identified macrophages as the principal subset of viru
