Genetic diversity among isolates of Paenibacillus larvae from Austria.

Genetic diversity of 214 Paenibacillus larvae strains from Austria was studied. Genotyping of isolates was performed by polymerase chain reaction (PCR) with primers corresponding to enterobacterial repetitive intergenic consensus (ERIC), BOX repetitive and extragenic palindromic (REP) elements (collectively known as rep-PCR) using ERIC primers, BOX A1R and MBO REP1 primers. Using ERIC-PCR technique two genotypes could be differentiated (ERIC I and II), whereas using combined typing by BOX- and REP-PCR, five different genotypes were detected (ab, aB, Ab, AB and alphab). Genotypes aB and alphab …

Intestinal myiasis caused by larvae of Telmatoscopus albipunctatus in a Taiwanese man.

Here we reported the first case in Taiwan of human intestinal myiasis caused by larvae of Telmatoscopus albipunctatus. The 21-year-old male patient experienced gastroenteritis and anal itching. During the patient's first hospital visit the doctor suspected Enterobius infection. After treatment with anthelmintic mebendazole, 5 live larvae were passed out in the feces. Symptoms were resolved a few days later. The larvae were brown and slender, measuring approximately 10 mm in length. The worms were located in the intestine. Subsequent examination showed that the cause was an infestation of larva…

Immunization against trichinellosis using microwaved larvae of Trichinela spiralis.

The present study evaluated the effect of microwave irradiation on infective larvae of Trichinella spiralis (T. spiralis) by the ultrastructure changes of the microwaved larvae (ML) using scanning electron microscope (SEM). The ML tested the ability to immunize mice against a challenge infection with T. spiralis. For the optimal dose and the best route of immunization inducing protection against challenge infection, two doses were used; 300 & 600 ML as one or two-dose regimen, each dose was given orally and intraperitoneally (IP). SEM revealed tegumental damage of the ML in the form of distort…

Unexpected high losses of Anopheles gambiae larvae due to rainfall.

BACKGROUND: Immature stages of the malaria mosquito Anopheles gambiae experience high mortality, but its cause is poorly understood. Here we study the impact of rainfall, one of the abiotic factors to which the immatures are frequently exposed, on their mortality. METHODOLOGY/PRINCIPAL FINDINGS: We show that rainfall significantly affected larval mosquitoes by flushing them out of their aquatic habitat and killing them. Outdoor experiments under natural conditions in Kenya revealed that the additional nightly loss of larvae caused by rainfall was on average 17.5% for the youngest (L1) larvae …

Inflammatory responses to migrating Brugia pahangi third-stage larvae.

Despite being central to parasite establishment and subsequent host pathological and immunologic responses, host-parasite interactions during early third-stage filarial larva (L3) migration are poorly understood. These studies aimed to define early tissue migration of Brugia pahangi L3 in the gerbil (Meriones unguiculatus) and measure host cellular responses during this period. Gerbils were intradermally inoculated in the hind limb with 100 B. pahangi L3, and necropsies were performed at various times. At 3 h, most L3 (96.3%) were recovered from tissues associated with the infection site, with…

Drug analysis in blowfly larvae and in human tissues: a comparative study.

The present study investigates the correlation between concentrations of drugs in human tissues and Diptera larvae feeding on these tissues. Samples of liver were taken from 18 cases in which preliminary toxicological screening indicated the presence of drugs. Blowfly larvae (Diptera: Calliphoridae) were reared on these samples and subsequently analyzed for drug content. Toxicological analyses were carried out using ONLINE Abuscreen (Roche) and GC/MS for available body fluids (blood, urine and bile) as well as liver samples and maggots. All drugs detected in human tissues were also detected in…

Morphological identification of nematode larvae of small ruminants and cattle simplified.

A simplified system has been developed to facilitate identification of nematode larvae of the common nematodes of cattle, sheep and goats. Firstly, in addition to the characteristics conventionally used for identification (such as the shape of the cranial extremity and numbers of intestinal cells), the lengths of the infective sheath tails of infective larvae of each genus/species are related to that of Trichostrongylus spp. instead of using measurements for differentiation. For instance, if the mean length of the sheath tail (the distance the sheath extends caudad beyond the caudal tip of the…

Trichinella encysted larvae in slaughtered pigs in Cairo (1995-1999).

Trichinosis is one of the most serious meat borne parasites. Examination of pigs in the slaughtered houses over the last five years showed an overall rate of 1.691%. The trichinoscope and sometimes the muscle digestion technique were adopted to diagnose infection. Trichinosis as a zoonotic parasitic disease was discussed.

Predilection sites of Trichinella spiralis larvae in naturally infected horses.

A total of 120 muscle tissues from three horses naturally infected with Trichinella spiralis were examined. The head was the most infected site. In particular, the muscles harbouring the highest number of larvae were: musculus buccinator (12, 411 and 1183 larvae g-1), the tongue (11, 615 and 1749 larvae g-1), m. levator labii maxillaris (17,582 and 1676 larvae g-1), and the masseter (4.9, 289 and 821 larvae g-1). Compared with the diaphragm, the number of larvae per gram was from 3.5 to 6.8 times higher in the tongue, from 3.5 to 6.5 higher in m. levator labii maxillaris, and from 2.5 to 4.6 h…

Human infestation with rodent botfly larvae: a new route of entry?

A 3-year-old child was infested with two larvae of the rodent botfly (insect order Diptera, genus Cuterebra), one on the back and one on the neck. Both larvae produced tracklike lesions in the skin. After week 1, the back lesion did not develop further, but the neck lesion continued to enlarge and formed a boil-like lesion. Despite examination by several physicians, the case was not recognized as myiasis for more than 3 weeks. Details of the case are presented and discussed, including the possibility that infestation resulted from direct deposition of eggs or larvae on the patient.

Excretion of ammonia by Lucilia cuprina larvae suppresses immunity in sheep.

Attempts to immunise sheep against natural infestations by Lucilia cuprina larvae have not been effective. Yet it is known that the larvae excrete the immunosuppressant ammonium bicarbonate. The effect of larval ammonium and nonionic ammonia on immunopathobiology was evaluated in 12 infested sheep. The concentration of ammonium in veins draining infested sites was measured in another group of four sheep. Mean jugular unionized ammonia concentration increased 3.5 to 5.6 times above pre-infested control levels. Mean venous ammonium concentrations draining infested sites were 13 times higher than…

Poisoning of cattle and sheep in Uruguay by sawfly (Perreyia flavipes) larvae.

From June to early October of 1993, 1994 and 1995 at least 40 outbreaks of a highly lethal disease occurred in cattle and sheep in the centr … s membranes, ascites and dry content of the omasum, colon and rectum were also observed. Invariably larval body fragments and heads of P flavipes were found in the rumen and omasum. The diagnosis of sawfly poisoning was confirmed by experimental feeding of 3 sheep and 2 calves with 9 to 40 g of P flavipes larvae/kg body weight.

Ultrastructure of the alimentary tract of third-stage larvae of Litomosoides carinii.

The ultrastructural morphology of the alimentary tract of the third-stage larvae of Litomosoides carinii (Nematoda: Filarioidea) is described. The oesophagus possesses the characteristic tri-radiate lumen and is lined by cuticle. Teh lumen of the oesophagus is tightly occluded anteriorly by particulate material and posteriorly at the oesophagointestional junction. Three glands are present in the glandular region, one dorsal and two subventral. These glands contain secretory granules with similar morphological and staining properties. The lumen of the intestine is ill-defined without microvilli…

Detection of Trichinella spiralis muscle larvae in naturally infected horses.

Human trichinellosis outbreaks related to horsemeat consumption have been reported in France and Italy in recent years. In order to determine if Trichinella is present in horses slaughtered at an abattoir in the State of Mexico, diaphragm muscle tissue samples (22-37 g) from 80 horses were examined by artificial digestion. Four of these samples had larvae that were characterized as Trichinella sp. by morphological criteria and as Trichinella spiralis by the polymerase chain reaction.

Large scale collection of viable infective larvae of Loa loa.

A simple method to obtain large numbers of viable infective larvae (L3) of the human filarial parasite Loa loa in an endemic country is described: wild vector flies (Chrysops silacea and C. dimidiata) were caught in large numbers, sealed into pockets of mosquito netting, gently crushed on a glass plate and incubated in a funnel with medium ("Baermann technique"). 91-94% of the L3 actively left the crushed flies and accumulated in the funnel tip after 15 (80%) to 45 minutes (95%). During 111 catching days over a 12 month period, a single fly collector provided approximately 13,000 flies and 23,…

Detection of hookworm and hookworm-like larvae in human fecocultures in Suriname.

Estimation of the prevalence of hookworm infection is usually based on finding eggs of the parasite in the stool. Eggs of the different hookworm species are difficult to distinguish and can be confused with those of hookworm-like species. Proper identification can be made after hatching the eggs and cultivating larvae to the infective stage. We have used the Harada-Mori method to study the stools of Bushnegroes in Suriname and have identified the emerged larvae. Of the 804 individuals examined, 53.6% had positive fecal cultures. The identification of emerged larvae revealed that 75% of the pos…
