Targeted Lipidomic Analysis of Aqueous Humor Reveals Signaling Lipid-Mediated Pathways in Primary Open-Angle Glaucoma.

Primary open-angle glaucoma (POAG) is characterized by degeneration of retinal ganglion cells associated with an increase in intraocular pressure (IOP) due to hindered aqueous humor (AH) drainage through the trabecular meshwork and uveoscleral pathway. Polyunsaturated fatty acids and oxylipins are signaling lipids regulating neuroinflammation, neuronal survival and AH outflow. Among them, prostaglandins have been previously implicated in glaucoma and employed for its treatment. This study addressed the role of signaling lipids in glaucoma by determining their changes in AH accompanying IOP gro

Lipidomic profiling of plasma free fatty acids in type-1 diabetes highlights specific changes in lipid metabolism.

Type-1 diabetes mellitus (T1DM) is associated with metabolic changes leading to alterations in glucose and lipid handling. While T1DM-associated effects on many major plasma lipids have been characterised, such effects on plasma free fatty acids (FFA) have not been fully examined. Using gas chromatography-mass spectrometry, we measured the plasma concentrations of FFA species in individuals with T1DM (n = 44) and age/sex-matched healthy controls (n = 44). Relationships between FFA species and various parameters were evaluated. Plasma concentrations of myristate (14:0), palmitoleate (16:1), pal

Lipidomic Profiles of the Heart and Circulation in Response to Exercise versus Cardiac Pathology: A Resource of Potential Biomarkers and Drug Targets.

Exercise-induced heart growth provides protection against cardiovascular disease, whereas disease-induced heart growth leads to heart failure. These distinct forms of growth are associated with different molecular profiles (e.g., mRNAs, non-coding RNAs, and proteins), and targeting differentially regulated genes has therapeutic potential for heart failure. The effects of exercise on the cardiac and circulating lipidomes in comparison to disease are unclear. Lipidomic profiling was performed on hearts and plasma of mice subjected to swim endurance training or a cardiac disease model (moderate o

Dietary saturated fatty acid type impacts obesity-induced metabolic dysfunction and plasma lipidomic signatures in mice.

Saturated fatty acid (SFA) intake is associated with obesity, insulin resistance, and hepatic steatosis, but scant work examines the impact of SFA type upon these outcomes. We tested the hypothesis that an obesogenic diet prepared with medium chain SFA (MCSFA), mostly as lauric acid-derived from coconut oil, reduces obesity-induced outcomes compared to obesogenic diets prepared with increasing amounts long chain SFA (LCSFA), primarily palmitic acid. Mice were fed (16 weeks) a control, low fat diet or obesogenic diets prepared with differing content of MCSFA or LCSFA in which polyunsaturated an

Genomic, Lipidomic and Metabolomic Analysis of Cyclooxygenase-null Cells: Eicosanoid Storm, Cross Talk, and Compensation by COX-1.

The constitutively-expressed cyclooxygenase 1 (COX-1) and the inducible COX-2 are both involved in the conversion of arachidonic acid (AA) to prostaglandins (PGs). However, the functional roles of COX-1 at the cellular level remain unclear. We hypothesized that by comparing differential gene expression and eicosanoid metabolism in lung fibroblasts from wild-type (WT) mice and COX-2(-/-) or COX-1(-/-) mice may help address the functional roles of COX-1 in inflammation and other cellular functions. Compared to WT, the number of specifically-induced transcripts were altered descendingly as follow

Lipidomic-based investigation into the regulatory effect of Schisandrin B on palmitic acid level in non-alcoholic steatotic livers.

Schisandrin B (SchB) is one of the most abundant bioactive dibenzocyclooctadiene derivatives found in the fruit of Schisandra chinensis. Here, we investigated the potential therapeutic effects of SchB on non-alcoholic fatty-liver disease (NAFLD). In lipidomic study, ingenuity pathway analysis highlighted palmitate biosynthesis metabolic pathway in the liver samples of SchB-treated high-fat-diet-fed mice. Further experiments showed that the SchB treatment reduced expression and activity of fatty acid synthase, expressions of hepatic mature sterol regulatory element binding protein-1 and tumor n

Comparison of electrospray ionization, atmospheric pressure chemical ionization and atmospheric pressure photoionization for a lipidomic analysis of Leishmania donovani.

A comparison of electrospray ionization (ESI), atmospheric pressure chemical ionization (APCI) and atmospheric pressure photoionization (APPI) for the analysis of a wide range of lipids has been performed on standard mixtures and extracts of Leishmania donovani promastigotes resistant to Amphotericin B (AmB). Calibration model, precision, limits of detection and quantification (LOD and LOQ) were assessed for each source. APPI provided the highest signal, signal-to-noise (S/N), and sensitivity for non-polar and low-polarity lipids, while ESI and APCI gave better results for the most polar ones.

Sex-specific lipidomic profiling of the marine mussel Mytilus coruscus: A comparative analysis of male and female lipid metabolism.

Sex-dependent lipid metabolism plays a pivotal role in physiological adaptation and reproduction in marine bivalves, yet its molecular basis remains poorly understood. Here, UPLC-HRMS-based lipidomics was applied to characterize sex-specific lipid profiles in mussels. Females showed higher levels of glycerophospholipids, particularly phosphatidylcholine and phosphatidylethanolamine enriched with polyunsaturated fatty acids, whereas males contained more glycerolipids, especially saturated fatty acid-rich triacylglycerols. Principal component analysis and orthogonal partial least squares-discrim…

Hydrogen gas with hypothermic machine perfusion induces lipidomic alterations in donation-after-cardiac-death rat livers.

Hypothermic machine perfusion (HMP) combined with hydrogen gas has previously been shown to mitigate ischemia-reperfusion injury (IRI) in rat liver, but without full recovery of function. This study examined how hydrogen gas modulates hepatic lipid metabolism during HMP using donation after cardiac death (DCD) rat livers. Untargeted liquid chromatography-mass spectrometry was employed to perform comprehensive lipidomic profiling of liver samples. The analysis results revealed distinct lipid metabolic alterations across cold storage, machine perfusion (MP), and hydrogen-supplemented perfusion (

Diagnostic accuracy of circulating microRNAs and lipidomic biomarkers for early breast cancer detection: A systematic review and meta-analysis.

BACKGROUND: Breast cancer outcomes improve substantially with earlier detection, yet mammography performance can be limited in dense breasts and may lead to false-positive investigations. Circulating microRNAs (miRNAs) and lipidomic/metabolomic signatures have emerged as promising minimally invasive biomarkers that could complement imaging for early-stage detection. The aim of this systematic review and meta-analysis is to evaluate the diagnostic accuracy of circulating microRNAs and lipidomic/metabolomic biomarkers for early breast cancer detection and to explore between-study heterogeneity.

Lipidomic Remodeling by HILPDA in Granulosa Cells Impairs Ovarian Reserve Through Disruption of Mitochondrial and Endoplasmic Reticulum Homeostasis.

Diminished ovarian reserve (DOR) is characterized by a decline in oocyte quantity and/or quality and represents a growing cause of female infertility, particularly among young women, yet its pathogenesis remains incompletely understood. Disordered lipid metabolism impairs folliculogenesis and steroidogenesis, thereby contributing to the pathological changes of DOR. Here, we identify hypoxia-inducible lipid droplet-associated protein (HILPDA), a regulator of lipid droplet biogenesis, as a potential contributor to DOR. We found that HILPDA was significantly upregulated in granulosa cells (GCs) f

HDL-mimetic peptide treatment reverses APOE4-induced transcriptomic and lipidomic alterations in the brain of humanized APOE mice.

INTRODUCTION: The apolipoprotein E4 (APOE4) allele is the strongest genetic risk factor for late-onset Alzheimer's disease (AD). ApoE4 has reduced lipidation capacity and impaired lipid transport, disrupting neuronal maintenance. The high-density lipoprotein (HDL)-mimetic peptide 4F offers a potential therapeutic strategy. METHODS: To investigate how APOE4 alters brain gene expression and lipid metabolism and to evaluate the therapeutic potential of 4F, we performed dual-omics analysis in APOE4/4 and APOE3/3 mice treated intraperitoneally with D-enantiomer of 4F (D4F) or vehicle for 12 weeks

Spatial lipidomic profiling of cutaneous melanoma using electroporation-based biopsy reveals subtype-specific metabolic alterations.

Cutaneous melanoma is a highly aggressive skin cancer with rising global incidence. Although understanding melanoma metabolism requires spatially resolved molecular information, conventional excisional sampling offers limited access to spatial gradients within tumors. Here, we introduce a handheld electroporation-based biopsy (e-biopsy) device, a newly engineered tool that delivers controlled pulsed electric fields to transiently permeabilize cells and extract intracellular molecules directly from tissue. This minimally invasive, probe-based approach enables rapid, localized molecular harvesti

Lipidomic adaptation and membrane physical changes underlie tamoxifen resistance in breast cancer: molecular dynamics simulation study.

Drug resistance in breast cancer (BC) is strongly linked to plasma membrane lipid remodeling, notably the loss of lipid asymmetry and elevated cholesterol (CHL) and sphingomyelin (SM) levels. We performed multi-scale molecular dynamics (MD) simulations on eight distinct lipid bilayer models-ranging from normal asymmetric (N1, N2) to cancer-mimetic symmetric states (C1-C5)-at all-atom and coarse-grained resolutions to investigate their impact on tamoxifen (TAM) permeation. Unbiased MD and umbrella sampling were used to compute potential of mean force (PMF) profiles. Our results indicate that me
