Early course of microcirculatory perfusion in eye and digestive tract during hypodynamic sepsis.

INTRODUCTION: The aim of the study was to evaluate and compare the microcirculatory perfusion during experimental sepsis in different potentially available parts of the body, such as sublingual mucosa, conjunctiva of the eye, and mucosa of jejunum and rectum. METHODS: Pigs were randomly assigned to sepsis (n = 9) and sham (n = 4) groups. The sepsis group received a fixed dose of live Escherichia coli infusion over a 1-hour period (1.8 × 10(9)/kg colony-forming units). Animals were observed 5 hours after the start of E. coli infusion. In addition to systemic hemodynamic assessment, we performe

Quantitative photoacoustic evaluation of graded ischemic stroke and the therapeutic efficacy of low-intensity transcranial ultrasound stimulation.

Ischemic stroke represents a leading cause of global mortality and long-term disability. Consequently, rapid quantification of cerebral damage severity, coupled with timely neuromodulatory intervention, is imperative for improving clinical prognosis. In this study, we present a label-free quantitative monitoring framework utilizing optical-resolution photoacoustic microscopy (OR-PAM) to assess microvascular alterations and evaluate the therapeutic efficacy of low-intensity transcranial ultrasound stimulation (LITUS). A graded ischemic stroke model was established by modulating photothrombotic

Retinal Biomarkers of Folate and Vitamin B12 Metabolic Dysfunction: A Framework for Machine Learning-Assisted Detection of Cerebral Folate Deficiency.

Disruptions in folate (vitamin B9) and vitamin B12 metabolism, including nutritional deficiency, folate receptor alpha (FRα) autoantibodies, and MTHFR/DHFR polymorphisms, impair one-carbon metabolism and produce measurable retinal structural, microvascular, and functional changes, offering a non-invasive window into systemic and cerebral metabolic dysfunction, particularly cerebral folate deficiency (CFD). This review synthesizes peer-reviewed evidence on retinal alterations linked to folate/B12 deficiency, hyperhomocysteinemia, FRα autoantibody syndromes, and MTHFR/DHFR variants, alongside ar

Rapid manufacturing of angiogenic cellular collagen patches for ischemic cardiomyopathy.

BACKGROUND: One in ten Americans carry a lifetime risk of ischemic heart failure, the most severe form of ischemic heart disease. Carrying a nearly 50% five‑year mortality rate, no interventional therapy exists to treat the underlying cause, microvascular malperfusion. In efforts to combat microvascular malperfusion, our group has utilized synergistic application of endothelial progenitor cells (EPCs) and smooth muscle cells (SMCs) to induce angiogenesis in ischemic myocardium. METHODS: Cells are then embedded into a rapidly manufacturable compressed collagen (CC) patch to provide a biosimila

Morphologic Mapping of the Sublingual Microcirculation in Healthy Volunteers.

PURPOSE: Monitoring the sublingual and oral microcirculation (SM-OM) using hand-held vital microscopes (HVMs) has provided valuable insight into the (patho)physiology of diseases. However, the microvascular anatomy in a healthy population has not been adequately described yet. METHODS: Incident dark field-based HVM imaging was used to visualize the SM-OM. First, the SM was divided into four different fields; Field-a (between incisors-lingua), Field-b (between the canine-first premolar-lingua), Field-c (between the first-second premolar-lingua), Field-d (between the second molar-wisdom teeth-l

OBJECTIVES: Systemic lupus erythematosus (SLE) is a chronic autoimmune disease with potential vascular complications. Ultrawide-field swept-source optical coherence tomography angiography (SS-OCTA) may serve as a non-invasive tool to assess retinal and choroidal microvasculature. We aimed to evaluate its utility in SLE for early detection and monitoring of SLE vasculopathy. METHODS: 142 eyes of 71 SLE patients (median age 35 years, IQR 23-42) and 194 eyes of 97 healthy controls (median age 33 years, IQR 26-39.25) underwent ultrawide-field SS-OCTA imaging to quantify vascular density (VD) in r

Virtual fluorescent labeling of engineered vascular networks with embedded tracer particles.

Functional microvascular networks in engineered tissues depend on coordinated endothelial-stromal interactions and evolving extracellular matrix (ECM) mechanics, yet fluorescent staining precludes longitudinal studies and is incompatible with repeated particle-based microrheology and traction force measurements. We develop a deep-learning virtual labeling approach that recovers nuclear, cytoskeletal, and endothelial fluorescence from label-free images acquired in fibrin scaffolds. Human umbilical vein endothelial cells and lung fibroblasts were co-cultured in three-dimensional (3D) fibrin hydr

Three dimensional evaluation of cerebrovascular density and branching in chronic traumatic encephalopathy.

Chronic traumatic encephalopathy (CTE) is a neurodegenerative disease associated with exposure to repetitive head impacts (RHI) and characterized by perivascular accumulations of hyperphosphorylated tau protein (p-tau) at the depths of the cortical sulci. Studies of living athletes exposed to RHI, including concussive and nonconcussive impacts, have shown increased blood-brain barrier permeability, reduced cerebral blood flow, and alterations in vasoreactivity. Blood-brain barrier abnormalities have also been reported in individuals neuropathologically diagnosed with CTE. To further investigat

New insights into systemic sclerosis related microcirculatory dysfunction by assessment of sublingual micr\ocirculation and vascular glycocalyx layer. Results from a preliminary study.

BACKGROUND: Initial morphological and functional markers of systemic sclerosis (SSc) are evidenced in microvascular structural damage. However, nailfold videocapillaroscopy (NVC) explores only morphological abnormalities. Sidestream Dark Field (SDF) imaging of sublingual microcirculation enables assessment of both morphological and functional capillary impairment and allows measurement of the glycocalyx layer, which is an indicator of endothelial dysfunction. OBJECTIVE: To describe and validate sublingual abnormalities assessed by SDF device in comparison with NVC findings and to measure the
