[Pathogenic spectrum of enteroviruses associated with hand, foot and mouth disease by a GeXP™-based multiplex reverse transcription-PCR assay in Jinan, China, 2009-2012].

We investigated the pathogenic spectrum of enteroviruses associated with hand, foot and mouth disease (HFMD) in Jinan, China. A total of 274 specimens with a clinical diagnosis of HFMD in Jinan from 2009 to June 2012 were used. A GenomeLab™ (GeXP)-based multiplex reverse transcription-polymerase chain reaction (RT-PCR) assay was employed to simultaneously detect 15 serotypes of human enteroviruses: human enterovirus (EV)71; coxsackievirus A (CVA)16, 4, 5, 6, 9, and 10; CVB1, 3 and 5; echovirus (Echo) 6, 7, 11, 13 and 19. Results showed that all samples were enterovirus-positive, with the most

Evidence of presence of Mycobacterium tuberculosis in bovine tissue samples by multiplex PCR: possible relevance to reverse zoonosis.

Bovine tuberculosis, caused by Mycobacterium bovis, remains one of the most important zoonotic health concerns worldwide. The transmission of Mycobacterium tuberculosis from humans to animals also occurs especially in countries where there is close interaction of humans with the animals. In the present study, thirty bovine lung tissue autopsy samples from an organized dairy farm located in North India were screened for the presence of Mycobacterium tuberculosis complex by smear microscopy, histopathological findings and PCR. Differential diagnosis of M. tuberculosis and M. bovis was made based

Rapid multiplex PCR assay to identify respiratory viral pathogens: moving forward diagnosing the common cold.

Upper respiratory tract infections (URIs) can be a serious burden to the healthcare system. The majority of URIs are viral in etiology, but definitive diagnosis can prove difficult due to frequently overlapping clinical presentations of viral and bacterial infections, and the variable sensitivity, and lengthy turn-around time of viral culture. We tested new automated nested multiplex PCR technology, the FilmArray(®) system, in the TAMC department of clinical investigations, to determine the feasibility of replacing the standard viral culture with a rapid turn-around system. We conducted a feas

A one-tube autocatalytic transcription-driven CRISPR cascade for ultrasensitive mRNA detection.

The rapid identification and precise quantification of cancer biomarkers are essential for the purposes of diagnosis, classification, and therapeutic intervention. Traditional molecular diagnostic methodologies, such as polymerase chain reaction (PCR), provide considerable sensitivity; however, they depend on exponential amplification and advanced instrumentation, thereby constraining their applicability for point-of-care testing. In this study, we developed a Transcription-driven CRISPR Cascade Amplification (TCCA) for one-tube detection of mRNA at the concentration of 0.6 copies/μL within ju

Echinococcus multilocularis--adaptation of a worm egg isolation procedure coupled with a multiplex PCR assay to carry out large-scale screening of red foxes (Vulpes vulpes) in Norway.

Echinococcus multilocularis, causing alveolar echinococcosis in humans, is a highly pathogenic emerging zoonotic disease in central Europe. The gold standard for the identification of this parasite in the main host, the red fox, namely identification of the adult parasite in the intestine at necropsy, is very laborious. Copro-enzyme-linked immunosorbent assay (ELISA) with confirmatory polymerase chain reaction (PCR) has been suggested as an acceptable alternative, but no commercial copro-ELISA tests are currently available and an in-house test is therefore required. Published methods for taeni

[Very rare and life-threatening complications of bocavirus bronchiolitis: pneumomediastinum and bilateral pneumothorax].

Human bocavirus (HBoV), that was first identified in 2005 and classified in Parvoviridae family, is a small, non-enveloped, single-stranded DNA virus, responsible for upper and lower respiratory tract infections, especially in young children. Although HBoV generally causes self-limited influenza-like illness, it may also lead to pneumonia, bronchiolitis, croup and asthma attacks. In this report, a case of acute bronchiolitis complicated with pneumomediastinum and bilateral pneumothorax caused by HBoV has been presented. A three-year-old boy was referred to our pediatric intensive care unit wit

Evaluation of a novel multiplex RT-qPCR assay for the quantification of leukemia-associated BCR-ABL1 translocation.

Although monitoring of BCR-ABL1 translocation has become an established practice in the management of chronic myeloid leukemia (CML), the detection limit of the BCR-ABL1 transcripts needs more standardization. The aim of the present study was to evaluate the clinical performances of a novel assay for the quantification of BCR-ABL1 fusion transcripts (e13a2 and e14a2) and ABL1 in a single reaction. This assay is based on the real-time reverse transcription polymerase chain reaction (RT-qPCR) in multiplex format. In a retrospective comparative clinical study performed in a reference laboratory,

Circulation and in vivo distribution of duck hepatitis A virus types 1 and 3 in infected ducklings.

The circulation of duck hepatitis A virus types 1 (DHAV-1) and 3 (DHAV-3) in Southeast Asia has resulted in a continuously changing epidemiological scenario. In this study, a duplex real-time PCR assay for simultaneous quantitative detection of DHAV-1 and DHAV-3 was established, and 200 liver samples from dead ducklings collected from 31 different flocks in Shandong province, China, were tested. Fifty-eight (29.0 %) samples from 13 flocks were positive for DHAV-1 single infection, 113 (56.5 %) samples from 13 other flocks were positive for DHAV-3 single infection, and 24 samples (12.0 %) from

A novel quantitative multiplex real-time RT-PCR for the simultaneous detection and differentiation of West Nile virus lineages 1 and 2, and of Usutu virus.

An increase in activity of two mosquito-borne flaviviruses, West Nile virus (WNV) and Usutu virus (USUV), has been reported in Europe in recent years. The current epidemiological situation calls for RT-PCR methods that are able to detect not only the widespread lineage 1 (L1) WNV, but also lineage 2 (L2) WNV. In addition, the presence in Europe of the closely related USUV requires methods that can identify these three flaviviruses and permit an efficient and accurate differential diagnosis. Here we describe a new one-step real-time multiplex RT-PCR that detects and differentiates efficiently W

Spontaneous Regression of a Retinal Mass With Epstein-Barr Virus-Positive and Cytomegalovirus-Positive Panuveitis in Methotrexate-Associated Lymphoproliferative Disorder.

Methotrexate (MTX) has been widely used for patients with autoimmune diseases as a corticosteroid-sparing agent. We here report an 86-year-old Japanese woman treated with MTX for rheumatoid arthritis who presented with panuveitis along with a yellowish retinal mass. Both Epstein-Barr virus (EBV) and cytomegalovirus (CMV) were detected in aqueous humor and vitreous fluid by quantitative polymerase chain reaction. The retinal mass gradually regressed over eight months after MTX withdrawal. The present case indicates that intraocular MTX-lymphoproliferative disorder (MTX-LPD) should be considered

Beyond the Central Nervous System: Neisseria meningitidis as an Unusual Cause of Urogenital Infection.

Neisseria meningitidis is a well-known cause of invasive diseases such as meningitis and septicemia, but is increasingly recognized as an emerging urogenital pathogen. We report the case of a 36-year-old man who presented to a tertiary hospital in Riyadh, Saudi Arabia, with persistent dysuria and purulent urethral discharge. Despite initial treatment for nonspecific urethritis and subsequent management for chronic prostatitis with levofloxacin and azithromycin, his symptoms persisted. Although standard multiplex polymerase chain reaction (PCR) assays for common sexually transmitted infections …

Masquerading Yeast: A Case Report of Lomentospora prolificans Fungemia With a Diagnostic Twist.

Lomentospora prolificans is an environmental mold that is increasingly recognized as an opportunistic pathogen, particularly in immunocompromised hosts. Here we present a case of disseminated Lomentospora in a patient with refractory acute myeloid leukemia. In general, disseminated mold infections can be challenging to diagnose. The diagnosis in this case was delayed due to a false positive direct from a blood culture multiplex polymerase chain reaction result for Candida tropicalis due to amplification of nonviable DNA and misidentification of conidia as yeast forms on gram stain. The Lomento…

Saliva and Anterior Nasal Samples for Detecting Respiratory Viruses in Children.

For detecting respiratory viruses, a nasopharyngeal swab sample for polymerase chain reaction (PCR) assays has been the benchmark. Less discomfort related to sampling is requested especially for children, and thus other sample types have been suggested. We examined anterior nasal samples and saliva samples of 59 pediatric patients with respiratory symptoms and compared the results to nasopharyngeal swab samples. The collection of saliva and anterior nasal samples was tolerated better than nasopharyngeal samples in children. Anterior nasal samples were more accurate than saliva samples in detec

Alpha-thalassaemia trait as a cause of unexplained microcytosis in a South African population.

BACKGROUND: Red cell microcytosis is a common abnormality detected in a full blood count, which often prompts clinicians to investigate further for a cause. In the absence of iron deficiency and anaemia of chronic disease, the differential diagnosis includes β-thalassaemia trait and α-thalassaemia trait. METHODS: We investigated the contribution of α-thalassaemia trait in South African subjects with unexplained microcytosis. Iron studies, haemoglobin subfractionation and multiplex polymerase chain reaction (PCR) analysis for α-globin gene deletions were performed on 97 controls and 86 patient…

Emphysematous rumenitis in a Japanese black calf caused by Clostridium perfringens with secondary Balantidium coli infection.

A 4-month-old Japanese black calf exhibited anorexia and respiratory distress. Necropsy revealed a spongiform appearance of the rumen wall and histologically diagnosed as emphysematous rumenitis. The ruminal mucosa exhibited characteristic lesions of chemical rumenitis, so ruminal acidosis is suspected. Gram-positive bacilli were discernible on the degenerated ruminal mucosal surface and reacted with Clostridium species antibodies. Anaerobic cultures yielded Clostridium perfringens (C. perfringens), identified by multiplex polymerase chain reaction (PCR) as C. perfringens type A. The lumen and…
