Lats2 deficiency protects the heart against myocardial infarction by reducing inflammation and inhibiting mitochondrial fission and STING/p65 signaling.

Large tumor suppressor kinase 2 (Lats2) is a member of the Hippo pathway, a critical regulator of organ size. Since Lats2 activity may trigger mitochondrial dysfunction, a key pathogenic factor in acute myocardial infarction (AMI), this study sought to investigate whether Lats2 deletion confers cardioprotection in AMI. AMI was induced in cardiomyocyte-specific Lats2 knockout (Lats2Cko) and control (Lats2flox) mice. Twenty-eight days after AMI surgery, myocardial performance and mitochondrial homeostasis were impaired in Lats2floxmice. In contrast, Lats2Cko mice exhibited markedly preserved car

Porcine Circovirus type 2 infected myocardial tissue transcriptome signature.

The goal of this study was to compare the global gene expression profile in cardiac tissues of pig infected with porcine circovirus 2 (PCV2) to that of healthy cells. Since PCV2 infection causes severe cardiovascular lesions, the myocardial tissue model was chosen for this study. In High-throughput transcriptome analysis, DESeq2 and CLC genomics workbench analyses revealed a total of 196 significantly differentially expressed genes (DEGs) (p-value < 0.05). Furthermore, 194 transcripts were upregulated, while only two were downregulated (HSPA6 and DNAJA1), with fold changes ranging from 16.293

The heart of cardiac reprogramming: The cardiac fibroblasts.

Cardiovascular disease is the leading cause of death worldwide, outpacing pulmonary disease, infectious disease, and all forms of cancer. Myocardial infarction (MI) dominates cardiovascular disease, contributing to four out of five cardiovascular related deaths. Following MI, patients suffer adverse and irreversible myocardial remodeling associated with cardiomyocyte loss and infiltration of fibrotic scar tissue. Current therapies following MI only mitigate the cardiac physiological decline rather than restore damaged myocardium function. Direct cardiac reprogramming is one strategy that has p

Calenduloside E protects against myocardial ischemia-reperfusion injury induced calcium overload by enhancing autophagy and inhibiting L-type Ca2+ channels through BAG3.

Calenduloside E (CE) is a saponin isolated from Aralia elata (Miq) Seem, which has anti-cardiovascular disease effects. This study aims to evaluate the anti-myocardial ischemia-reperfusion injury (MIRI) mechanisms of CE and regulation of BAG3 on calcium overload. We adopted siRNA to interfere with BAG3 expression in H9c2 cardiomyocytes and used adenovirus to interfere with BAG3 expression (Ad-BAG3) in primary neonatal rat cardiomyocytes (PNRCMs) to clarify the role of BAG3 in mitigating MIRI by CE. The results showed that CE reduced calcium overload, and Ad-BAG3 had a significant regulatory ef

Unexpected death of a young woman: is myocardial bridging significant?--A case report and review of literature.

Myocardial bridging (MB) refers to a congenital condition where a segment of a major coronary artery courses within the myocardium for a variable distance, and has been reported as an incidental finding in many cases. However, in some situations it has been shown to be associated with acute coronary syndromes, arrhythmias, myocardial ischemia and sudden death. We present the case of a 26-year-old Chinese woman with no previous history of disease who died unexpectedly in hospital. She had a 'common cold' with its associated symptoms. Autopsy revealed the presence of MB of the left anterior desc

Nongeometric quantitative assessment of right and left ventricular function: myocardial performance index in normal children and patients with Ebstein anomaly.

Assessment of ventricular systolic function has been based on the geometric models of ventricular shape. This study was designed to define normal values for a nongeometric myocardial performance index (MPI) in children and to evaluate the utility of MPI in congenital heart disease. The MPI measures the ratio of total time spent in isovolumic activity (isovolumic contraction time and isovolumic relaxation time) to the ejection time. The right ventricular (RV) and left ventricular (LV) MPI were measured in 152 normal children (ages 3 to 18 years) and 45 preoperative patients with Ebstein anomaly

The relationship between prolapsing mitral leaflet syndrome and angina and normal coronary arteriograms.

Patients with a prolapsing mitral leaflet frequently have chest pain while their coronary arteriograms are normal. In this regard, these patients are similar to the group of patients with angina and normal coronary arteriograms. In the present study, clinical, electrocardiographic, cardiac hemodynamic, angiographic, and metabolic findings in 20 patients with a prolapsing mitral leaflet were compared to those of 16 patients with angina and normal coronary arteriograms. Except for the presence of mitral leaflet prolapse and systolic clicks, the findings were similar in both groups. We postulate

Systolic time intervals as a measure of left ventricular function in viscose rayon workers exposed to carbon disulfide.

In a group of viscose rayon workers exposed to carbon disulfide the systolic time intervals were calculated. They were determined from the simultaneous recording of an ECG, a phonocardiogram, and arterial pulse tracings. Results indicate a shortened left ventricular ejection time (lvet) and a prolonged isovolumetric contraction time (ICT), together with an increase in the ratio of ICT to LVET. These alterations, which occurred in the absence of any clinical signs or symptoms of heart disease, are interpreted as a consequence of an impaired left ventricular contractility, comparable to mild cor

Low use of statins for secondary prevention in primary care: a survey in a northern Swedish population.

BACKGROUND: Cholesterol-lowering therapy with statins is recommended in established cardiovascular disease (CVD) and should be considered for patients at high cardiovascular risk. We surveyed statin treatment before first-time myocardial infarction in clinical practice compared to current guidelines, in patients with and without known CVD in primary care clinics with general practitioners (GPs) on short-term contracts vs. permanent staff GPs. METHODS: A total of 931 patients (345 women) in northern Sweden were enrolled in the study between November 2009 and December 2014 and stratified by pri

Visualization of regenerating and repairing hearts.

With heart failure continuing to become more prevalent, investigating the mechanisms of heart injury and repair holds much incentive. In contrast with adult mammals, other organisms such as teleost fish, urodele amphibians, and even neonatal mammals are capable of robust cardiac regeneration to replenish lost or damaged myocardial tissue. Long-term high-resolution intravital imaging of the behaviors and interactions of different cardiac cell types in their native environment could yield unprecedented insights into heart regeneration and repair. However, this task remains challenging for the he

Cardiac Memory - from Theory to Clinical Practice.

Cardiac memory (CM) is defined as changes in T wave polarity and vector that appear after cessation of a period of abnormal ventricular depolarization of various causes. The mechanisms responsible for CM development are initiation by local stretch, requiring myocardial contraction, followed by a cascade of intracellular signals that lead to a reduction in repolarization currents, especially Ito. In practice, CM is a frequently encountered ECG phenomenon, especially in patients with intermittent ventricular pacing, and knowledge of the ECG pattern of CM may help quick differential diagnosis fro…

[Effects of ventricular hypertrophy following experimental renovascular hypertension on the elasticity of the papillary muscle].

A precise method was developed for detecting muscular stiffness changes in the left ventricular hypertrophy by chronic pressure overload in rats (2 kidney-1 clip renal hypertensive rats). The technique of Quick-Release allowed the measurement of the "stress-strain" relation (delta-epsilon), the elastic stiffness (d delta/d epsilon) and the stiffness constant (K) in isolated papillary muscle from normal and hypertrophied hearts at rest and at the maximal time course of activation during isometric contraction. (Table: see text). The results of this study reported that the cardiac hypertrophy ind

Beta blockade and the effects of stress on the normal and ischaemic heart.

There is evidence that the beta blocking drugs are useful in angina pectoris, inhibiting the tachycardia of exercise and other forms of stress, such as emotion. In myocardial infarction, there is experimental evidence in animals and man for the effectiveness of these drugs in restoring the supply/demand balance of the ischaemic areas, and thus limiting infarct size. They may act partly by redistributing the coronary blood flow from areas of relatively good perfusion to those less well supplied. Cardiac pain is reduced in angina and also post-infarction. In contrast to the usual haemodynamic ef

[Changes in serum and urinary magnesium during and after extracorporeal circulation in cardiac surgery in children].

Magnesium plays an essential role in the processes of myocardial contraction and conduction. In cardiovascular surgery, extracorporeal circulation (ECC) in haemodilution results in various electrolyte distrubances, with invariably hypomagnesaemia. A group of 13 children undergoing open heart surgery was compared with a group of 17 children operated upon under the same conditions but with the addition of magnesium to the filling solution for the extracorporeal circulation. Changes in blood and urinary electrolytes during and after ECC were studied. Hypomagnesaemia was seen in both groups after …

Use of a QDOT MICRO catheter to identify a premature ventricular contraction origin in the right ventricular anterior papillary muscle in a repaired truncus arteriosus.

The ventricular papillary muscles (VPMs) can be a source of premature ventricular contractions (PVCs). Catheter ablation of VPM PVCs is challenging because of the anatomical complexity, such as the apical structures in proximity to the ventricular walls. The QDOT MICRO catheter (Biosense Webster, Diamond Bar, CA, USA) has microelectrodes embedded along the circumference of its distal tip and can provide information on which side of its tip myocardial activation is earlier. This repaired truncus arteriosus case demonstrates the usefulness of the microelectrode recording in identifying a PVC ori

Paediatric chest wall trauma causing delayed presentation of ventricular arrhythmia.

This report describes a paediatric patient presenting with haemodynamically stable non-sustained ventricular tachycardia 1 day after minor blunt chest trauma. Initial laboratory studies, chest X-ray and echocardiography were normal; however, cardiac MRI revealed precordial haematoma, myocardial contusion and small pericardial effusion. Throughout her hospital course, she remained asymptomatic aside from frequent couplets and triplets of premature ventricular contractions. Ectopy was controlled with oral verapamil. This case highlights how significant cardiac injury may be missed with standard

Physics-Informed Reconstruction of Transmural Myocardial Deformation from 3D Echocardiography: Validation Against Cardiac MRI.

PURPOSE: Quantitative assessment of myocardial deformation is increasingly important in clinical cardiology, yet conventional two-dimensional (2D) echocardiography and standard three-dimensional (3D) approaches remain limited by out-of-plane motion and incomplete characterization of transmural mechanics. To address these limitations, we introduce a physics-informed framework for 3D echocardiography that reconstructs the full finite strain tensor across the entire myocardial wall. As an initial methodological study, we demonstrate the framework and validate it against cardiac magnetic resonance

Breaking the Heat Tolerance Response: The Crucial Role of MFAP4 in Combating Heat-Induced Cardiac Injury.

Heat exposure increases cardiac workload and impairs thermoregulation, leading to cardiac injury and heatstroke (HS). The involvement of Microfibrillar-associated protein 4 (MFAP4) in HS remains unclear. This study aims to investigate the role and mechanisms of MFAP4 in heat-induced damage. Whole body MFAP4-knockout (MFAP4-KO) mice and MFAP4-overexpressing cells were generated. Physiological and pathological responses were assessed in vivo/vitro experiments. An HS model exposed mice to (39.5 ± 0.5)°C/(55 ± 5)% humidity; and cell to 44°C with 5% CO₂ for 2 h. Transcriptomic analysis was performe
