Evaluation of IL-31 and IL-31RA Expression in Lichen Planopilaris: A Retrospective Immunohistochemistry Study.

BACKGROUND: Lichen planopilaris (LPP) is an autoimmune cicatricial alopecia leading to irreversible follicular destruction. Interleukin-31 (IL-31) and its receptor complex (IL-31RA/OSMRβ) are implicated in neurogenic inflammation and pruritus in inflammatory dermatoses, including lichen planus. This study aimed to evaluate IL-31/IL-31RA expression in LPP and to investigate their associations with disease extent and pruritus. METHODS: Thirty patients with classical-type LPP and 30 age- and sex-matched controls were retrospectively analyzed. Disease extent and pruritus severity were assessed us…

Growth hormone-releasing hormone attenuates amyloid deposition and neuroinflammation in Alzheimer's disease models.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by amyloid-β (Aβ) accumulation, tau hyperphosphorylation, neuroinflammation, and synaptic loss. Existing therapies provide only modest symptomatic relief and fail to slow disease progression. Beyond its role in promoting pituitary growth hormone (GH) secretion, growth hormone-releasing hormone (GHRH) has shown neuroprotective effects in experimental ischemic stroke and spinal muscular atrophy. Here, we explored the therapeutic potential of GHRH and its agonist MR-409 in AD models. In vitro, GHRH(1-44)NH₂ promote

Antinociceptive, anti-inflammatory, and antiophidic potential of Punica granatum leaves extract and fraction: an in vitro and in vivo evaluation.

Punica granatum (Punicaceae) is a shrub with a worldwide distribution that has been used in folk medicine for the treatment of inflammatory disorders. The study investigated the therapeutic potential of spray-dried hydroethanolic extract (SDE) and spray-dried ethyl acetate fraction (EAF) from Punica granatum leaves in models of pain, inflammation, and snakebite envenomation. Phytochemical analysis highlighted the presence of polyphenols, particularly luteolin and hydrolysable tannins in SDE, and a significant concentration of flavonoids in EAF. Both samples demonstrated safety in murine fibrob

Melatonin as an anti-inflammatory hormone bridging migraine relief and cancer immunity enhancement: a literature review.

Melatonin, once relegated to the circadian periphery, has resurfaced as a pleiotropic immunomodulator capable of quelling neurogenic inflammation while invigorating antitumour defence. Migraine-and its disabling, episodic neurovascular pain-shares an "inflammatory genome", defined here as a conserved danger-response gene set (NF-κB, NLRP3, IL1B, NOS2), with the metabolic chaos that subdues cytotoxic immunity in solid cancers; both ignite NF-κB, NLRP3 and reactive-oxygen cascades that erode tissue homeostasis. Emerging evidence shows that endogenous melatonin declines precede migraine attacks, …

Overexpressing neurogenic differentiation factor 1 in Müller cells improves retinal function after optic nerve crush injury in adult mice.

Optic nerve injury leads to axonal degeneration and the death of retinal ganglion cells, which ultimately causes vision loss. Notably, current treatments are limited. In the present study, we explored whether neurogenic differentiation factor 1 (NeuroD1 or ND1) overexpression in retinal Müller cells may repair the retina after optic nerve crush in mice. Adult mice were subjected to optic nerve crush followed by intravitreal AAV-7m8-GFAP-GFP-ND1 virus injection. Immunofluorescent staining, multi-electrode array recording, electroretinogram, and visual behavior tests were then performed to exami

Functional Assessment of Genetically Modified Infrapatellar Fat Pad Mesenchymal Stem/Stromal Cell-Derived Extracellular Vesicles (EVs): Potential Implications for Inflammation/Pain Reversal in Osteoarthritis.

Osteoarthritis (OA) is a debilitating joint disease affecting over 500 million people globally, characterized by cartilage degradation, chronic pain, and failed tissue repair. Neurogenic inflammation, driven by neuropeptides including Substance P (SP) and calcitonin gene-related peptide (CGRP), plays a key role in the pathogenesis of OA. This study explores the therapeutic potential of extracellular vesicles (EVs) derived from infrapatellar fat pad mesenchymal stem/stromal cells (IFP-MSCs) transduced with CGRP antagonist CGRP8-37 (aCGRP IFP-MSC EVs). These EVs are enriched in anti-inflammatory…

Variability of autonomic nerve activity in dry eye with decreased tear stability.

The autonomic nervous system plays a crucial role in the maintenance of homeostasis. Neurogenic inflammation due to decreased stability of tear film may induce changes in autonomic nerve activity, which could be associated with symptom expression. This study aimed to measure biological parameters that represent autonomic nerve activity in dry eye (DE)s caused by tear film dysfunction and investigate their relationship with symptom intensity. This prospective, cross-sectional, comparative study evaluated 34 eyes of 34 participants (mean age: 52.5 ± 13.4 years; range: 20-81 years) without keratoc…

Solitary chemosensory cells are innervated by trigeminal nerve endings and autoregulated by cholinergic receptors.

BACKGROUND: Solitary chemosensory cells (SCCs) in the murine nasal epithelium are discrete specialized cells that respond to irritants and activate trigeminal nerve fibers through the release of acetylcholine (ACh), resulting in local neurogenic inflammation. In addition to releasing ACh, SCCs are the exclusive epithelial source of interleukin (IL)-25. In humans, SCCs are significantly expanded in sinonasal polyps (NPs). However, the SCC-trigeminal synapse has yet to be demonstrated in human sinonasal epithelium. METHODS: Immunofluorescence for trigeminal nerve fiber markers, nicotinic ACh re…

Role of neurokinin type 1 receptor in nociception at the periphery and the spinal level in the rat.

OBJECTIVES: Noxious stimuli activate small to medium-sized dorsal root ganglion (DRG) neurons. Intense noxious stimuli result in the release of substance P (SP) from the central terminals of these neurons. It binds to the neurokinin type 1 receptor (NK1r) and sensitises the dorsal horn neurons. SP is also released from the peripheral terminals leading to neurogenic inflammation. However, their individual contribution at spinal and peripheral levels to postincisional nociception has not been delineated as yet. METHODS: Sprague-Dawley rats were administered different doses (3-100 μg) of an NK1r…

The neurochemical changes in the innervation of human colonic mesenteric and submucosal blood vessels in ulcerative colitis and Crohn's disease.

BACKGROUND: Neurogenic inflammation involves vasodilation, oedema and sensory nerve hypersensitivity. Extrinsic sensory nerves to the intestinal wall mediate these effects and functional subsets of these extrinsic nerves can be characterized by immunohistochemical profiles. In this study such profiles were examined in samples from patients with inflammatory bowel disease (IBD), in particular ulcerative colitis (UC) and Crohn's disease (CD). METHODS: Healthy margins from cancer patients were compared to specimens from IBD patients. All nerve fibres were labelled by PGP 9.5. Double and triple l…

A substance P antagonist increases brain intracellular free magnesium concentration after diffuse traumatic brain injury in rats.

OBJECTIVE: Magnesium (Mg) deficiency has been shown to increase substance P release and induce a pro-inflammatory response that can be attenuated with the administration of a substance P-antagonist. Neurogenic inflammation has also been implicated in traumatic brain injury (TBI), a condition where brain intracellular free magnesium (Mg(f)) decline is known to occur and has been correlated with functional outcome. We therefore examined whether a substance P antagonist restores brain intracellular free magnesium concentration following TBI. METHODS: Male, adult Sprague-Dawley rats were injured …
