Reconstruction of the mouse extrahepatic biliary tree using primary human extrahepatic cholangiocyte organoids.

The treatment of common bile duct (CBD) disorders, such as biliary atresia or ischemic strictures, is restricted by the lack of biliary tissue from healthy donors suitable for surgical reconstruction. Here we report a new method for the isolation and propagation of human cholangiocytes from the extrahepatic biliary tree in the form of extrahepatic cholangiocyte organoids (ECOs) for regenerative medicine applications. The resulting ECOs closely resemble primary cholangiocytes in terms of their transcriptomic profile and functional properties. We explore the regenerative potential of these organ…

Human colon organoid Transwell model for investigating epithelial growth and repair.

Disruption of the intestinal epithelial barrier is a hallmark of Inflammatory Bowel Disease (IBD), yet mechanisms of epithelial repair remain poorly understood. We developed a scalable human colon organoid-derived model in a 96-well Transwell system that recapitulates the cellular diversity and barrier function of the native epithelium. Using this platform, we investigated how various growth factors and cytokines influence epithelial maturation and repair following simulated inflammatory damage. Our results demonstrate that restorative factors, particularly EGF and TGFα, significantly enhance

Tumor organoid-immune cell co-culture systems for precision oncology.

Three-dimensional tumor organoids, particularly patient-derived organoids (PDOs), recapitulate key morphological, genetic, and functional features of original tumors. Co-culture with immune cells enables studies of the tumor immune microenvironment (TIME) and holds promise for personalized immunotherapy. In this review, we critically evaluate established methodologies for tumor organoid-immune cell co-culture, including reductionist, holistic (tumor slice culture and air-liquid interface), and organoid-on-a-chip approaches. We provide quantitative benchmarking of success rates, immune cell per…

Microfluidic Control of Dorsal-Ventral Patterning Within a Single Forebrain Organoid.

How distinct regional identities emerge within a single developing brain remains poorly understood. Current in vitro models address this by fusing independently generated organoids, but this introduces variability in size, maturation state, and connectivity, confounding the study of regionalization itself. Here, we present a microfluidic platform that supports the co-development of different tissue identities within a single, continuous 3D culture domain. The device integrates controlled microfluidic flow with real-time fluorescence imaging, providing stable perfusion and high-resolution track…

Retinoblastoma: unveiling molecular pathogenesis and pioneering organoid-driven therapeutic innovations.

Retinoblastoma (RB), which is the most common pediatric intraocular malignancy driven by RB1 inactivation, presents with clinical challenges, such as treatment toxicity, relapse, and resistance. Traditional models inadequately replicate human RB genetics or tumor heterogeneity, warranting the development of advanced in vitro platforms. Retinal organoids generated from human pluripotent or patient-specific stem cells enable three-dimensional(3D) modeling of the tumor microenvironment, drug screening, and mechanistic studies. This review summarizes RB pathogenesis, including RB1 loss, MYCN ampli…

Radiation-induced interferon-I response impairs thyroid organoid function.

BACKGROUND AND AIM: Radiotherapy is a standard cancer treatment, but radiation exposure to surrounding healthy tissues may lead to adverse side effects that compromise patient quality of life. In patients with head and neck cancer treated with radiotherapy, thyroid damage is a frequent complication, resulting in hypothyroidism or secondary thyroid malignancies. Although clinically recognized, the molecular mechanisms underlying these side effects remain mostly unexplored. This study aims to characterize the radiation-induced molecular alterations in thyroid organoids. METHODS: Bulk RNA sequen…

Physical, chemical, and structural properties of human gastric organoid-derived mucus.

The gastric mucus layer protects the epithelium from gastric acid and ingested pathogens. However, studies of human gastric mucus have been limited due to poor accessibility of native human mucus and the abundance of contaminants in these samples. Here, we explored the potential of human gastric organoids as models for mucus production. Immunofluorescence staining confirmed that the organoids produced mucus containing MUC5AC and MUC6. The luminal mucus had viscoelastic properties similar to those of native human gastric mucus, as determined by particle tracking microrheology. To collect organo…

Cadmium Toxicity in Testis Organoids: Validation of an In Vitro Model for Testicular Toxicology.

Environmental toxicants are increasingly implicated in male reproductive disorders, highlighting the need for physiologically relevant in vitro testicular models for toxicological testing. We previously developed a testis organoid system that recapitulates key architectural and endocrine features of native tissue; however, its suitability for toxicological applications requires validation. In this study, cadmium, a well-characterized environmental heavy metal and testicular toxicant, was used as a positive control to evaluate model responsiveness. Testis organoids were exposed to increasing co…

The application and progress of AI-based image analysis in tumor organoid research.

The development of preclinical models that recapitulate the physiological and pathological features of human tumors remains a central challenge in cancer research. Advances in cell biology have enabled the generation of three-dimensional tumor organoids, which closely mirror patient-specific therapeutic responses and facilitate the study of disease mechanisms. However, the trend of these models necessitates a shift from traditional, invasive analytical methods toward non-invasive, high-throughput imaging approaches. Here, we review the current state of tumor organoid culture and the emerging a…

Nanofibers for organoid cultivation advances from chemical, physical and biological dimensions.

Organoids are high-fidelity in vitro models with substantial potential in biomedicine. However, traditional animal-derived matrix gels contain undefined components and have unpredictable mechanical properties, which greatly hinder the standardization and functional maturation of organoids. In recent years, nanofibers have emerged as an ideal substrate for organoid cultivation because of their structural similarity to natural extracellular matrices and their easily tunable properties. Accordingly, this article systematically reviews the synergistic effects of nanofibers across chemical, physica…

Looping patchworks and imaginations in a concept: Building the patchwork of "organoid" in contemporary biotechnology.

Organoids have become a central but contested notion across developmental biology, stem cell research, tissue engineering and regenerative medicine. I argue that the term organoid functions not as a single, well-defined scientific kind but as a special kind of patchwork concept that I call looping patchwork concept: a coordination device whose stability depends on overlapping inferential patches distributed across material practices, ethical deliberations, and promissory imaginaries. Drawing on Hacking's looping kinds, Jasanoff's co-production framework, and recent philosophical work on patchw…

Smart organoid-on-chip systems: From microfluidics to sensors, control, and AI.

Organoid systems have opened the door to modeling human tissues with a level of biological realism that conventional 2D cultures could never achieve. Yet, as many laboratories have experienced firsthand, organoids are extremely sensitive to their microenvironment and difficult to standardize across experiments. The growing shift toward smart, sensor-integrated organoid-on-a-chip platforms reflects an attempt to address these persistent challenges. These hybrid systems combine organoid biology with microfluidics, real-time sensors, microcontroller-based actuation, and increasingly, AI-driven in…
